Currently, little information has been published on the application of ternary eluent compositions in supercritical fluid chromatography for separating peptides. This work investigates the benefits of adding acetonitrile to methanol as the modifier. Three cyclic antibiotic peptides (bacitracin, colistin, and daptomycin) ranging between 1000 and 2000 Da were chosen as model substances. The ternary mixture of carbon dioxide, methanol, and acetonitrile is optimized to increase the resolution of the peptide's fingerprint. In addition, varying compositions of methanol and acetonitrile were found to change the elution order of the analytes, which is a valuable tool during method development. An individual gradient method using two Torus 2-PIC columns (each 100 × 3.0 mm, 1.7 μm), carbon dioxide, and a modifier consisting of acetonitrile/methanol/water/methanesulfonic acid (60:40:2:0.1, v:v:v:v) was optimized for each of the peptides. Subsequently, a generic method development protocol applicable to polypeptides is proposed.  相似文献

In real-world applications of optimization, optimal solutions are often of limited value, because disturbances of or changes to input data may diminish the quality of an optimal solution or even render it infeasible. One way to deal with uncertain input data is robust optimization, the aim of which is to find solutions which remain feasible and of good quality for all possible scenarios, i.e., realizations of the uncertain data. For single objective optimization, several definitions of robustness have been thoroughly analyzed and robust optimization methods have been developed. In this paper, we extend the concept of minmax robustness (Ben-Tal, Ghaoui, & Nemirovski, 2009) to multi-objective optimization and call this extension robust efficiency for uncertain multi-objective optimization problems. We use ingredients from robust (single objective) and (deterministic) multi-objective optimization to gain insight into the new area of robust multi-objective optimization. We analyze the new concept and discuss how robust solutions of multi-objective optimization problems may be computed. To this end, we use techniques from both robust (single objective) and (deterministic) multi-objective optimization. The new concepts are illustrated with some linear and quadratic programming instances.  相似文献

It is proved that the solutions to the singular stochastic pp-Laplace equation, p∈(1,2)p(1,2) and the solutions to the stochastic fast diffusion equation with nonlinearity parameter r∈(0,1)r(0,1) on a bounded open domain Λ⊂RdΛRd with Dirichlet boundary conditions are continuous in mean, uniformly in time, with respect to the parameters pp and rr respectively (in the Hilbert spaces L2(Λ)L2(Λ), H−1(Λ)H1(Λ) respectively). We propose a parallel implementation of the Preconditioned Conjugate Gradient algorithm on a GPU platform. The preconditioning matrix is an approximate inverse derived from the SSOR preconditioner. Used through sparse matrix–vector multiplication, the proposed preconditioner is well suited for the massively parallel GPU architecture. As compared to CPU implementation of the conjugate gradient algorithm, our GPU preconditioned conjugate gradient implementation is up to 10 times faster (8 times faster at worst).  相似文献

Thin droplets spreading on a solid substrate are investigated, with a special focus on temperature effects. The aim is to manipulate the fingering instability which may occur in the spreading in a spin coating process. The analysis bases on lubrication approximation, valid for flat thin droplets, which usually is the case. The dynamic of the wetting is implemented by using a generalized law of Tanner, coupling the contact angle (CA) of the droplet at the (apparent) contact line (CL) with its speed. A one-way coupling is used to investigate, whether viscous heating has to be taken into account. It can be derived that its role is negligible in the spreading process of a thin droplet, even for a relatively large viscous influence (large capillary number). Analyzing the results of a linear stability analysis of the fingering instability and taking Marangoni-stresses (MS) into account reveals, that the instability may be suppressed by cooling the ambient gas or heating the substrate during the spreading. Unfortunately an comparison with experiments for spreading droplets in a heated gas shows deviations for larger spreading radii. The influence of temperature on density is investigated and on the way a criteria, from which it may be obtained whether a simple Boussinesq-approximation (BA) is appropriate or not.
