Low-energy CAD product-ion spectra of various molecular species of phosphatidylserine (PS) in the forms of [M−H]− and [M−2H+Alk]− in the negative-ion mode, as well as in the forms of [M+H]+, [M+Alk]+, [M−H+2Alk]+, and [M−2H+3Alk]+ (where Alk=Li, Na) in the positive-ion mode contain rich fragment ions that are applicable for structural determination.
Following CAD, the [M−H]− ion of PS undergoes dissociation to eliminate the serine moiety (loss of C3H5NO2) to give a [M−H−87]− ion, which equals to the [M−H]− ion of a phoshatidic acid (PA) and give rise to a MS3-spectrum that is identical to the MS2-spectrum of PA. The major fragmentation process for the [M−2H+Alk]− ion of PS arises from primary loss of 87 to give rise to a [M−2H+Alk−87]− ion, followed by loss of fatty acid substituents as acids (RxCO2H, x=1,2) or as alkali salts (e. g., RxCO2Li, x=1,2). These fragmentations result in a greater abundance of [M−2H+Alk−87−R2CO2H]− than [M−2H+Alk−87−R1CO2H]− and a greater abundance of [M−2H+Alk−87−R2CO2Li]− than [M−2H+Alk−87−R1CO2Li]−; while further dissociation of the [M−2H+Alk−87−R2(or 1)CO2Li]− ions gives a preferential formation of the carboxylate anion at sn-1 (R1CO2−) over that at sn-2 (R2CO2−). Other major fragmentation process arises from differential loss of the fatty acid substituents as ketenes (loss of Rx′CH=CO, x=1,2). This results in a more prominent [M−2H+Alk−R2′CH=CO]− ion than [M−2H+Alk−R1′CH=CO]− ion. Ions informative for structural characterization of PS are of low abundance in the MS2-spectra of both the [M+H]+ and the [M+Alk]+ ions, but are abundant in the MS3-spectra. The MS2-spectrum of the [M+Alk]+ ion contains a unique ion corresponding to internal loss of a phosphate group probably via the fragmentation processes involving
rearrangement steps. The [M−H+2Alk]+ ion of PS yields a major [M−H+2Alk−87]+ ion, which is equivalent to an alkali adduct ion of a monoalkali salt of PA and gives rise to a greater abundance of [M−H+2Alk−87−R1CO2H]+ than [M−H+2Alk−87−R2CO2H]+. Similarly, the [M−2H+3Alk]+ ion of PS also yields a prominent [M−2H+3Alk−87]+ ion, which undergoes consecutive dissociation processes that involve differential losses of the two fatty acyl substituents.
Because all of the above tandem mass spectra contain several sets of ion pairs involving differential losses of the fatty
acid substituents as ketenes or as free fatty acids, the identities of the fatty acyl substituents and their positions on
the glycerol backbone can be easily assigned by the drastic differences in the abundances of the ions in each pair. 相似文献

The present study describes a new application of ruthenium(II) tris(bipyridine) (Ru(bpy)3(2+)) and osmium(II) tris(bipyridine) (Os(bpy)3(2+)) as phosphorescent labels for the quantification of surface binding of molecules to gold and silver nanoparticles. The fraction of Ru(bpy)3(2+) and Os(bpy)3(2+) that is in solution can be distinguished from the surface-bound fraction by the relative lifetimes and integrated emission yields as determined by time-correlated single-photon counting (TCSPC) spectroscopy. Complementary steady-state measurements were carried out to confirm surface attachment of the phosphorescent label molecules. Although the emission of solutions of Ru(bpy)3(2+) and Os(bpy)3(2+) is quenched proportional to the concentration of 10 nm Au or 20 nm Ag nanoparticles, the quenching is static and not diffusional quenching observed in Stern-Volmer plots. The results demonstrate that time-resolved spectroscopy provides a rapid method for the measurement of surface binding of labeled molecules on metallic nanoparticles. While steady-state measurements require the preparation of a series of samples with varying quencher concentrations and a reference, the method described herein requires a single sample plus reference. The mechanism for phosphorescence quenching on Au and Ag nanoparticles is discussed in terms of energy and electron transfer theories. 相似文献

The formation of the superoxide radical anion (O2*-) adduct of the nitrone 5,5-dimethyl-1-pyrroline N-oxide (DMPO) as detected by electron paramagnetic resonance (EPR) spectroscopy is one of the most common techniques for O2*- detection in chemical and biological systems. However, the nature of DMPO-O2H has confounded spin-trapping investigators over the years, since there has been no independently synthesized DMPO-O2H to date. A density functional theory (DFT) approach was used to predict the isotropic hyperfine coupling constants arising from the N, beta-H, and gamma-H nuclei of DMPO-O2H using explicit interactions with water molecules as well as via a bulk dielectric effect employing the polarizable continuum model (PCM). Theoretical calculation on the thermodynamics of DMPO-O2H decay shows favorable intramolecular rearrangement to form a nitrosoaldehyde and a hydroxyl radical as products, consistent with experimental observations. Some pathways for the bimolecular decomposition of DMPO-O2H and DMPO-OH have also been computed. 相似文献

We describe in situ neutron reflectivity (NR) and RAIRS studies of the chemical modification of films of a polypyrrole-based conducting polymer derived from the pentafluorophenyl ester of poly(pyrrole-N-propanoic acid) (PFP) electrodeposited on electrode surfaces. We explore the role of the solvent in controlling the rate of reaction with solution-based nucleophiles (amines, which react with the ester to form amides). By varying the identity of the solvent (water vs acetonitrile) and the neutron contrast (deuteration), we find that both the identity of the solvent and its population within the film are paramount in determining chemical reactivity and electroactivity. IR signatures allow monitoring of the reaction of solution-based amine-tagged species such as amino-terminated poly(propylene glycol), ferrocene ethylamine, and lysine with film-based ester functionalities: the carbonyl bands show ester/amide interconversion and some hydrolysis to acid. Time-dependent spectral analysis shows marked variations in reaction rate with (i) (co-)polymer composition (replacement of some fluorinated ester-functionalized pyrrole with unfunctionalized pyrrole), (ii) the solvent to which the polymer film is exposed, and (iii) the rate of polymer deposition. NR data provide solvent profiles as a function of distance perpendicular to the interface, the variations of which provide an explanation for film reactivity patterns. Homopolymer films are relatively hydrophobic, thus hindering reaction with species present in water solutions. Incorporating pyrrole groups raises the solvent population-dramatically for water-thereby facilitating entry and reaction of aqueous-based lysine. Changing film deposition rate yields films with different absolute levels of solvent and reactivity patterns that are dependent on the size of the reactant molecules: more rapid deposition of polymer gives films with a more open structure leading to a higher solvent content and thence increased reactivity. These results, supported by XPS and AFM data, allow assembly of composition-structure-reactivity correlations, in which the controlling feature is film solvation. 相似文献

Lactobacillus crispatus is the dominant species in the vagina of many women. With the potential for strains of this species to be used as a probiotic to help prevent and treat dysbiosis, we investigated isolates from vaginal swabs with Lactobacillus-dominated and a dysbiotic microbiota. A comparative genome analysis led to the identification of metabolic pathways for synthesis and degradation of three major biogenic amines in most strains. However, targeted metabolomic analysis of the production and degradation of biogenic amines showed that certain strains have either the ability to produce or to degrade these compounds. Notably, six strains produced cadaverine, one produced putrescine, and two produced tyramine. These biogenic amines are known to raise vaginal pH, cause malodour, and make the environment more favourable to vaginal pathogens. In vitro experiments confirmed that strains isolated from women with a dysbiotic vaginal microbiota have higher antimicrobial effects against the common urogenital pathogens Escherichia coli and Enterococcus faecium. The results indicate that not all L. crispatus vaginal strains appear suitable for probiotic application and the basis for selection should not be only the overall composition of the vaginal microbiota of the host from which they came, but specific biochemical and genetic traits. 相似文献

The possibility of using one- and two-dimensional Kohonen self-organizing maps (SOMs) to recognize similarities in low-resolution
vapor-phase infrared spectra without any additional information, i.e., in an unsupervised mode, has been investigated. Full-range
vapor-phase FT-IR reference spectra were first used to train the networks and the trained networks were then used to classify
the reference spectra into several groups. The feasibility of reducing the spectral range to be consistent with the atmospheric
windows used in open-path FT-IR spectrometry was also studied. Kohonen networks are shown to be relatively immune to the presence

Chromatographic retention factors (k′) of a series of eight β-adrenoceptor antagonist compounds (β-adrenolytic drugs) were determined employing an immobilized artificial membrane column (IAM.PC.DD). The influence of mobile phase pH, ionic strength, and organic modifier composition was studied in order to examine column performance. After the IAM.PC.DD columns were exposed to approximately 7000 column volumes of a 0.01 M PBS mobile phase, five out of six columns tested showed significant peak broadening and decreased k′ values indicative of premature column failure. The data suggested that the immobilized phospholipids stationary phase was removed by the 0.01 M PBS mobile phase. The β-adrenolytic drug's log k′IAM values obtained with an IAM.PC.DD column were compared to an esterIAM.PC.MG column for predicting drug membrane interactions. For the linear regression analysis between log k′IAM and the logarithm of the n-octanol–water partition coefficients (rIAM.PC.DD=0.8710 vs. rIAM.PC.MG=0.9538), the C18 HPLC retention factors (rIAM.PC.DD=0.8408 vs. rIAM.PC.MG=0.9380), the liposome partition coefficients (rIAM.PC.DD=0.8887 vs. rIAM.PC.MG=0.9187), and various pharmacokinetic parameters, significantly better correlations were obtained with the esterIAM.PC.MG column than the IAM.PC.DD column. 相似文献
