The positions of a given fold always occupied by strong hydrophobic amino acids (V, I, L, F, M, Y, W), which we call “topohydrophobic positions”, were detected and their properties demonstrated within 153 non-redundant families of homologous domains, through 3D structural alignments. Sets of divergent sequences possessing at least four to five members appear to be as informative as larger sets, provided that their mean pairwise sequence identity is low. Amino acids in topohydrophobic positions exhibit several interesting features: they are much more buried than their equivalents in non-topohydrophobic positions, their side chains are far less dispersed; and they often constitute a lattice of close contacts in the inner core of globular domains. In most cases, each regular secondary structure possesses one to three topohydrophobic positions, which cluster in the domain core. Moreover, using sensitive alignment processes such as hydrophobic cluster analysis (HCA), it is possible to identify topohydrophobic positions from only a small set of divergent sequences. Amino acids in topohydrophobic positions, which can be identified directly from sequences, constitute key markers of protein folds, define long-range structural constraints, which, together with secondary structure predictions, limit the number of possible conformations for a given fold. Oxidations of 14α-hydroxy-5α-cholestan-3β-yl acetate ( 5 ) with lead tetraacetate under thermal or photolytic conditions or in the presence of iodine proceed mainly by fragmentation of the C(13)−C(14) bond to give as the primary products the 13,18-didehydro-13,14-seco derivative 6 and the (E )-Δ12-13,14-seco ketone 11 , respectively. Further transformations of these compounds under conditions of their formation afforded, in addition, the acetoxy derivatives 7 – 9 (from 6 ), and the D-homo-C-nor compound 12 and (12R,13R)-epoxide 13 (from 11 ). Unexpectedly, the photolytic lead-tetraacetate oxidation of 5 resulted partly (to ca. 20%) in a reversible fragmentation involving scission and recombination of the C(8)−C(14) bond followed by formation of the 14β,22-ether 10 . Possible mechanisms for the observed transformations are discussed.  相似文献

Using a recently proposed non-empirical Heisenberg hamiltonian, the ground and excited covalent surfaces of the styrene molecule are calculated as functions of the external double-bond twisting angle θ, including full bond-length optimisation. The lowest excited singlet surface exhibits a barrier at 0 ?~ 45° resulting from a weakly avoided crossing of two neutral diabatic surfaces. The isomerisation should proceed from the twisted funnel. This model and the calculated transition energies are in good agreement with experiment.  相似文献

The multiple-quantum magic-angle spinning (MQMAS) and satellite-transition magic-angle spinning (STMAS) experiments refocus second-order quadrupolar broadening of half-integer quadrupolar spins in the form of two-dimensional experiments. Isotropic shearing is usually applied along the indirect dimension of the 2D spectra such that an isotropic projection free of anisotropic quadrupolar broadening can be obtained. An alternative shear transformation by a factor equal to the coherence level (quantum number) selected during the evolution period is proposed. Such a transformation eliminates chemical shift along the indirect dimension leaving only the second-order quadrupolar-induced shift and anisotropic broadening, and is expected to be particularly useful for disordered systems. This transformation, dubbed Q-shearing, can help avoid aliasing problems due to large chemical shift ranges and spinning sidebands. It can also be used as an intermediate step to the isotropic representation for expanding the spectral window of rotor-synchronized experiments.  相似文献

A triad D-Ir-A, where Ir is an Ir(III) bisterpyridine complex connected through an amidophenyl spacer to D, a triphenylamine electron donor, and to A, a naphthalene bisimide electron acceptor, has been synthesized and electrochemically investigated. The photoinduced processes in the triad, which is more than 4-nm long, have been characterized by steady-state and time-resolved optical spectroscopy by comparison with the model dyads D-Ir, Ir-A, and the reference monomers D, Ir, and A. A sequential electron transfer occurs upon excitation of the D and Ir units, leading to the charge-separated state D+-Ir- -A in 100 % yield and subsequently to D+-Ir-A- in about 10 % yield. The final charge-separated state has a lifetime at room temperature of 120 micros in air-free acetonitrile and of 100 micros in air-equilibrated solvent. Excitation of the A units does not yield intramolecular reactivity, but the resulting triplet-excited state localized on the acceptor, D-Ir-3A, displays intermolecular reactivity.  相似文献

Carboxylated pyrrole (Pyr, a index)- and carbazole (Cbz, b index)-containing monomers 6-7a/b and 9a/b have been readily synthesized from the monobenzyl ester of L-glutamic acid and triamine 2 using Clauson-Kaas and amide coupling reactions. In contrast to Pyr-containing compounds 6-7a, and 9a, the three Cbz-containing monomers 6-7b, and 9b have been found electroactive and were successfully electropolymerized on a Pt electrode resulting in the deposition of corresponding insoluble electroconducting polyCOOH polyCbz-films poly(6-7b) and poly(9b).  相似文献

The investigation of ultrafast dynamics, taking place on the few to sub‐picosecond time scale, is today a very active research area pursued in a variety of scientific domains. With the recent advent of X‐ray free‐electron lasers (XFELs), providing very intense X‐ray pulses of duration as short as a few femtoseconds, this research field has gained further momentum. As a consequence, the demand for access strongly exceeds the capacity of the very few XFEL facilities existing worldwide. This situation motivates the development of alternative sub‐picosecond pulsed X‐ray sources among which femtoslicing facilities at synchrotron radiation storage rings are standing out due to their tunability over an extended photon energy range and their high stability. Following the success of the femtoslicing installations at ALS, BESSY‐II, SLS and UVSOR, SOLEIL decided to implement a femtoslicing facility. Several challenges were faced, including operation at the highest electron beam energy ever, and achievement of slice separation exclusively with the natural dispersion function of the storage ring. SOLEIL's setup also enables, for the first time, delivering sub‐picosecond pulses simultaneously to several beamlines. This last feature enlarges the experimental capabilities of the facility, which covers the soft and hard X‐ray photon energy range. In this paper, the commissioning of this original femtoslicing facility is reported. Furthermore, it is shown that the slicing‐induced THz signal can be used to derive a quantitative estimate for the degree of energy exchange between the femtosecond infrared laser pulse and the circulating electron bunch.  相似文献

The factors governing the stability and the reactivity towards cyclic esters of heteroleptic complexes of the large alkaline earth metals (Ae) have been probed. The synthesis and stability of a family of heteroleptic silylamido and alkoxide complexes of calcium [{LO(i)}Ca-Nu(thf)(n)] supported by mono-anionic amino ether phenolate ligands (i = 1, {LO(1)}(-) = 4-(tert-butyl)-2,6-bis(morpholinomethyl)phenolate, Nu(-) = N(SiMe(2)H)(2)(-), n = 0, 4; i = 2, {LO(2)}(-) = 2,4-di-tert-butyl-6-{[2-(methoxymethyl)pyrrolidin-1-yl]methyl}phenolate, Nu(-) = N(SiMe(2)H)(2)(-), n = 0, 5; i = 4, {LO(4)}(-) = 2-{[bis(2-methoxyethyl)amino]methyl}-4,6-di-tert-butylphenolate, Nu(-) = N(SiMe(2)H)(2)(-), n = 1, 6; Nu(-) = HC≡CCH(2)O(-), n = 0, 7) and those of the related [{LO(3)}Ae-N(SiMe(2)H)(2)] ({LO(3)}(-) = 2-[(1,4,7,10-tetraoxa-13-azacyclopentadecan-13-yl)methyl]-4,6-di-tert-butylphenolate Ae = Ca, 1; Sr, 2; Ba, 3) have been investigated. The molecular structures of 1, 2, [(4)(2)], 6, and [(7)(2)] have been determined by X-ray diffraction. These highlight Ae???H-Si internal β-agostic interactions, which play a key role in the stabilization of [{LO(i)}Ae-N(SiMe(2)H)(2)] complexes against ligand redistribution reactions, in contrast to regular [{LO(i)}Ae-N(SiMe(3))(2)]. Pulse-gradient spin-echo (PGSE) NMR measurements showed that 1, 4, 6, and 7 are monomeric in solution. Complexes 1-7 mediate the ring-opening polymerization (ROP) of L-lactide highly efficiently, converting up to 5000?equivalents of monomer at 25?°C in a controlled fashion. In the immortal ROP performed with up to 100?equivalents of exogenous 9-anthracenylmethanol or benzyl or propargyl alcohols as a transfer agent, the activity of the catalyst increased with the size of the metal (1<2<3). For Ca-based complexes, the enhanced electron-donating ability of the ancillary ligand favored catalyst activity (1>6>4≈5). The nature of the alcohol had little effect over the activity of the binary catalyst system 1/ROH; in all cases, both the control and end-group fidelity were excellent. In the living ROP of L-LA, the HC≡CCH(2)O(-) initiating group (as in 7) proved superior to N(SiMe(2)H)(2)(-) or N(SiMe(3))(2)(-) (as in 6 or [{LO(4)}Ca-N(SiMe(3))(2)] (B), respectively).  相似文献

A formal [2 + 2] cycloaddition-cycloreversion (CA-CR) between N,N'-dicyanoquinone diimides (DCNQIs) and electron-rich alkynes was explored, providing a new class of π-conjugated donor-acceptor chromophores. These DCNQI adducts exist in the solid state as single diastereoisomers, but as two interconverting diastereoisomers in solution. Solid- and solution-state evidence for intramolecular charge transfer (CT) was obtained; additionally, the DCNQI adducts exhibit positive solvatochromism and significant solution-state third-order polarizabilities.  相似文献
