Time-resolved fluorescence resonance energy transfer (FRET) measurements were used to measure distance distributions and intramolecular dynamics (site-to-site diffusion) of a 28-residue single-domain zinc finger peptide in the absence and presence of zinc ion. Energy transfer was measured between TRP14 and a N-terminal DNS group. As expected, the TRP-to-DNS distance distribution for zinc-bound peptide is shorter and narrower (R av=11.2 Å,hw=2.8 Å) than the metal-free peptide (R av=20.1 Å,hw=14.5 Å). The degree of mutual donor-to-acceptor diffusion (D) was also determined for these distributions. For zinc-bound peptide there is no detectible diffusion (D0.2 Å2/ns), whereas for metal-free peptide a considerable amount of motion is occurring between the donor and the acceptor (D=12 Å2/ns). These results indicate that the zinc-bound peptide folds into a unique, well-defined conformation, whereas the metal-free conformation is flexible and rapidly changing. The absence of detectible mutual site-to-site diffusion between the donor and the acceptor in the metal-bound zinc finger peptide indicates that intramolecular motion is essentially frozen out, on the FRET time scale, as a consequence of zinc coordination.Dedicated to the memory of Barbara D. We examined the time-dependent intensity decays ofN-acetyl-l-tryptophanamide (NATA) when collisionally quenched by acrylamide in propylene glycol over a range of temperatures. The intensity decays of NATA became increasingly heterogeneous in the presence of acrylamide. The NATA intensity decays were not consistent with the Collins-Kimball radiation boundary condition (RBC) model for quenching. The steady-state Stern-Volmer plots show significant upward curvature, and quenching of NATA by acrylamide was observed even in vitrified propylene glycol, where translational diffusion cannot occur during the lifetime of the excited state. These frequencydomain and steady-state data indicate a through-space quenching interaction between NATA and acrylamide, and the results are consistent with a rate constant for quenching that depends exponentially on the fluorophore-quencher separation distance. The exponential distance-dependent rate of quenching also explains the upward curvature of the Stern-Volmer plot, and the steady-state data aid in determining the interaction distance between NATA and acrylamide. These results suggest that the distance-dependent quenching rates need to be considered in the interpretation of acrylamide quenching of proteins.  相似文献

Summary Radiochemical neutron activation procedures using liquid-liquid extraction with tetraphenylarsonium chloride in chloroform from 1M HCl and solid extraction with ALIQUAT 336 incorporated in a polyacrylonitrile binding matrix from 0.1M HCl were developed for accurate determination of rhenium in biological and environmental samples at the sub-ng . g-1 level. Concentrations of Re in the range of 0.1 to 2.4 ng . g-1 were determined in several botanical reference materials (RM), while in a RM of road dust a value of ~10 ng . g-1 was found. Significantly elevated values of Re, up to 90 ng . g-1 were found in seaweed (brown algae). Results for Re in the brown algae Fucus vesiculosus in which elevated 99Tc values had previously been determined suggested possible competition between Re and Tc in the accumulation process.  相似文献

The extraction of Am, Eu, Ce(III), Zr(IV), and Sr from aqueous nitric acid— nitrate media by dibutyl N,N-diethylcarbamylphosphonate and dibutyl N,N-diethylcar-bamylmethylenephosphonate dissolved in carbon tetrachloride was studied. The trivalent actinide and lanthanide elements may be separated in certain aqueous phase acidity regions from Fe(III), Zr(IV), and Sr, whereas the separation of actinide elements from the lanthanide elements is poor. The extractant to metal ratio in the extracted complexes of Am, Eu, and Ce(III) is 3. The interferences in the extraction due to acidic impurities in the extracting agent are discussed.  相似文献

To determine this substance it was necessary to find experimental conditions that would allow eliminating the influence of other components of the tablets. Trimellitic anhydride (TMA) is widely used industrially to make epoxy and alkyd resins, plasticizers and surfactants. The purpose of this study was to investigate whether contact hypersensitivity (CHS) is induced by repeated TMA challenge and the role of TNF-alpha and IgE in the TMA-induced CHS. The repetition of the challenge enlarged the extent of an early and a late phase of CHS in TNF-alpha+/+ (B6129SF2/J) and Balb/c mice. In the late phase of TMA-induced CHS, the peak of ear swelling responses by single challenge showed at 24 h after challenge, but the peak was observed at 8 h after repeated challenge. In the TNF-alpha knockout TNF-alpha-/- (B6;129S-Tnftm1Gk1) mice, the repetition of the TMA challenges enlarged the extent of the late phase of CHS, but less than those in TNF-alpha+/+ mice. Injection of anti-TNF-alpha antibody into the peritoneal cavity of Balb/c mice significantly decreased the extent of the late phase of CHS. Subcutaneous injection of anti-IgE antibody into Balb/c mice also decreased the extent of the late phase of CHS in dose-dependent manner. Histologically, infiltration of polymorphonuclear leukocytes and eosinophils was more pronounced in repeatedly TMA-challenged TNF-alpha+/+ and Balb/c mice than in the TNF-alpha-/- mice and anti-TNF-alpha or anti-IgE antibodies treated Balb/c mice. These results indicate that mice sensitized by TMA could possibly offer a useful model to study the mechanism of CHS, and TNF-alpha and IgE may act as potential modulators in the late phase of TMA-induced CHS. Neutralization of TNF-alpha and IgE by anti-TNF-alpha or anti-IgE antibodies may provide therapeutic tools for the treatment of TMA-induced CHS.  相似文献

A set of certified reference materials of four natural soils denoted as CRM 7001-4 was prepared. They consist of light sandy soils with normal and elevated, silty clay loam with normal, and loam with elevated element levels. Certified and/or information values were established for the total contents of the elements As, Ba, Be, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb, V, Zn, and their fractions, extractable with aqua regia, and boiling and cold 2 mol L–1 nitric acid, from results of a 28 laboratory intercomparison using various methods of atomic absorption spectrometry, optical emission spectrometry, mass spectrometry, electrochemical methods, spectrophotometry, instrumental neutron activation analysis, and X-ray fluorescence.  相似文献

As a part of our general interest in the UV-Vis spectroscopy of multidentate mixed-donor ligands, the (salicylideneethylenediamine)Cu(II) complex has been prepared and characterized by elemental analyses, solubility in common solvents, molar conductivities, and ultraviolet (UV), and visible (Vis) spectroscopy. The combined results of spectrophotometric measurements and EPR spectra, as well as known the X-ray structure for solids, were used to determine the structure of the investigated complex in solutions. The spectra of [Cu(salen)] (H2salen = salicylideneethylenediamine), were measured in various solvents at room temperature, resolved by Gaussian analysis, and angular overlap model (AOM) treated in C 2v symmetry. Because of overparametrization problems, the bis(salicylaldehyde)Cu(II) complex has been characterized and AOM treated. The results of this have been used for AOM studies of [Cu(salen)]. The effect of the solvents upon the - and -bonding ligand abilities is discussed.  相似文献

The solid-state ternary complex of terbium chloride with L-tyrosine and glycine, [Tb(Tyr)(Gly)3Cl3·3H2O], was synthesized and characterized. Using a solution-reaction isoperibol calorimeter, the enthalpy of reaction for the following reaction, TbCl3·6H2O(s)+Tyr(s)+3Gly(s)=Tb(Tyr)(Gly)3Cl3·3H2O(s)+3H2O(l), was determined to be (5.1±0.6) kJ mol-1. The standard enthalpy of formation of Tb(Tyr)(Gly)3Cl3-3H2O at T=298.15 K has been derived as -(4267.3±2.3) kJ mol-1. The thermal decomposition kinetics of the complex was studied by non-isothermal thermogravimetry in the temperature range of 325-675 K. Two main mass loss stages existed in the process of the decomposition of the complex, the kinetic parameters for the second stage were analyzed by means of differential and integral methods, respectively. Comparing the results of differential and integral methods, mechanism functions of the thermal decomposition reaction for its second stage were proposed. The kinetic equation can be expressed as: d/dt=Aexp(-E/RT)(1-)2. The average values of the apparent activation energy E and pre-exponential factor A were 213.18 kJ mol-1 and 2.51·1020 s-1, respectively.  相似文献
