High-valent tetraalkylcuprates(iii ) and -argentates(iii ) are key intermediates of copper- and silver-mediated C−C coupling reactions. Here, we investigate the previously reported contrasting reactivity of [RMiii Me3] complexes (M=Cu, Ag and R=allyl) with energy-dependent collision-induced dissociation experiments, advanced quantum-chemical calculations and kinetic computations. The gas-phase fragmentation experiments confirmed the preferred formation of the [RCuMe] anion upon collisional activation of the cuprate(iii ) species, consistent with a homo-coupling reaction, whereas the silver analogue primarily yielded [AgMe2], consistent with a cross-coupling reaction. For both complexes, density functional theory calculations identified one mechanism for homo coupling and four different ones for cross coupling. Of these pathways, an unprecedented concerted outer-sphere cross coupling is of particular interest, because it can explain the formation of [AgMe2] from the argentate(iii ) species. Remarkably, the different C−C coupling propensities of the two [RMiii Me3] complexes become only apparent when properly accounting for the multi-configurational character of the wave function for the key transition state of [RAgMe3]. Backed by the obtained detailed mechanistic insight for the gas-phase reactions, we propose that the previously observed cross-coupling reaction of the silver complex in solution proceeds via the outer-sphere mechanism.  相似文献

We consider the electrical signals recorded from a subdural array of electrodes placed on the pial surface of the brain for chronic evaluation of epileptic patients before surgical resection. A simple and computationally fast method to analyze the interictal phase synchrony between such electrodes is introduced and developed with the aim of detecting and localizing the foci of the epileptic seizures. We evaluate the method by comparing the results of surgery to the localization predicted here. We find an indication of good correspondence between the success or failure in the surgery and the agreement between our identification and the regions actually operated on.  相似文献

A key problem in solid-state NMR is resolving overlapping isotropic signals. We present here a two-dimensional method which can enable sites with the same isotropic chemical shift to be distinguished according to their chemical shift anisotropy and asymmetry. The method involves correlating sideband spectra at different effective spinning rates using CSA-amplification pulse sequences. The resulting two-dimensional correlation pattern allows very accurate determination of the chemical shift principal values in addition to the recovery of parameters for two overlapping patterns which allows the resolution of overlapping signals.  相似文献

Redox-active Cu(II) complexes are able to form reactive oxygen species (ROS) in the presence of oxygen and reducing agents. Recently, Faller et al. reported that ROS generation by Cu(II) ATCUN complexes is not as high as assumed for decades. High complex stability results in silencing of the Cu(II)/Cu(I) redox cycle and therefore leads to low ROS generation. In this work, we demonstrate that an exchange of the α-amino acid Gly with the β-amino acid β-Ala at position 2 (Gly2→β-Ala2) of the ATCUN motif reinstates ROS production (OH and H2O2). Potentiometry, cyclic voltammetry, EPR spectroscopy and DFT simulations were utilized to explain the increased ROS generation of these β-Ala2-containing ATCUN complexes. We also observed enhanced oxidative cleavage activity towards plasmid DNA for β-Ala2 compared to the Gly2 complexes. Modifications with positively charged Lys residues increased the DNA affinity through electrostatic interactions as determined by UV/VIS, fluorescence, and CD spectroscopy, and consequently led to a further increase in nuclease activity. A similar trend was observed regarding the cytotoxic activity of the complexes against several human cancer cell lines where β-Ala2 peptide complexes had lower IC50 values compared to Gly2. The higher cytotoxicity could be attributed to an increased cellular uptake as determined by ICP-MS measurements.  相似文献

The ubiquitin–proteasome system (UPS) has been successfully targeted by both academia and the pharmaceutical industry for oncological and immunological applications. Typical proteasome inhibitors are based on a peptidic backbone endowed with an electrophilic C‐terminus by which they react with the active proteolytic sites. Although the peptide moiety has attracted much attention in terms of subunit selectivity, the target specificity and biological stability of the compounds are largely determined by the reactive warheads. In this study, we have carried out a systematic investigation of described electrophiles by a combination of in vitro, in vivo, and structural methods in order to disclose the implications of altered functionality and chemical reactivity. Thereby, we were able to introduce and characterize the class of α‐ketoamides as the most potent reversible inhibitors with possible applications for the therapy of solid tumors as well as autoimmune disorders.  相似文献

Chemical dimerizers are powerful tools for non‐invasive manipulation of enzyme activities in intact cells. Here we introduce the first rapidly reversible small‐molecule‐based dimerization system and demonstrate a sufficiently fast switch‐off to determine kinetics of lipid metabolizing enzymes in living cells. We applied this new method to induce and stop phosphatidylinositol 3‐kinase (PI3K) activity, allowing us to quantitatively measure the turnover of phosphatidylinositol 3,4,5‐trisphosphate (PIP3) and its downstream effectors by confocal fluorescence microscopy as well as standard biochemical methods.  相似文献

We described a simple and quick miniaturized sequencing gel system for DNA analysis. Two major modifications were made to the previously reported miniaturized DNA sequencing gel system to achieve high-resolution hydroxyl radical cleavage analysis: including formamide in the miniaturized gel and providing uniform heating during electrophoresis. Our method enables one to reduce the cost for chemicals and to significantly reduce electrophoresis time. Furthermore, minimal gel handling simplifies the entire process. We show that the resolution of DNA fragments obtained by hydroxyl radical cleavage for the miniaturized gel is similar to that of a large conventional sequencing gel.  相似文献

The first general single-step route to dendritic or cascade polyaryl ethers analogous to common linear polyaryl ethers is described. The sodium salts of four AB2 monomers each containing a single phenolic hydroxyl group and two aryl fluorides activated toward nucleophilic substitution by carbonyl, sulphonyl, or tetrafluorophenyl moieties are shown to polymerize in hot N, N-dimethylacetamide. The products are high molecular weight polymers (7000 < Mn < 36000), have narrow polydispersities (1.50 < Mw/Mn < 4.50), and are highly soluble in organic solvents. The molecular weights of two of the polymers increase with monomer concentration. The polymers are thermally stable (500 °C under N2) and have glass transition temperatures ranging from 135 to 231 °C.  相似文献

In contrast to many cellulose derivatives, the cellulose O-silyl ethers with bulky side groups exhibit scarcely a change in glass transition temperature by a variation of the degree of substitution (DS) or degree of polymerization (DP) or by introducing phenyl carbamate groups for the remaining hydroxyls along the main chain. However, a substitution of these hydroxyls by flexible acetate groups lowers the glass transition temperature considerably. The secondary dispersion (relaxation) behavior is strongly influenced by the various substituents and can be correlated to specific motions of the molecules. The dynamic mechanical properties are also dependent on the kind of preconditioning of the samples.  相似文献

The Cu2+‐dependent ligation DNAzyme is implemented as a biocatalyst for the colorimetric or chemiluminescence detection of Cu2+ ions, Hg2+ ions, or cocaine. These sensing platforms are based on the structural tailoring of the sequence of the Cu2+‐dependent ligation DNAzyme for specific analytes. The tethering of a subunit of the hemin/G‐quadruplex DNAzyme to the ligation DNAzyme sequence, and the incorporation of an imidazole‐functionalized nucleic‐acid sequence, which acts as a co‐substrate for the ligation DNAzyme that is tethered to the complementary hemin/G‐quadruplex subunit. In the presence of different analytes, Cu2+ ions, Hg2+ ions, or cocaine, the pretailored Cu2+‐dependent ligation DNAzyme sequence stimulates the respective ligation process by combining the imidazole‐functionalized co‐substrate with the ligation DNAzyme sequence. These reactions lead to the self‐assembly of stable hemin/G‐quadruplex DNAzyme nanostructures that enable the colorimetric analysis of the substrate through the DNAzyme‐catalyzed oxidation of 2,2′‐azinobis(3‐ethylbenzothiazoline‐6‐sulfonic acid), ABTS2?, by H2O2 into the colored product ABTS.?, or the chemiluminescence detection of the substrate through the DNAzyme‐catalyzed oxidation of luminol by H2O2. The detection limits for the sensing of Cu2+ ions, Hg2+ ions, and cocaine correspond to 1 nM , 10 nM and 2.5 μM , respectively. These different sensing platforms also reveal impressive selectivities.  相似文献
