Triglucosylated high-mannose-type tetradecasaccharide (Glc3Man9GlcNAc2), the oligosaccharide part of the donor substrate of oligosaccharyl transferase (OST) complex, and diglucosylated tridecasaccharide (Glc2Man9GlcNAc2) were synthesized. These oligosaccharides were assembled in a convergent and stereoselective manner. Undecasaccharide 5 was employed as the common intermediate, and coupling with trisaccharide (4) and disaccharide (3) donor afforded fully protected tetradeca-(17) and tridecasaccharide (16), respectively. These oligosaccharides were deprotected to give Glc3Man9GlcNAc2 and Glc2Man9GlcNAc2, respectively.  相似文献

Metal complexes of covalently bound porphyrins are used as sensitive probes for several investigations. Substituted derivatives of tetraphenyl-porphin, phthalocyanine, and naphthalocyanine are synthesized at positively and negatively charged as well as uncharged polymers. The photo-redox activities were studied under irradiation with visible light in the presence of a donor and an acceptor. The triplet life times of covalently bound porphyrin moieties are strongly enhanced compared with the analogous monomeric porphyrins. In addition, the polymer binding results in higher photocatalytic activity. The electron-transfer reactions of Mn(III)-containing porphyrins using the reducing agent dithionite are strongly influenced by the polymer environment. In contrast to monomeric Mn(III)-porphyrins, the porphyrins containing polymers exhibit a two-step reduction which may be due to the change of the conformation of the polymer coil. The catalytic epoxidation of 2,5-dihydrofuran with hypochlorite with formation of 3,4-epoxytetrahydrofuran occurs with water-soluble porphyrins in water. No influence of the polymer environment exists. The different reactions require reaction times from milliseconds up to hours.  相似文献

Anti‐MUC1 monoclonal antibodies (mAbs) are powerful tools that can be used to recognize cancer‐related MUC1 molecules, the O‐glycosylation status of which is believed to affect binding affinity. We demonstrate the feasibility of using a rapid screening methodology to elucidate those effects. The approach involves i) “one‐bead‐one‐compound”‐based preparation of bilayer resins carrying glycopeptides on the shell and mass‐tag tripeptides coding O‐glycan patterns in the core, ii) on‐resin screening with an anti‐MUC1 mAb, iii) separating positive resins by utilizing secondary antibody conjugation with magnetic beads, and (iv) decoding the mass‐tag that is detached from the positive resins pool by using mass spectrometric analysis. We tested a small library consisting of 27 MUC1 glycopeptides with different O‐glycosylations against anti‐MUC1 mAb clone VU‐3C6. Qualitative mass‐tag analysis showed that increasing the number of glycans leads to an increase in the binding affinity. Six glycopeptides selected from the library were validated by using a microarray‐based assay. Our screening provides valuable information on O‐glycosylations of epitopes leading to high affinity with mAb.  相似文献

Alanine synthesis by reductive amination of pyruvate was performed by the combination of NADH regeneration system and alanine dehydrogenase (AlaDH). The conversion of pyruvate to alanine was 99% after 1 h. Leucine synthesis was also carried out by the combination of NADH regeneration system and leucine dehydrogenase (LeuDH). A series of systematic tensile and microbend tests were conducted on copper foil specimens with different thicknesses. The specimens were made of a copper foil having almost unidirectional crystal orientations that was considered to be nearly single-crystal. In order to investigate the effects of slip system interactions, two different crystal orientations relative to the tensile direction were considered in the tests: one is close to coplanar double-slip orientation, and the other is close to the ideal cube orientation (the tensile direction nearly coincides to [0 0 1]) that yields multi-planar multi-slip deformation. We extended the microbend test method to include the reversal of bending, and we attempted to divide the total amount of strain-hardening into isotropic and kinematic hardening components. In the tensile tests, no systematic tendency of size dependence was observed. In the microbend tests, size-dependent kinematic hardening behavior was observed for both the crystal orientations, while size dependence of isotropic hardening was observed only for the multi-planar multi-slip case. We introduce an extended crystal plasticity model that accounts for the effects of the geometrically necessary dislocations (GNDs), which correspond to the spatial gradients of crystallographic slips. Through numerical simulations performed using the model, the origin of the size-dependent behavior observed in the microbend tests is discussed.  相似文献

Poly(p‐oxybenzoyl) (POB) crystals were prepared by reaction‐induced crystallization during direct polymerization of p‐hydroxybenzoic acid in the presence of boronic anhydrides. Polymerizations were carried out at 300 °C in dibenzyltoluene at a concentration of 1% with three kinds of anhydrides of boronic acid such as 3,4,5‐trifluorophenylboronic acid (TFB), 4‐methoxyphenylboronic acid (MPB) and 4‐biphenylboronic acid (BPB). The POB crystals were formed as precipitates in the solution and the morphology was considerably influenced by both the structure of the boronic anhydride and its concentration (cB). Needle‐like crystals were firmed in the presence of TFB anhydride (TFBA) at cBs of 5 and 10 mol % by the spiral growth of lamellae. Spherical aggregates of slab‐like crystals were formed at cBs from 50 to 100 mol %. The polymerization with MPB anhydride and BPB anhydride (BPBA) also yielded the needle‐like crystals at cBs of 50 and 5 mol %, respectively. The polymerization with TFBA at lower cB was favorable to prepare the needle‐like crystal. Molecular weight was also influenced by the structure of the boronic anhydride and cB. Mn increased generally with cB and BPBA gave the highest Mn of 14.7 × 103 at cB of 100 mol %. The loose packing of the molecules in the crystal caused by the bulkiness of the end‐groups made the polymerization in the crystals more efficiently. Morphology and molecular weight of the POB crystals could be controlled by the chemical structure and the content of boronic anhydride. © 2010 Wiley Periodicals, Inc.
