Benzo(a)pyrene is a chemical produced during the process of making fried, roasted, and smoked foods. It remains unclear whether benzo(a)pyrene affects the early development of human organs. In this study, we used the transgenic zebrafish line Tg(wt1b:GFP) as a model to assess benzo(a)pyrene-induced kidney malformation. By soaking zebrafish embryos in benzo(a)pyrene at various doses (2, 20, and 200 ppb), only a minor effect on the survival rate was detected (0 ppb: 97.8 ± 1.9 %; 2–200 ppb: 89.1 ± 5.8–91.5 ± 8.3 %). However, benzo(a)pyrene significantly affected the development of the kidney (malformation rates ranges from 50.0 ± 3.5 to 77.4 ± 5.3 %). Various abnormalities, such as unusual curving of pronephric tubes, swollen glomerulus, and incomplete development of pronephric ducts, were observed. This study provides a rapid and effective protocol for the evaluation of the notable effects of benzo(a)pyrene on embryonic kidney development. 相似文献

The photocatalyzed ortho-selective migration on a pyridyl ring has been achieved for the site-selective trifluoromethylative pyridylation of unactivated alkenes. The overall process is initiated by the selective addition of a CF3 radical to the alkene to provide a nucleophilic alkyl radical intermediate, which enables an intramolecular endo addition exclusively to the ortho-position of the pyridinium salt. Both secondary and tertiary alkyl radicals are well-suited for addition to the C2-position of pyridinium salts to ultimately provide synthetically valuable C2-fluoroalkyl functionalized pyridines. Moreover, the method was successfully applied to the reaction with P-centered radicals. In the present study, we investigated the molecular mechanisms of adenosine for its hair growth promoting effect. Adenosine stimulated the Wnt/β-catenin pathway by modulating the activity of Gsk3β in cultured human dermal papilla cells. It also activated adenosine receptor signaling, increasing intracellular cAMP level, and subsequently stimulating the cAMP mediated cellular energy metabolism. The phosphorylation of CREB, mTOR, and GSK3β was increased. Furthermore, the expression of β-catenin target genes such as Axin2, Lef1, and growth factors (bFGF, FGF7, IGF-1) was also enhanced. The inhibitor study data conducted in Wnt reporter cells and in cultured human dermal papilla cells demonstrated that adenosine stimulates Wnt/β-catenin signaling through the activation of the adenosine receptor and Gsk3β plays a critical role in transmitting the signals from the adenosine receptor to β-catenin, possibly via the Gαs/cAMP/PKA/mTOR signaling cascade. 相似文献

Striae distensae (SD) or stretch marks are common linear scars of atrophic skin with disintegrating extracellular matrix (ECM) structures. Although fibroblasts contribute to the construction of ECM structure in SD, some studies have reported that mast cell degranulation causes the disruption of ECM in early SD lesions. Lagerstroemia indica flower (LIF) has traditionally been used in India as a diuretic. However, little is known about the effect and molecular action of Lagerstroemia indica flower extract (LIFE) on alleviating SD. This study evaluated the effects of LIFE on mast cell degranulation and the synthesis of ECM components in fibroblasts. LIFE inhibits the adhesion of rat basophilic leukemia (RBL) cells, RBL-2H3 on fibronectin (FN) and the expression of integrin, a receptor for FN, thereby reducing focal adhesion kinase (FAK) phosphorylation. In addition, LIFE attenuated the allergen-induced granules and cytokine interleukin 3 (IL-3) through the adhesion with FN. Moreover, the conditioned medium (CM) of activated mast cells decreases the synthesis of ECM components, and LIFE restores the abnormal expressions induced by activated mast cells. These results demonstrate that LIFE suppresses FN-induced mast cell activation and promotes the synthesis of ECM components in fibroblast, which indicates that LIFE may be a useful cosmetic agent for SD treatment. 相似文献

ABSTRACTTIACA-I, TIACA-II were synthesized by changing the substitution position of the imidazole group in the diazocine core. TIACA-I, TIACA-II in the film state showed absorption in the range of 354 to 392 nm and exhibited blue photoluminescence (PL) emissions at 448 and 462 nm, respectively. The PL wavelength of TIACA-II is red-shifted by 14 nm than that of TIACA-I due to the electron-donating intensity depending on the position of the imidazole group. By using the formulation of the reconstruction, we explicitly construct models of k-essence, which unify the inflation in the early universe and the late accelerating expansion of the present universe by a
single scalar field. Due to the higher derivative terms, the solution describing the unification can be stable in the space
of solutions, which makes the restriction for the initial condition relaxed. The higher derivative terms also eliminate tachyon.
Therefore we can construct a model describing the time development, which cannot be realized by a usual inflaton or quintessence

We aimed to evaluate the suitability of a glass dosimeter (GD) for high-energy photon and electron beams in experimental and clinical use, especially for radiation therapy. We examined the expanded dosimetric characteristics of GDs including dose linearity up to 500 Gy, uniformity among GD lots and for individual GDs, the angular dependence, and energy dependence of 4 therapeutic x-ray qualities. In addition, we measured the dosimetric features (dose linearity, uniformity, angular dependence, and energy dependence) of the GD for electron beams of 10 different electron energy qualities. All measurements with the exception of dose linearity for photon beam were performed in a water phantom. For high-energy photon beams, dose linearity has a linear relationship for a dose ranging from 1 to 500 Gy with the coefficient of determination; R2 of 0.998. The uniformity of each GD of dose measurements was within ±0.5% for four GD lots and within ±1.2% for 80 GDs. In terms of the effects of photon beam angle, lower absorbed doses of within 1.0% were observed between 60° and 105° than at 90°. The GD energy dependence of 4 photon beam energy qualities was within ±2.0%. On the other hand, the result of the dose linearity for high-energy electron beams showed well fitted regression line with the coefficient of determination; R2 of 0.999 between 6 and 20 MeV. The uniformity of GDs exposed to the nominal electron energies 6, 9, 12, 16, and 20 MeV was ±1.2%. In terms of the angular dependence to electron beams, absorbed doses were within 2.0% between 60° and 105° than at 90°. In evaluation of the energy dependence of the GD at nominal electron energies between 5 and 20 MeV, we obtained responses between 1.1% and 3.5% lower than that for a cobalt-60 beam. Our results show that GDs can be used as a detector for determining doses when a high-energy photon beam is used, and that it also has considerable potential for dose measurement of high-energy electron beam. 相似文献

Tunneling magnetoresistance was found to be suppressed with decreasing temperature for magnetic tunnel junctions (MTJs) oxidized under high plasma power. A strong temperature dependence of the junction resistance was observed, along with zero-bias anomalies of dynamic resistance at low temperatures. Resistance shows a logarithmic dependence on temperature, and resistance versus temperature exhibits a scaling behavior. Our experimental data can be explained in a consistent way by the Kondo effect in the MTJs with the Kondo temperature TK=20-30 K. 相似文献
