Municipal wastewater has been examined for steroids, β2-agonists, stimulants, diuretics, and phosphodiesterase type V inhibitors (PDE type V inhibitors), which are “dual-use-drugs”
applied either as anabolic, doping, and lifestyle drugs or for treatment of diverse diseases. To identify their origin, fitness
centre discharges under suspicion of being point sources and sewage-treatment plant feed and effluents were sampled and concentrations
determined. Sensitive and selective methods for determination and quantification based on solid-phase extraction (SPE) followed
by high-performance liquid chromatography–high resolution mass and tandem mass spectrometry (HPLC–(HR)MS and HPLC–MS–MS) were
developed and established for analysis of these compounds in wastewater and to assess their effect on the environment. The
methods developed enabled quantification at trace concentrations (limit of quantification (LOQ): 5 ng L−1). Of the steroids and stimulants under investigation, testosterone, methyltestosterone, and boldenone or ephedrine, amphetamine,
and MDMA (3,4-methylendioxy-N-methylamphetamine) were observed at up to 5 μg L−1 (ephedrine). Of the β2-agonists salbutamol only, and of the diuretics furosemide and hydrochlorothiazide were confirmed in the extracts. Quite high
concentrations of the PDE type V inhibitors sildenafil, tadalafil, and vardenafil and their metabolites were confirmed in
fitness centre discharges (sildenafil: 1,945 ng L−1) whereas their concentrations in municipal wastewater did not exceed 35 ng L−1. This study identified anabolic and doping drugs in wastewater for the first time. Results obtained from wastewater treatment
plant effluents proved that these “dual-use-drugs”, with the exception of hydrochlorothiazide, were mostly eliminated. 相似文献

In this work, a method for simultaneous determination of amfepramone, fenproporex, sibutramine and fluoxetine was developed by capillary zone electrophoresis with capacitively coupled contactless conductivity detection (C4D) using a homemade capillary electrophoretic system. The optimized conditions for the separation of the pharmaceuticals by CZE were as follows: 50 mmol L− 1 phosphate buffer (pH 5.0) in 50/50 (v/v) mixture of water/acetonitrile as the working electrolyte, 15 kV separation voltage, 25 °C separation temperature, hydrodynamic injection by gravity using 20 cm injection height and 60 s injection time. The detection by C4D was carried out by using a homemade detector, which employs a sinusoidal wave generator operating at 600 kHz frequency and 2 Vpp wave amplitude. The optimized and validated CZE-C4D method was applied for the determination of the studied pharmaceuticals as adulterants in phytotherapeutic formulations commercialized in Brazil for slimming purposes. 相似文献

Synthesis and characterisation of the SrxBa1−xFeO3−y-system and the fluorinated phases SrxBa1−xFeO2F

Compounds in the system SrxBa1?xFeO3?y have been prepared under different partial pressures of oxygen. In this system, different perovskite-type structures are found depending highly on the values of x and y. Fluorination using polyvinylidenedifluoride (PVDF) gives oxyfluoride materials of composition SrxBa1?xFeO2F, which normally crystallize in the cubic perovskite structure. Rietveld refinement results provide information about the packing density for oxide and oxyfluoride samples and allow a general comparison between these two different types of materials. Furthermore, the determination of the average iron oxidation state also showed that the oxygen deficiency, y, depends significantly on the value of x and the structure determined by the Sr/Ba ratio. 相似文献

The αvβ6 integrin binds the RGD‐containing peptide of the foot and mouth disease virus with high selectivity. In this study, the long binding helix of this ligand was downsized to an enzymatically stable cyclic peptide endowed with sub‐nanomolar binding affinity toward the αvβ6 receptor and remarkable selectivity against other integrins. Computational studies were performed to disclose the molecular bases underlying the high binding affinity and receptor subtype selectivity of this peptide. G‐protein‐coupled receptors (GPCRs) are involved in a wide range of physiological processes, and they have attracted considerable attention as important targets for developing new medicines. A central and largely unresolved question in drug discovery, which is especially relevant to GPCRs, concerns ligand selectivity: Why do certain molecules act as activators (agonists) whereas others, with nearly identical structures, act as blockers (antagonists) of GPCRs? To address this question, we employed all‐atom, long‐timescale molecular dynamics simulations to investigate how two diastereomers (epimers) of dihydrofuroaporphine bind to the serotonin 5‐HT1A receptor and exert opposite effects. By using molecular interaction fingerprints, we discovered that the agonist could mobilize nearby amino acid residues to act as molecular switches for the formation of a continuous water channel. In contrast, the antagonist epimer remained firmly stabilized in the binding pocket. 相似文献

When applied to a pure component suspension in an apolar solvent, a strong inhomogeneous electric field induces particle movement, and the particles are collected at the surface of one of the two electrodes. This new phenomenon was used to separately isolate two organic crystalline compounds, phenazine and caffeine, from their suspension in 1,4‐dioxane. First, crystals of both compounds were collected at different electrodes under the influence of an electric field. Subsequent cooling crystallization enabled the immobilization and growth of the particles on the electrodes, which were separately collected after the experiment with purities greater than 91 %. This method can be further developed into a technique for crystal separation and recovery in complex multicomponent suspensions of industrial processes. 相似文献

A convenient synthesis of novel bifunctional poly(amino carboxylate) chelating agents allowing chemoselective attachment to highly functionalized biomolecules is described. Based on the well known chelator 1,4,7,10-tetraazacyclodecane-1,4,7,10-tetraacetic acid (DOTA), we synthesized novel bifunctional chelating agents bearing additional functional groups by alkylating 1,4,7,10-tetraazacyclododecane (cyclen) with one equivalent of para-functionalized alkyl 2-bromophenyl-acetate and three equivalents of tert-butyl 2-bromoacetate. The resulting compounds, which contain an additional carbonyl or alkyne functionality, allow site specific labeling of appropriately functionalized unprotected biomolecules in a rapid manner via click reactions. This was demonstrated by the attachment of our new DOTA derivatives to the somatostatin analogue Tyr3-octreotate by chemoselective oxime ligation and CuI-catalyzed azide-alkyne cycloaddition. Initial biodistribution studies in mice with the radiometalated compound demonstrated the applicability of the described DOTA conjugation. 相似文献
