Exosomes are membrane vesicles of endocytic origin released by many cell types. The molecular composition of exosomes reflects the specialised functions of their original cells. For example, these vesicles can mediate communication through their ability to bind to target cells, facilitating processes such as vascular homeostasis and antigen presentation. Although the proteomes of exosomes from several cell types are known, exploration of exosomes from additional cell types may improve our understanding of their potential physiological roles. Here, we describe the isolation and characterisation of exosomes isolated from the culture medium of murine fibroblast NIH3T3 cells and Ras-transformed NIH3T3 cells. The vesicular nature and size (30-100 nm) of the purified fibroblast exosomes was confirmed by electron microscopy. 2-D difference gel electrophoresis (DIGE) was used to compare protein profiles of exosomes secreted from NIH3T3 cells and Ras-transformed NIH3T3 cells. LC-MS/MS sequencing identified proteins in 188 protein spots in the exosomes from the two cell lines, many of which have been previously identified in exosomes from other cell types. However, some proteins identified are novel for fibroblast exosomes, such as Serpin B6. Over 34 proteins, including milk fat globule EGF factor 8 (lactadherin), collagen alpha-1 (VI), 14-3-3 isoforms, guanine nucleotide-binding proteins (G proteins), the eukaryotic translation initiation factors elF-3 gamma and elF-5A accumulated (>2-fold) in exosomes upon Ras-induced oncogenic transformation. Significantly, the 10.4-fold increase in v-Ha-Ras p21 protein in exosomes derived from Ras-transformed NIH3T3 cells suggests that exosome secretion may be implicated in eradication of obsolete proteins.  相似文献

We study 2-->n inelastic fermion-(anti)fermion scattering into multiple longitudinal weak gauge bosons and derive universal upper bounds on the scales of fermion mass generation by imposing unitarity of the S matrix. We place new upper limits on the scales of fermion mass generation, independent of the electroweak symmetry breaking scale. Strikingly, we find that the strongest 2-->n limits fall in a narrow range, 3-170 TeV (with n=2-24), depending on the observed fermion masses.  相似文献

Ferrocenoylacetone I reacts with terephthaloyl hydrazide in the presence of a catalytic amount of p-toluenesulfonic acid to afford complex 1 whereas acylation of 3(5)-ferrocenyl-5(3)-methylpyrazole II with terephthaloyl chloride leads to an isomer of complex 1, viz. complex 2. Two new complexes have been characterized by elemental analyses and IR,1H NMR, UV spectra and structurally characterized by single-crystal X-ray crystallography. Complex 1 (C36H30Fe2N4O2) crystallizes in the monoclinic space group P21/c, with lattice constants: a = 11.593(2) ?, b = 12.9962(17) ?, c = 10.0030(17) ?, β = 113.227(4)°, V = 1462.1(4) ?3, Z = 4, D c = 1.505 g·cm−3, F(000) = 684, R 1 = 0.0307, wR 2 = 0.0739. Complex 2 (C36H30Fe2N4O2) crystallizes in the monoclinic space group P21/n, with lattice constants: a = 12.2388(18) ?, b = 7.3200(15) ?, c = 17.288(4) ?, β = 90.89(3)°, V = 1466.9(5) ?3, Z = 2, D c = 1.500 g·cm−3, F(000) = 684, R 1 = 0.0410, wR 2 = 0.1247. Heteroaggregates composed of cationic porphyrin, meso-tetra(p-trimethylamino)-porphyrin iodide, its zinc complex and anionic phthalocyanine sodium hydroxygallium 4,4',4",4"'-tetrasulfonated phthalocyanine have been investigated by absorption and fluorescence spectroscopy. It has been found that both free base cationic porphyrin and its zinc complex can form very stable heterodimers with anionic phthalocyanine in water, methanol and dhnethylfonnamide. The stability of the aggregate depends on the polarity as well as the ligation ability of the solvent. No evidence of higher aggregates was detected. Besides axial coordination, steric hindrance which influence the relative orientation of the macrocycles are considered.  相似文献

First-principals calculations show that magnetization reversal is accompanied by the opposite sense of rotation of the neighboring oxygen octahedra along the [1 1 1] direction which is called the antiferrodistortive displacement in BiCrO3.The coupling between magnetization and antiferrodistortive distortion is mainly caused by Dzyaloshinskii-Moriya interaction which is driven by the eg-eg states antiferromagnetic interaction in Cr-3d.A critical value of on-site Coulomb interaction prohibiting the Dzyaloshinskii-Moriya interaction and thus the magnetization reversal is found to be 1.3eV.  相似文献

A cycle in a matroid is a disjoint union of circuits. This paper proves that every regular matroidM without coloops has a set S of cycles whose union is E(M) such that every element is in at most three of the cycles in S. It follows immediately from this that, on average, each element of M is in at most three members of the cycle cover S. This improves on a 1989 result of Jamshy and Tarsi who proved that there is a cycle cover for which this average is at most 4, and conjectured that a cycle cover exists for which the average is at most 2.  相似文献
