Unique α‐hemiaminal ether gold carbene intermediates were accessed by a gold‐catalysed 1,1‐carboalkoxylation strategy and evolved through a highly selective 1,2‐N‐migration. This skeletal rearrangement gave functionalised indenes, and isotopic labelling confirmed the rare C?N bond cleavage of the ynamide moiety. The effect of substituents on the migration has been explored, and a model is proposed to rationalise the observed selectivity. 相似文献

Capillary gel electrophoresis (CGE) methods with UV detection were developed for reduced and non‐reduced mAb analysis. These methods can be used to evaluate mAb purity, offering more reproducible quantitation compared with that of traditional SDS‐PAGE methods. These CGE methods have been utilized as platform technology for bioprocess development, formulation development, mAb characterization, drug substance/drug product release testing as well as a required methodology for stability testing. We have found these CGE methods to be applicable across a platform of mAbs in preclinical and clinical development, with the majority of mAbs requiring no modification to the method conditions. This methodology has been ICH validated and transferred to several supporting organizations. The data presented herein describes the development of CGE methodology, platform application to mAb purity analysis, ICH validation, reliability metrics, and considerations on technology enhancement for improved performance and throughput. 相似文献

The prevalence of noise in the riding of motorcycles has been a source of concern to both riders and researchers in recent times. Detailed flow field information will allow insight into the flow mechanisms responsible for the production of sound within motorcycle helmets. Flow field surveys of this nature are not found in the available literature which has tended to focus on sound pressure levels at ear as these are of interest for noise exposure legislation. A detailed flow survey of a commercial motorcycle helmet has been carried out in combination with surface pressure measurements and at ear acoustics. Three potential noise source regions are investigated, namely, the helmet wake, the surface boundary layer and the cavity under the helmet at the chin bar. Extensive information is provided on the structure of the helmet wake including its frequency content. While the wake and boundary layer flows showed negligible contributions to at-ear sound the cavity region around the chin bar was identified as a key noise source. The contribution of the cavity region was investigated as a function of flow speed and helmet angle both of which are shown to be key factors governing the sound produced by this region. 相似文献

Gold carbene reactivity patterns were accessed by ynamide insertion into a C(sp3) H bond. A substantial increase in molecular complexity occurred through the cascade polycyclization of N‐allyl ynamides to form fused nitrogen‐heterocycle scaffolds. Exquisite selectivity was observed despite several competing pathways in an efficient gold‐catalyzed synthesis of densely functionalized C(sp3)‐rich polycycles and a copper‐catalyzed synthesis of fused pyridine derivatives. The respective gold–keteniminium and ketenimine activation pathways have been explored through a structure–reactivity study, and isotopic labeling identified turnover‐limiting C H bond‐cleavage in both processes. 相似文献

A new method is presented for the prediction of unsteady axisymmetric inviscid flows. By combining a triangulated vortex approach with a novel evaluation technique for the Biot–Savart integrals, a Lagrangian vortex method is developed which eliminates the singularities usually present in axisymmetric methods, without recourse to normalizations or other approximations. Furthermore, the computational effort scales as the number of control points N and, in the large N limit, depends only on the order of quadrature employed. The accuracy and computational effort are assessed by comparison with the velocity field of a Gaussian core vortex ring and the use of the technique is illustrated by computation of the motion of Norbury rings and of vortex ring pairing. 相似文献
