Glycosyltransferase activity of 13 Leuconostoc mesenteroides strains isolated from Bulgarian fermented vegetables was investigated. All the strains displayed a mucoid phenotype on sucrose-containing agar media. Strains were characterized according to carbohydrate fermentation, species-specific multiple PCR using several primers, repetitive element-PCR fingerprinting using (GTG)5 primers and glycosyltransferase activity. Level of activity and cellular localization (soluble or cell-associated) were variable among strains. Precipitation of exopolysaccharides produced from sucrose by the soluble fractions from these strains allowed recovery of only glucans and further characterization by 1H and 13C NMR analysis and enzymatic digestion with dextranase revealed dextran production. However, levans could be detected in presence of raffinose as fructosyl donor. Both fructosyltransferase and glucosyltransferase encoding genes were detected by PCR and both active enzymes were detected after functional characterization by SDS-PAGE electrophoresis and in situ polymer production after incubation with sucrose. This work therefore showed that concomitant production of glucosyltransferase and fructosyltransferase is widespread in L. mesenteroides strains. 相似文献
Off frame reading of protein coding sequences reveals numerous stop codons. The genetic code and genomic codon usages maximize hidden stop numbers, minimizing effects of ribosomal slippages. This increases efficiency and homogeneity of protein expression. Here I test whether developmental stability in bilateral morphological traits of lizards and primates increases with numbers of hidden stops in mitochondrial protein coding genes. Expected effects are weak, but are apparent in 14 among 16 independent taxonomic groups for the non-coding +1 frame, and 12 among 16 for the ?1 frame. Results stress that many molecular factors determine phenotypes in a complex manner, frequently undetectable by classical quantitative genetics. Explicit molecular hypotheses can partially palliate this shortcoming, and uncover unsuspected links between genotype and phenotype, especially when molecular epistasis (here interactions between hidden stops and ribosomes), is involved. Effects of hidden stops on morphological developmental stability suggest that mitochondrial off frame stops might also affect human cumulative neurodegenerative diseases. Results suggest adaptive rather than neutralist interpretations of the ambush hypothesis. 相似文献
Spin‐noise appeal : Detection of NMR spin‐noise is very appealing when dilute hyperpolarized species are considered. Continuous monitoring of the noise absorption at the Larmor frequency enables determination of T1 and T2*, independently of the static magnetic field. An inductively coupled microcoil located inside the NMR tube (see picture) allows acquisition of 129Xe spin‐noise spectra without radio‐frequency excitation.
Proteins of the 2S albumin family, such as Ara h2 and Ara h6, are most frequently involved in peanut allergy. We have developed
a reverse enzyme allergo-sorbent test (EAST) in which total serum IgE antibodies are first captured by immobilised anti-human
IgE monoclonal antibodies, and then the binding of the anti-Ara h2 and anti-Ara h6 specific IgE to the corresponding labelled
allergens is measured. This reverse immunoassay was used either as a direct EAST or as an EAST inhibition assay to study the
interactions of whole peanut protein extract and purified Ara h2 and Ara h6 with IgE antibodies from peanut-allergic patients.
Finally, we identified some IgE-binding epitopes on Ara h6 using a format of EAST in which the protein is immobilised in a
particular, well defined, manner through interactions with specific monoclonal antibodies (mAbs) coated on the micro-plates.
The fine specificity of those mAbs has been characterised at the epitope level, and their binding to the allergen thus masks
a known particular epitope and makes it unavailable for recognition by IgE antibodies. The reverse EAST increased the ratio
specific signal/background. It avoids interferences with competitors such as anti-peanut protein IgG antibodies and allows
the study of the specificity and/or affinity of the interactions between IgE antibodies and Ara h2 or Ara h6 with a higher
sensitivity and accuracy than the conventional EAST. The EAST results obtained when the allergens are presented by specific
mAbs suggest that the homologous molecular domain(s) in peanut 2S albumins encompass major IgE epitope(s) and are strongly

The synthesis of novel meso-/macroporous SiO2 monoliths by combining a nano-building-blocks-based approach with the confined geometry of a tailored air-liquid foam structure is described. The resulting macrostructure in which ordered close-packed colloidal silica nanoparticles constitute the monolith's scaffolds very closely resembles the tailored periodic air-liquid foam template. The void spaces between adjacent particles create textural mesoporosity; therefore, the as-prepared silica networks are characterized by hierarchical porosity at the macroscopic and mesoscopic length scales. The fine-tuning of both the liquid foam's fraction and the bubble size allows a rational design over the macroscopic cell morphologies (shape, Plateau border's length, and width). Striking results of this approach are the weak shrinkage of the as-synthesized opal-like scaffolds during the thermally induced sintering process and, in contrast with previous studies, the formation of closed-cell structures. Particle organization and the foam film surface roughness are investigated by atomic force microscopy (AFM), showing the influence of the liquid flow, within the foams' Plateau borders and films, on the final assemblies. 相似文献
We experimentally demonstrate that continuous-wave supercontinuum generation in optical fibers can be significantly enhanced by using both multiwavelength pumping and dispersion management. We show by detailed spectral analysis that continuum enhancement is achieved mainly through a combination of Raman-assisted modulation instabilities, soliton compression, and dispersive wave generation. With this technique, an 800 nm wide (from 1.2 to 2.0 microm) 2 W supercontinuum source is reported that uses a three-wavelength pump and a dispersion-tailored four-optical fibers arrangement. 相似文献
The layer-multiple-scattering method is developed to study wave propagation through two-dimensional lattices of cylindrical inclusions in an elastic medium. The lattices are a series of periodically spaced infinite one-dimensional periodic gratings (or rows) of inclusions. The layer-multiple-scattering method allows the analysis of the reflection and transmission properties of the two-dimensional lattice, provided those of each row are known. These are later determined by means of an exact multiple scattering formalism based on modal series developments. A new characteristic equation is obtained that describes the Bloch wave propagation into the infinite lattice. Lattices with empty and fluid-filled inclusions are compared. The comparison shows the existence of pass and stop bands due to the resonances of the fluid-filled inclusions. Resonant inclusions allow the opening of narrow pass bands inside phononic stop band, which is an interesting phenomenon for demultiplexing problems. It is worth noting that inclusion resonances have nothing to do with resonances due to defects, as they involve the whole lattice. In addition, it is shown that stop bands, at an oblique incidence, due to a strong coupling between longitudinal and transverse waves, are related to dispersive guided waves that propagate in the direction of the reticular planes of the lattices. 相似文献
