Monolithic capillary columns were prepared by copolymerization of styrene and divinylbenzene inside a 200 microm i.d. fused silica capillary using a mixture of tetrahydrofuran and decanol as porogen. Important chromatographic features of the synthesized columns were characterized and critically compared to the properties of columns packed with micropellicular, octadecylated poly(styrene-co-divinylbenzene) (PS-DVB-C18) particles. The permeability of a 60 mm long monolithic column was slightly higher than that of an equally dimensioned column packed with PS-DVB-C18 beads and was invariant up to at least 250 bar column inlet pressure, indicating the high-pressure stability of the monolithic columns. Interestingly, monolithic columns showed a 3.6 times better separation efficiency for oligonucleotides than granular columns. To study differences of the molecular diffusion processes between granular and monolithic columns, Van Deemter plots were measured. Due to the favorable pore structure of monolithic columns all kind of diffusional band broadening was reduced two to five times. Using inverse size-exclusion chromatography a total porosity of 70% was determined, which consisted of internodule porosity (20%) and internal porosity (50%). The observed fast mass transfer and the resulting high separation efficiency suggested that the surface of the monolithic stationary phase is rather rough and does not feature real pores accessible to macromolecular analytes such as polypeptides or oligonucleotides. The maximum analytical loading capacity of monolithic columns for oligonucleotides was found to be in the region of 500 fmol, which compared well to the loading capacity of the granular columns. Batch-to-batch reproducibility proved to be better with granular stationary phases compared to monolithic stationary phase, in which each column bed is the result of a unique column preparation process. 相似文献

An elliptically polarized light scattering (EPLS) technique for the size and shape characterization of ellipsoidal hematite particles is presented. The hematite particles are synthesized by aging aqueous FeCl(3) or Fe(NO(3))(3) solutions at 100 degrees C. Different reaction conditions create particles of aspect ratios between 1 (spherical) and 8 (elongated). Cross-sectional diameter and aspect ratio are observed as a function of reaction (aging) time. Growth of the elongated particles, as well as their fractal aggregation behavior, is characterized using EPLS and comparisons are made with photon correlation spectroscopy and TEM measurements. 相似文献

The rates of the epoxidation reactions of aldehydes, of the aziridination reactions of aldimines, and of the cyclopropanation reactions of α,β-unsaturated ketones with aryl-stabilized dimethylsulfonium ylides have been determined photometrically in dimethyl sulfoxide (DMSO). All of these sulfur ylide-mediated cyclization reactions as well as the addition reactions of stabilized carbanions to N-tosyl-activated aldimines have been shown to follow a second-order rate law, where the rate constants reflect the (initial) CC bond formation between nucleophile and electrophile. The derived second-order rate constants (log k(2)) have been combined with the known nucleophilicity parameters (N, s(N)) of the aryl-stabilized sulfur ylides 4a,b and of the acceptor-substituted carbanions 4c-h to calculate the electrophilicity parameters E of aromatic and aliphatic aldehydes (1a-i), N-acceptor-substituted aromatic aldimines (2a-e), and α,β-unsaturated ketones (3a-f) according to the linear free-energy relationship log k(2) = s(N)(N + E) as defined in J. Am. Chem. Soc.2001, 123, 9500-9512. Enantiotopic discrimination in the NMR spectra of prochiral rigid solutes in chiral liquid crystals (CLC), by the ordering mechanism, is limited to molecules possessing one of the four, so called, "allowed" symmetries, D(2d), C(2v), C(s), and S(4). So far, such spectral discrimination was demonstrated only for solutes possessing one of the first three symmetries. In this work, we present deuterium NMR measurements on a rigid S(4) compound dissolved in a chiral nematic solvent and demonstrate, for the first time, enantiotopic discrimination in such symmetry. The measurements were performed on the isotopically normal icosane derivative (1) and on its isotopomer (1-d(8)), specifically deuterated in its four core methylene groups. As a CLC solvent, a lyotropic mesophase, consisting of a solution of poly-γ-benzyl-L-glutamate (PBLG) in pyridine, was employed. For comparison with a corresponding achiral liquid crystal (ALC) solvent, a solution of a racemic mixture of poly-γ-benzylglutamate (PBG) of similar composition in the same co-solvent was used. The spectra were recorded at 92.1 MHz using the 2D Q-COSY Fz sequence with proton decoupling. In the CLC solvents they exhibited clear discrimination due to different enantiotopic sites, with components displaced symmetrically, at frequencies below and above those in the corresponding ALC, as expected for discrimination by ordering. Two procedures were employed for correlating the enantiotopic sites in the CLC spectra. For 1-d(8) the dipolar cross-peaks in a 2D (2)H-(2)H COSY-90 experiment provided identification of signals belonging to the same methylene (and hence the same enantiotopic) groups. For 1 the correlation was achieved using a least-square-deviation fitting of the experimental quadrupole splittings with respect to those expected from the molecular geometry. These results, with appropriate symmetry considerations were used to determine the symmetric (S(zz)) and antisymmetric (S(xy) and S(xx)-S(yy)) components of the Saupe ordering matrix. Interpretation of the NMR spectra of prochiral solutes in CLC suffers from the ambiguity in identifying the signals with specific enantiotopic groups. For this reason only the relative (but not the absolute) signs of the antisymmetric elements of the ordering matrix can be determined. For the S(4) group this leads to sign ambiguity in the rhombic term in the diagonalized ordering matrix. Similar (but not identical) ambiguities occur for solutes belonging to the other allowed groups. In a concluding section of the paper, the ambiguities in the antisymmetric order parameters for the various allowed groups are compared and their physical meaning are discussed. 相似文献

An optical biosensor for the determination of hydrogen peroxide based on immobilized horseradish peroxidase is described. The fluorescence of the dimeric product of the enzyme catalysed oxidation of homovanillic acid is utilized to determine the concentration of H2O2. The membrane-bound enzyme is attached to a bifurcated fibre bundle permitting excitation and detection of the fluorescence by a fluorometer. The response of the sensor is linear from 1 to 130 M hydrogen peroxide; the coefficient of variation is 3%. The sensor is stable for more than 10 weeks. The operating pH for maximal sensor response is 8.15. This allows the sensor to be used in combination with oxidase reactions producing hydrogen peroxide, as is demonstrated with a co-immobilized lactate oxidase-horseradish peroxidase optode for the determination of L-lactate. The fluorescence intensity of this sensor depends linearly on the concentration of lactate between 3 and 200 M and a throughput of 10 samples per hour is possible. The precision is in the same range as that of the monoenzyme optode. The lifetime of the bienzyme sensor for lactate is considerably shorter than that of the peroxidase sensor; it is limited by the stability of the immobilized lactate oxidase enzyme. The determination of extractable trace metal contents in soil using single extraction procedures is currently performed in many laboratories to assess the bioavailable metal fraction (and related phytotoxic effects) and the accessability to the environment (e.g. contamination of ground waters). Owing to the need for validation of the extraction schemes used and of the analytical techniques, the EC Measurements and Testing Programme (formerly BCR) has organized a project for improving the quality of determination of extractable trace metal contents in soil, the first step being an interlaboratory study to adopt common extraction procedures and the second being a certification campaign to certify two soils for their extractable trace element contents following these procedures. The reaction of N-heterocyclic silylene (NHSi) L [L = CH{(C[double bond, length as m-dash]CH(2))(CMe)(2,6-iPr(2)C(6)H(3)N)(2)}Si] with benzoylhydrazine, 1,2-dicarbethoxyhydrazine, 1,2-diacetylhydrazine and 1,2-bis(tert-butoxycarbonyl)hydrazine in 1 : 1 molar ratio resulted in compounds 1-4 with an almost quantitative yield and five coordinate silicon atoms. Compounds 1-4 were formed by double N-H bond activation by deliberate selection of N,N'-bis-substituted hydrazine compounds bearing the -C(O)NHNH- unit. Compounds 1-4 were characterized by NMR spectroscopy, EI-MS and elemental analysis. The molecular structures of compounds 1-3 were unambiguously established by single crystal X-ray structural analysis. 相似文献
