Two linear isomers, OPN and ONP, were formed in an Ar matrix at 16 K by ArF laser (λ = 193 nm) photolysis of phosphoryl triazide, OP(N(3))(3), and comprehensively characterized by their mid- and far-IR spectra as well as (14/15)N isotopic data. Two intermediates, (N(3))(2)P(O)N and N(3)P(O)N(4), have been isolated in an Ar matrix and spectroscopically characterized. Photolysis of these intermediates using near-UV/vis light (λ > 320 nm) was shown to yield exclusively ONP, which under ArF laser irradiation selectively photo-isomerized to OPN. The photochemical and molecular properties of OPN and ONP are discussed on the basis of experimental and ab initio calculated results.  相似文献

Soluble epoxide hydrolase (sEH) is a promising therapeutic target for the treatment of hypertension, pain, and inflammation-related diseases. In order to enable the development of sEH inhibitors (sEHIs), assays are needed for determination of their potency. Therefore, we developed a new method utilizing an epoxide of arachidonic acid (14(15)-EpETrE) as substrate. Incubation samples were directly injected without purification into an online solid phase extraction (SPE) liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS-MS) setup allowing a total run time of only 108 s for a full gradient separation. Analytes were extracted from the matrix within 30 s by turbulent flow chromatography. Subsequently, a full gradient separation was carried out on a 50X2.1 mm RP-18 column filled with 1.7 μm core-shell particles. The analytes were detected with high sensitivity by ESI-MS-MS in SRM mode. The substrate 14(15)-EpETrE eluted at a stable retention time of 96 ± 1 s and its sEH hydrolysis product 14,15-DiHETrE at 63 ± 1 s with narrow peak width (full width at half maximum height: 1.5 ± 0.1 s). The analytical performance of the method was excellent, with a limit of detection of 2 fmol on column, a linear range of over three orders of magnitude, and a negligible carry-over of 0.1% for 14,15-DiHETrE. The enzyme assay was carried out in a 96-well plate format, and near perfect sigmoidal dose-response curves were obtained for 12 concentrations of each inhibitor in only 22 min, enabling precise determination of IC(50) values. In contrast with other approaches, this method enables quantitative evaluation of potent sEHIs with picomolar potencies because only 33 pmol L(-1) sEH were used in the reaction vessel. This was demonstrated by ranking ten compounds by their activity; in the fluorescence method all yielded IC(50) ≤ 1 nmol L(-1). Comparison of 13 inhibitors with IC(50) values >1 nmol L(-1) showed a good correlation with the fluorescence method (linear correlation coefficient 0.9, slope 0.95, Spearman's rho 0.9). For individual compounds, however, up to eightfold differences in potencies between this and the fluorescence method were obtained. Therefore, enzyme assays using natural substrate, as described here, are indispensable for reliable determination of structure-activity relationships for sEH inhibition.  相似文献

In this paper, we report the most comprehensive equilibrium superacidity scale that is available to date. Contrary to most of the past works, this scale is set up in a medium of constant composition and the obtained acidity values characterize the acidities of molecules rather than acidities of media. The current scale is thus complementary to the well-known H(0) scale in the information that it provides. The solvent used is 1,2-dichloroethane (DCE). DCE has very weak basic properties (but sufficiently high polarity) and is an appropriate solvent for measuring acidities of very strong acids of diverse chemical nature. DCE acidities of well-known superacids (CF(3)SO(2)OH, (CF(3)SO(2))(2)NH, cyanocarbon acids, etc.) as well as common mineral acids (H(2)SO(4), HI, HBr, etc.) are reported. Acidities of altogether 62 acids have been determined from 176 interlinked relative acidity measurements. The scale spans 15 orders of magnitude (from picric acid to 1,1,2,3,3-pentacyanopropene) and is expected to be a useful tool in design, use, and further acidity measurements of superacidic molecules.  相似文献

A new group of porphyrin-fullerene dyads with an azobenzene linker was synthesized, and the photochemical and photophysical properties of these materials were investigated using steady-state and time-resolved spectroscopic methods. The electrochemical properties of these compounds were also studied in detail. The synthesis involved oxidative heterocoupling of free base tris-aryl-p-aminophenyl porphyrins with a p-aminophenylacetal, followed by deprotection to give the aldehyde, and finally Prato 1,3-dipolar azomethineylide cycloaddition to C60. The corresponding Zn(II)-porphyrin (ZnP) dyads were made by treating the free base dyads with zinc acetate. The final dyads were characterized by their 1H NMR, mass, and UV-vis spectra. 3He NMR was used to determine if the products are a mixture of cis and trans stereoisomers, or a single isomer. The data are most consistent with the isolation of only a single configurational isomer, assigned to the trans (E) configuration. The ground-state UV-vis spectra are virtually a superimposition of the spectral features of the individual components, indicating there is no interaction of the fullerene (F) and porphyrin (H2P/ZnP) moieties in the ground state. This conclusion is supported by the electrochemical data. The steady-state and time-resolved fluorescence spectra indicate that the porphyrin fluorescence in the dyads is very strongly quenched at room temperature in the three solvents studied: toluene, tetrahydrofuran (THF), and benzonitrile (BzCN). The fluorescence lifetimes of the dyads in all solvents are sharply reduced compared to those of H2P and ZnP standards. In toluene, the lifetimes of the free base dyads are 600-790 ps compared to 10.1 ns for the standard, while in THF and BzCN the dyad lifetimes are less than 100 ps. For the ZnP dyads, the fluorescence lifetimes were 10-170 ps vs 2.1-2.2 ns for the ZnP references. The mechanism of the fluorescence quenching was established using time-resolved transient absorption spectroscopy. In toluene, the quenching process is singlet-singlet energy transfer (k approximately 10(11) s-1) to give C60 singlet excited states which decay with a lifetime of 1.2 ns to give very long-lived C60 triplet states. In THF and BzCN, quenching of porphyrin singlet states occurs at a similar rate, but now by electron transfer, to give charge-separated radical pair (CSRP) states, which show transient absorption spectra very similar to those reported for other H2P-C60 and ZnP-C60 dyad systems. The lifetimes of the CSRP states are in the range 145-435 ns in THF, much shorter than for related systems with amide, alkyne, silyl, and hydrogen-bonded linkers. Thus, both forward and back electron transfer is facilitated by the azobenzene linker. Nonetheless, the charge recombination is 3-4 orders of magnitude slower than charge separation, demonstrating that for these types of donor-acceptor systems back electron transfer is occurring in the Marcus inverted region.  相似文献

The new tetracyanoborate K[B(CN)4]·CH3CN was synthesized by dissolution of the solvent‐free K[B(CN)4] in acetonitrile and subsequent careful crystallization. The crystal structure has been determined by single‐crystal X‐ray diffraction. It crystallizes in the orthorhombic space group P212121 with Z = 4. The in vitro metabolic pattern of BAL19403, a novel macrolide antibiotic, was investigated by capillary liquid chromatography/quadrupole time-of-flight mass spectrometry (LC/QTOF-MS) in incubations with human microsomes. For the elucidation of the metabolic pathway, BAL19403 labeled with four deuterium atoms (D4) was used, and detection of metabolites performed using mixtures of the unlabeled (H4) BAL19403 and its D4 analogue (1:1) as substrate. All metabolites appeared with similar chromatographic behavior. MS/MS spectra of BAL19403 and its metabolites are dominated by non-informative fragment ions. Therefore, the structure of the metabolites was elucidated mainly by accurate mass measurements with subsequent proposals of elemental compositions. Main biotransformations were N-demethylation, lactone ring hydrolysis, and oxidation. Additionally, N-dealkylation of the aromatic moiety was identified. This dealkylation results not only in formation of an aldehyde, according to the classical pathway, but also in formation of the corresponding alcohol and carboxylic acid. Final elucidation of their structures was possible, since this dealkylation takes place vicinal to the deuterium-labeled part of BAL19403 and interferes with D/H exchange. The degree of D/H exchange, determined by analysis of the metabolite isotopic pattern, was used to elucidate the adjacent functional group.  相似文献
