Synthesis of carbazole derived aza[7]helicenes

Synthesis of four derivatives of symmetrical aza[7]helicenes is achieved by I₂-THF mediated photocyclization of corresponding bis-styryl derivatives. The cyclization furnished the desired angularly fused aza-helicene in moderate yields. The structures are established by NMR spectroscopy and single crystal X-ray analysis for the difluoro derivative. The series of synthesized aza[7]helicenes showed remarkable thermal stability as indicated by DSC analysis.     

Positive isotropic curvature and self-duality in dimension 4

We study a positivity condition for the curvature of oriented Riemannian 4-manifolds: the half-PIC condition. It is a slight weakening of the positive isotropic curvature (PIC) condition introduced by M. Micallef and J. Moore. We observe that the half-PIC condition is preserved by the Ricci flow and satisfies a maximality property among all Ricci flow invariant positivity conditions on the curvature of oriented 4-manifolds. We also study some geometric and topological aspects of half-PIC manifolds.     

Process and purification for manufacture of a modified insulin intended for oral delivery

Oral delivery of insulin is convenient and physiologically desirable in the treatment of diabetes. However, this route of administration has presented substantial challenges as insulin is degraded enzymatically in the gut, resulting in low bioavailability. We have developed a conjugated insulin product (IN-105) that has high bioavailability and is currently undergoing clinical trials for the treatment of diabetes. A process for the manufacture of IN-105 was developed. Initially, recombinant human insulin was conjugated covalently with a monodisperse, short-chain methoxypolyethylene glycol derivative. The desired product, IN-105, was purified from its closely related species using RP-HPLC and cation exchange chromatography to a purity of 98.5%. The elution pool from cation exchange chromatography was crystallized and lyophilized into the dry active pharmaceutical ingredient.     

Synthesis of Some Newer Analogues of 4‐Hydroxyphenyl Acetic Acid as Potent Anti‐Inflammatory Agents

A series of 1,3,4‐oxadiazole and 1,2,4‐triazole derivatives of 4‐hydroxyphenyl acetic acid have been synthesized and evaluated for their anti‐inflammatory activity by carrageenan induced rat paw edema method. The compounds, which showed good anti‐inflammatory activity, were screened for their ulcerogenic and lipid peroxidation activities.     

Procedure for chromatography involving sample solvent with higher elution strength than the mobile phase

In preparative chromatography, often the solubility of the sample in the mobile phase is limited, making the mobile phase unsuitable as a solvent for preparation of load. Generally, solvents that have high solubility for the sample also have higher elution strengths than the mobile phase. Additionally, at high loading volumes, these strong sample solvents are known to adversely affect the band profiles leading to poor chromatographic performance. Here, we show that controlling the mobile phase strength during loading and post-load elution resulted in improved band profiles when the sample solvent was stronger than the mobile phase. Such an approach improves performance in preparative chromatography by allowing either higher sample loading or higher organic content in mobile phase (without loss of yield). Alternately, the approach can be used for improvement in performance by increase in yield or product purity.     

Positive scalar curvature and minimal hypersurfaces

We show that the minimal hypersurface method of Schoen and Yau can be used for the ``quantitative' study of positive scalar curvature. More precisely, we show that if a manifold admits a metric with or , where is the scalar curvature of , any 2-tensor on and the Weyl tensor of , then any closed orientable stable minimal (totally geodesic in the second case) hypersurface also admits a metric with the corresponding positivity of scalar curvature. A corollary pertaining to the topology of such hypersurfaces is proved in a special situation.
