Peptide-based molecular probes identified by bacteriophage (phage) display technology expand the peptide repertoire for in vivo diagnosis and therapy of cancer. Numerous peptides that bind cancer-associated antigens have been discovered by panning phage libraries. However, until now only few of the peptides selected by phage display have entered clinical applications. The success of phage derived peptides essentially depends on the quality of the library screened. This review summarizes the methods to achieve highly homogenous libraries that cover a maximal sequence space. Biochemical and chemical strategies for the synthesis of DNA libraries and the techniques for their integration into the viral genome are discussed in detail. A focus is set on the methods that enable the exclusion of disturbing sequences. In addition, the parameters that define the variability, the minimal numbers of copies per library and the use of alternating panning cycles to avoid the loss of selected hits are evaluated.  相似文献

The reactivity of aryl triflates in on-surface C−C coupling is reported. It is shown that the triflate group in aryl triflates enables regioselective homo coupling with preceding or concomitant hydrodetriflation on Cu(111). Three different symmetrical π-systems with two and three triflate functionalities were used as monomers leading to oligomeric conjugated π-systems. The cascade, comprising different intermediates at different reaction temperatures as observed for one of the molecules, proceeds via initial removal of the trifluoromethyl sulfonyl group to give an aryloxy radical which in turn is deoxygenated to the corresponding aryl radical. Thermodynamically driven regioselective 1,2-hydrogen atom transfer leads to a translocated aryl radical which in turn undergoes coupling. For a sterically more hindered bistriflate, where one ortho position was blocked, dehydrogenative coupling occurred at remote position with good regioselectivity. Starting materials, intermediates as well as products were analyzed by scanning tunneling microscopy. Structures and suggested mechanism were further supported by DFT calculations.  相似文献

Liquid chromatography in combination with spectroscopic methods like matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) or nuclear magnetic resonance (NMR) spectroscopy is a powerful method to characterize silsesquioxanes and silsesquioxane mixtures. As new examples, the formation of silsesquioxyl-substituted silsesquioxanes [(n-octyl)(7)(SiO(1.5))(8)](2)O and [(n-octyl)(7)(SiO(1.5))(8)O](2)[(n-octyl)(6)(SiO(1.5))(8)] as well as the cage rearrangement of octa-[(n-heptyl)silsesquioxane] to larger structures [(n-heptyl)SiO(1.5))](n) up to n=28 are shown.  相似文献

We have examined two-dimensional electrophoresis (2-DE) gel maps of polypeptides from the Gram-negative bacterium Methylococcus capsulatus (Bath) and found the same widespread trains of spots as often reported in 2-DE gels of polypeptides of other Gram-negative bacteria. Some of the trains of polypeptides, both from the outer membrane and soluble protein fraction, were shown to be generated during the separation procedure of 2-DE, and not by covalent post-translational modifications. The trains were found to be regenerated when rerunning individual polypeptide spots. The polypeptides analysed giving this type of trains were all found to be classified as stable polypeptides according to the instability index of Guruprasad et al. (Protein Eng. 1990, 4, 155-161). The phenomenon most likely reflects conformational equilibria of polypeptides arising from the experimental conditions used, and is a clear drawback of the standard 2-DE procedure, making the gel picture unnecessarily complex to analyse.  相似文献

The yellowing and IR-changes of spruce wood as a result of UV-irradiation were studied using two different types of xenon lamps (lambda>300 nm; I(o)=50 mW cm(-2) and lambda>280 nm; I(o)=17.5 mW cm(-2)). Changes in the IR spectra as well as the yellowing of the irradiated wood surfaces show the influence of UV light on the wood modules. The UV-irradiation (72 h; lambda>300 nm; I(o)=50 mW cm(-2)) decreased the lignin content on the surface by up to 20% of the original values. The colour difference of yellowing (deltaE) exhibited a systematic trend to higher values with increasing irradiation time. Our results show that the photoyellowing (UV-Vis detection) correlates very well with lignin degradation (IR detection). This result is in agreement with the quinone formation as the chromophoric reaction product of lignin decay. The degradation, yellowing, and oxidation kinetics differed only little using different light sources. The absorbed light intensity, which depends on wavelength, the intensity distribution of the light source and the absorption spectrum of lignin, influenced the degradation rate. Under the current experimental conditions, the absorption spectrum of lignin was the most important factor. High-temperature solvent gradient interaction chromatography (HT-SGIC) is a fast and efficient fractionation technique for the chemical composition analysis of olefin copolymers. The separation of ethylene–propylene random copolymers (EPRs) was achieved on a graphitic stationary phase, Hypercarb, at 160 °C by using linear solvent gradient elution from 1-decanol to 1,2,4-trichlorobenzene (TCB). In the present work, the solvent gradient profile was modified to improve the chromatographic separation of EPRs. With the aim to obtain a better resolution in separation, a slow increase in the volume fraction of TCB was applied. This allowed for a relatively large retention region for linear polyethylene (PE) chains on the column; thereby, a broader elution volume zone between the start of the gradient and the PE elution was achieved. The efficiency of this new gradient profile was demonstrated by analysing two fully amorphous EPR samples. Clear differences in the chemical composition of these EPR samples with similar ethylene contents have been proven by using this modified solvent gradient. The comprehensive chemical composition and microstructure analysis of the SGIC-separated fractions by FTIR revealed that ethylene/propylene (EP) copolymer chains were eluted according to their ethylene/propylene contents and E or P sequence lengths, even though they are distributed in a random manner. These results showed that the solvent composition is an important factor to affect the interactive adsorption or desorption behaviour of EP chains on Hypercarb. In this way, for the first time, the determination of the complex composition and chain structure of EPR samples was achieved within short analysis time, which is not possible till now using other fractionation techniques reported.

Asymmetrical flow field-flow fractionation (AF4) was used as a fractionation technique to investigate the molecular heterogeneity of poly(styrene-b-isoprene) diblock copolymers synthesized by either sequential living anionic polymerization or coupling of living precursor blocks. AF4 coupled to multi-angle laser light scattering (MALLS), refractive index (RI), and ultraviolet (UV) detectors was used to separate the diblock copolymers from the homopolymers and coupling products, and the molar masses of the different components were analyzed. In order to get more information about the separated block copolymers, homopolymers, and coupling products, fractions were collected directly after the AF4 channel. The collected fractions were analyzed offline by 1H NMR to provide identification of the different species and additional information on the true chemical composition, and the microstructure of the diblock copolymer was obtained.

Abstract

The synthesis of a new disc-like mesogenic compound permitted, for the first time, ferroelectric electrooptical switching in a tilted columnar liquid crystal. The spontaneous molecular dipole moment is induced by the bend between the dibenzopyrene core and part of the eight O-hexyllactic acid chains attached to it. Two similar new compounds displayed a weak electrooptical effect in the columnar phase.  相似文献
