Analysis of cocaine and its principal metabolites in waste and surface water using solid-phase extraction and liquid chromatography–ion trap tandem mass spectrometry

A validated method based on solid-phase extraction (SPE) and liquid chromatography-ion trap tandem mass spectrometry (LC-MS/MS) is described for the determination of cocaine (COC) and its principal metabolites, benzoylecgonine (BE) and ecgonine methyl ester (EME), in waste and surface water. Several SPE adsorbents were investigated and the highest recoveries (95.7 +/- 5.5, 91.8 +/- 2.2 and 72.5 +/- 5.3% for COC, BE and EME, respectively) were obtained for OASIS HLB(R) cartridges (6 mL/500 mg) using 100 mL of waste water or 500 mL of surface water. Extracts were analysed by reversed-phase (RP) or hydrophilic interaction (HILIC) LC-MS/MS in positive ion mode with multiple reactions monitoring (MRM); the latter is the first reported application of the HILIC technique for drugs of abuse in water samples. Corresponding deuterated internal standards were used for quantification. The method limits of quantification (LOQs) for COC and BE were 4 and 2 ng L(-1), respectively, when RPLC was used and 1, 0.5 and 20 ng L(-1) for COC, BE and EME, respectively, with the HILIC setup. For COC and BE, the LOQs were below the concentrations measured in real water samples. Stability tests were conducted to establish the optimal conditions for sample storage (pH, temperature and time). The degradation of COC was minimal at -20 degrees C and pH = 2, but it was substantial at +20 degrees C and pH = 6. The validated method was applied to a set of waste and surface water samples collected in Belgium.

Two‐Photon Absorption and Time‐Resolved Stimulated Emission Depletion Spectroscopy of a New Fluorenyl Derivative

The synthesis, comprehensive linear photophysical characterization, two‐photon absorption (2PA), steady‐state and time‐resolved stimulated emission depletion properties of a new fluorene derivative, (E)‐1‐(2‐(di‐p‐tolylamino)‐9,9‐diethyl‐9H‐fluoren‐7‐yl)‐3‐(thiophen‐2‐yl)prop‐2‐en‐1‐one ( 1 ), are reported. The primary linear spectral properties, including excitation anisotropy, fluorescence lifetimes, and photostability, were investigated in a number of aprotic solvents at room temperature. The degenerate 2PA spectra of 1 were obtained with open‐aperture Z‐scan and two‐photon induced fluorescence methods, using a 1 kHz femtosecond laser system, and maximum 2PA cross‐sections of ～400–600 GM were obtained. The nature of the electronic absorption processes in 1 was investigated by DFT‐based quantum chemical methods implemented in the Gaussian 09 program. The one‐ and two‐photon stimulated emission spectra of 1 were measured over a broad spectral range using a femtosecond pump–probe‐based fluorescence quenching technique, while a new methodology for time‐resolved fluorescence emission spectroscopy is proposed. An effective application of 1 in fluorescence bioimaging was demonstrated by means of one‐ and two‐photon fluorescence microscopy images of HCT 116 cells containing dye encapsulated micelles.     

Development and validation of an HPLC-MS/MS method to quantify clopidogrel acyl glucuronide, clopidogrel acid metabolite, and clopidogrel in plasma samples avoiding analyte back-conversion

A new sensitive and fast quantitative analytical method for the simultaneous determination of clopidogrel, its main metabolite clopidogrel carboxylic acid, and the newly described acyl glucuronide metabolite, in human plasma samples, is presented. The analytical procedures (plasma storage, handling, and extract storage in the autosampler) were optimized in order to avoid back-conversion; a known drawback in measurements of clopidogrel. Clopidogrel acyl glucuronide was confirmed as a major source of back-conversion to the parent drug in the presence of methanol, and thorough stability experiments were carried out to find the most appropriate conditions for an accurate analysis of clopidogrel and the two metabolites. The method was validated by assessing selectivity, sensitivity, linearity, accuracy, and precision for all three analytes, in accordance to Food and Drug Administration guidelines. Spiked quality controls in plasma as well as incurred samples were used to verify back-conversion in the selected conditions, with results meeting European Medicines Agency acceptance criteria (concentrations within 80–120% of the first reading). The method was then applied to a pharmacokinetic study, and for the first time, a pharmacokinetic curve of clopidogrel acyl glucuronide in human plasma is presented. The concentrations ranged up to 1,048.684 ng/mL, with a mean of 470.268 ng/mL, while clopidogrel had a mean C _max of 1.348 ng/mL; these orders of magnitude show how much the back-conversion of this metabolite may influence clopidogrel quantification if it is not properly controlled.     

Spectroscopic characteristics of Tm3+ in Tm and Tm,Nd, Yb:Sc2O3 ceramic

The spectroscopic investigation of Tm³⁺ or (Nd, Yb, Tm)-doped Sc₂O₃ transparent ceramics, as laser-active media for visible and IR emission, was performed. The spectra are dominated by Tm³⁺ ions in sites of C₂ symmetry and an energy level scheme and other spectral parameters were determined. The strong concentration quenching of ³H₄ level emission in Tm:Sc₂O₃ at 300 K is discussed by considering the resonant cross-relaxation (³H₄→³F₄)?(³H₆→³F₄) process and multipolar interactions of various orders. The main energy-transfer processes leading to the blue upconversion emission from the ¹G₄ Tm³⁺ level in (Tm, Nd, Yb):Sc₂O₃ ceramic, under pulsed pumping at 808.3 nm were evidenced.     

Surfaces Generation by Blending Interpolation on a Triangle with One Curved Side

We use some interpolation operators and some Bernstein type operators for construction of surfaces which satisfy some given conditions.     

Interpolation operators on a triangle with one curved side

We construct certain Lagrange, Hermite and Birkhoff-type operators, which interpolate a given function and some of its derivatives on the border of a triangle with one curved side, as well as some of their product and Boolean sum operators. We study the interpolation properties and the order of accuracy (degree of exactness and precision set) of the constructed operators, respectively the remainders of the corresponding interpolation formulas. Finally, we give some numerical examples.
