The first synthesis of a purine nucleoside analog containing a bridgehead nitrogen atom is here reported. The direct glycosylation of the trimethylsilyl derivative of s-triazolo[2,3-a] pyrimid-7-one has been shown to give 3-(β-D-ribofuranosyl)-s-triazolo[2,3-a]pyrimid-7-one (V) and 4-(β-D-ribof'uranosyl)-s-lriazolo[2,3-α]pyrimid-7-one (VII). The nueleoside V may he considered a close analog of inosine in which the nitrogen N1 and C5 of inosine have been interchanged. Bro-minalion of the tri-O-acelyl derivative IV gave, after deblocking, 6-bromo-3-(β-D-ribofurnaosyl)-s-triazolo[2,3-a] pyrimid-7-one (IX). Structural assignments of the nucleosides were made on the basis of comparison of the ultraviolet absorption spectral characteristics with 3-methyl-s-triazolo-[2,3-a]pyrimid-7-one (XI) and 4-methyl-s-lriazolo[2,3-a Jpyrimid-7-one (XII) prepared by a standard procedure from 7-methoxy-s-triazolo(2,3-a] pyrimidine (X). 相似文献

Thermal decomposition of configurationally pure 1,2-diarylpropenylcopper compounds Z-Vi2CU4Br2 and Z-Vi2Cu4R2 [Vi (2-Me2NC6H4)CC(Me)-(C6H4Me-4), R 2-Me2NC6H4 or 4-MeC6H4CC] predominantly results in the formation of ViH. In contrast, only dimers (ViVi) were formed on thermolysis of (Z-ViCu2OTf)η which is a further illustration of the influence of the counter anion on the reactivity of organocopper cluster compounds. However, in both cases partial inversion of configuration, giving mixtures of isomers, was observed. The thermolysis of (Z-ViCu2OTf)η is discussed in terms of OTf-enhanced intraaggregate electron-transfer processes. The formation of incipient vinyl cations which are η2-coordinated to the copper cluster can explain the observed isomerization.Also in the hydrolysis reaction of Z-Vi2Cu4Br2 mixtures of isomeric ViH compounds were obtained, the E/Z ratio being dependent on the type of reagent used. Mixtures of isomeric ViX compounds (X Br, Cl, I) were formed in the reaction of Z-Vi2Cu4Br2 with I2 and CuCl2. An explanation for the occurrence of isomerization is presented.Dimers (ViVi) were almost absent in the product mixture resulting from the reaction of Z-Vi2Cu4Br2 with CuCl2. In contrast, ViVi is obtained in about 50% yield from the reaction of ViLi with CuCl2 which is in accord with earlier observations for the reaction of aryllithium compounds with cupric halides.Highly selective E-ViBr formation was observed in the reaction of E-ViLi with AgBr. Site creation : Enantioselective artificial metalloenzymes have been created by grafting a new active site onto bovine pancreatic polypeptide through the introduction of an amino acid capable of coordinating a copper(II) ion. This hybrid catalyst gave good enantioselectivities in the Diels–Alder and Michael addition reactions in water (see scheme) and displayed a very high substrate selectivity.

We analytically calculate the gas-liquid critical endpoint (cep) for hard spheres with a Yukawa attraction. This cep is a boundary condition for the existence of a liquid. We use an analytical Helmholtz energy expression for the attractive Yukawa (hard) spheres based on the first-order mean spherical approximation to the attractive Yukawa potential by Tang and Lu (J. Chem. Phys. 1993, 99, 9828). This theory and our analytical simplification of it predict the gas-liquid and fluid-solid phase behavior, as found from computer simulations, very accurately as long as the range 1/kappa of attraction is not too short. We find that the cep is situated at kappasigma approximately 6 and at a contact potential around 2 kT. It follows that a liquid state is only possible when the attraction range is longer than (1/6) of the particle diameter sigma, and the attraction strength is smaller than 2 kT. The liquid region does not span more than 0.6 kT in strength, and there is also a relatively narrow window for the attraction range. 相似文献

Kinase-mediated resistance to antibiotics is a significant clinical challenge. These enzymes share a common protein fold characteristic of Ser/Thr/Tyr protein kinases. We screened 14 antibiotic resistance kinases against 80 chemically diverse protein kinase inhibitors to map resistance kinase chemical space. The screens identified molecules with both broad and narrow inhibition profiles, proving that protein kinase inhibitors offer privileged chemical matter with the potential to block antibiotic resistance. One example is the flavonol quercetin, which inhibited a number of resistance kinases in vitro and in vivo. This activity was rationalized by determination of the crystal structure of the aminoglycoside kinase APH(2″)-IVa in complex with quercetin and its antibiotic substrate kanamycin. Our data demonstrate that protein kinase inhibitors offer chemical scaffolds that can block antibiotic resistance, providing leads for co-drug design. 相似文献

Bile flow restoration is a crucial step in the recovery process post transplantation of the liver. Here, metabolic trajectories based on changes in bile secretion - a known marker of functionality - have been utilised as an approach for discovering bile fluxes during transplantation. A total of ten liver transplants were monitored and from these 68 bile samples from both donors and recipients were collected and analysed using ultra-performance LC-MS in combination with multivariate statistical analysis. Based on the principal component scores constructed from the total bile fingerprint, differentiation of the bile acid concentrations before and after transplantation was detected. A trend was also observed, by constructing metabolic trajectories, whereby the post-transplant profiles approached the position of pre-transplant profiles within 30-60?min of the restoration of bile secretion function. The ten major conjugated bile acid salts were measured and a significant increase in concentrations of taurocholic acid and taurochenodeoxycholic acid were seen after transplantation. In addition, the ratios of secondary bile acids detected in gall bladder and hepatic bile were measured before and after transplantation. This study suggests that bile acid ratios in the donor liver at the pre-transplant and post-transplant stage may be important and that profiling of secreted bile after transplantation may aid clinical assessment and progress post-transplantation. 相似文献

The reactivity of the bifunctionalized ligand NC(Br)N-I 1 [IC(6)H(2)(CH(2)NMe(2))(2)-3,5-Br-4] has been studied as a versatile synthon for organic and/or organometallic synthesis. Chemoselective metalation (M = Pd, Pt, Li) at the C(aryl)-I or C(aryl)-Br bonds was achieved by choosing the appropriate metal precursors. In this way a series of Pt(II) and Pd(II) complexes were prepared that have a second functional group available for further reactions. These Pt(II) and Pd(II) complexes were subjected to a wide range of organic and organometallic reactions, revealing the remarkable stability of their M-C sigma-bond and opening an easy route for the synthesis of mono- and (hetero)bimetallic building blocks. The scope of the chemistry of such building blocks shows that they are good candidates for use in the synthesis of dendrimers, bioorganometallic systems, or polymetallic materials. The X-ray crystal structures of the most representative complexes (2, 3a, 19, 20, and 24) are also reported. 相似文献

Studies on square planar iridium complexes of the type trans-Ir(PR(3))(2)(CO)Cl, where PR(3) is PhP[(C(5)Me(4))](2), PhP[Me(2)C(4)H(6)], or PhP[Pr(i)(2)C(4)H(6)], demonstrate that monodentate chiral phosphines impart exceptional degrees of diastereoselectivity in the oxidative addition of H(2). Thus, the oxidative addition of H(2) to the two faces of the meso isomer (R,S)-trans-Ir(PR(3))(2)(CO)Cl proceeds with a kinetic diastereoselectivity which exceeds that for related square planar iridium complexes employing bidentate chiral phosphine ligands. Furthermore, the kinetically favored dihydride is not favored thermodynamically, and the magnitude of the inversion of the kinetic and thermodynamic selectivities is greater than has previously been observed using bidentate phosphines. 相似文献
