There is considerable interest in maximising the amount of information obtained from animal product analyses, when screening for the presence of veterinary drug residues. One of the barriers to effective multi-residue analysis to date has been a lack of effective clean up procedures to isolate a wide range of residues from the potential interferents, which may be present in both simple and complex (including processed) foods. A cation-exchange clean up has, therefore, been developed for use with acetonitrile extracts of foods, when analysing for several basic drug groups (sulfonamides, benzimidazoles, levamisole, nitroimidazoles, tranquillisers and fluroquinolones). The clean up procedure has also been shown to be effective using a modified extraction solvent for malachite green and leucomalachite green in fish.Several of the key parameters that influence analyte recovery have been investigated and in an optimised procedure, tissue/biofluid samples containing sulfonamides, benzimidazoles, levamisole, nitroimidazoles, tranquillisers and fluoroquinolones are first extracted with acetonitrile. The extract is then dried with sodium sulfate and acidified with glacial acetic acid before loading onto a Bond Elut, strong cation-exchange (SCX) solid phase extraction (SPE) cartridge. Extracts from fish containing malachite green and leucomalachite green can be cleaned up using the same SCX SPE procedure following extraction with citrate buffer/acetonitrile. Typical recoveries of drugs from low level fortified tissues using the optimised procedure lie in the range 53-104% with the exception of carazolol from pig kidney (31%), malachite green from trout (42-51%) and ciprofloxacin from chicken muscle (44%) and from egg (21%).  相似文献

Salicylaldehyde reacts with dimethyl acetylenedicarboxylate in benzene solution to give a mixture of dimethyl o-formylphenoxymaleate, dimethyl o-formylphenoxyfumarate, 2,3-dicarbomethoxychrom - 2 - en - 4 - ol, 2,3 - dicarbomethoxychrom - 3 - en - 2 - ol, dimethyl (2,3 - dicarbomethoxychrom - 2 - en - 4 - yl) - oxalacetate and dimethyl fumarate. 2,3 - Dicarbomethoxychrom - 3 - en - 2 - ol in this reaction is formed from 2,3 - dicarbomethoxychrom - 2 - en - 4 - ol through a benzopyrylium intermediate. The reaction of salicylaldehyde with excess of dimethyl acetylenedicarboxylate, however, gives a mixture of 2,3 - dicarbomethoxychrom - 3 - en - 2 - ol and dimethyl (2,3 - dicarbomethoxychrom - 3 - en) - 2 - oxyfumarate. 2,3 - Dicarbomethoxychrom - 3 - en - 2 - ol itself reacts further with dimethyl acetylenedicarboxylate to give 2,3,12 - tricarbomethoxychrom - 3,4 - eno[2,3 - b] pyrone. Similarly, the reaction of o-hydroxyacetophenone with dimethyl acetylendicarboxylate gives a mixture of dimethyl o-acetylphenoxymaleate, dimethyl o-acetylphenoxyfumarate, 2,3 - dicarbomethoxy - 4 - methylchrom - 2 - en - 4 - ol and 2,3 - dicarbomethoxy - 4 - methylchrom - 3 - en - 2 - ol. Both 2-hydroxychalcone and 2 - hydroxy - 4′ - methoxychalcone give mixtures of chalcone - 2 - oxymaleate and chalcone - 2 - oxyfumarate. The reaction of 2,2t?-dihydroxychalcone, however, gives 2′-hydroxyflavone, in addition to the expected maleate and fumarate. Similar reactions of 5 - chloro - 2,2′ - dihydroxychalcone and of 3,5 - dibromo - 2,2′ - dihydroxychalcone, on the other hand, give 2,3 - dicarbomethoxy - 4 - (o - hydroxyphenacyl) - 6 - chlorochrom - 2 - ene and 2,3 - dicarbomethoxy - 4 - (o - hydroxyphenacyl) 6,8 - dibromochrom - 2 - ene, respectively, together with the corresponding maleates and fumarates.  相似文献

Soluble ethylene polymerization catalysts derived from (π-C5H5)2Ti(R)Cl and R ′AlCl2, where R = methyl or phenyl and R ′ = methyl or ethyl, were studied both by polymerization kinetics at 0°C and by diagnostic experiments. It was found that the first insertion of ethylene into the Ti? R bond is difficult when R = methyl or phenyl, and for this reason these catalysts show a different overall behavior than when R = ethyl or higher alkyl.  相似文献

Although previous studies have demonstrated that the predominant photochemistry of type I collagen under 254 nm irradiation may be attributed either to direct absorption by tyrosine/phenylalanine or to peptide bonds, direct collagen photochemistry via solar UV wavelengths is much more likely to involve several age- and tissue-related photolabile collagen fluorophores that absorb in the latter region. In this study, we compare and contrast results obtained from irradiation of a commercial preparation of acid-soluble calf skin type I collagen in solution with UVC (primarily 254 nm), UVA (335–400nm) and broad-band solar-simulating radiation (SSR; 290^1–00nm). Excitation spectroscopy and analysis of photochemically induced disappearance of fluorescence (fluorescence fading) indicates that this preparation has at least four photolabile fluorescent chromophores. In addition to tyrosine and L-3,4-dihydroxyphenylalanine, our sample contains two other fluorophores. Chromophore I, with emission maximum at 360 nm, appears to be derived from interacting aromatic moieties in close mutual proximity. Chromophore II, with broad emission at430–435 nm, may be composed of one or more age-related molecules. Collagen fluorescence fading kinetics are sensitive to excitation wavelength and to conformation. Under UVC, chromophore I fluorescence disappears with second-order kinetics, indicating a reaction between two proximal like molecules. Adherence to second-order kinetics is abrogated by prior denaturation of the collagen sample. A new broad, weak fluorescence band at400–420 nm, attributable to dityrosine, forms under UVC, but not under solar radiation. This band is photolabile to UVA and UVB wavelengths. Amino acid analysis indicates significant destruction of aromatic amino acids under UVC, but not under UVA or SSR. When properly understood, collagen fluorescence fading phenomena may act as a sensitive molecular probe of structure, conformation and reactivity.  相似文献

C7H7Mo(CO)(PN)I (I) (PN  (S)(+)-(C6H5)2PN(CH3)CH(CH3)(C6H5)) is prepared in 90% yield by reaction of C7H7Mo(CO)2I and PN. The two diastereo-isomers Ia and Ib differing only in the Mo-configuration exhibit chemical shift differences of their C7H7 and CH3 signals. Ia and Ib can be separated by fractional crystallization. In solution Ia epimerizes with respect to the Mo configuration. The half lives in benzene for the equilibration Ia ? Ib are 5.5, 30, and 104 min at 60, 50, and 40°C, respectively. Phosphine exchange experiments show that the epimerization proceeds via PN dissociation.An X-ray structure analysis was carried out on a single crystal of Ia. The absolute configuration at Mo was determined to be (R).  相似文献

Two tetrahydrofuran-based γ-amino acids [2,4-cis and 2,4-trans] were subjected to iterative peptide-coupling procedures to afford dimeric, tetrameric and hexameric carbopeptoids in good yield. These homooligomers were prepared for secondary structural study—to ascertain the conformational preference inherent in the monomer units. The l-xylo oligomers were protected with triethylsilyl ethers to increase the range of solvents suitable for structural investigation. Initial secondary structure data indicate the presence of hydrogen-bonded conformations in the l-ribo series.  相似文献

Capillary zone electrophoresis (CZE) and micellar electrokinetic chromatography (MEKC) were applied to the simultaneous separation of cyanobacterial toxins (anatoxin-a, microcystin-LR, cylindrospermopsin). The analytical performance data of both methods, optimized for the three toxins, were similar with a precision of migration times smaller than 0.8 RSD% and a detection limit in the range of 1-4 microg/mL, using spectrophotometric detection at 230 nm. Both methods were applied to an analysis of cyanotoxins in water bloom samples and crude cyanobacterial extracts. The results obtained indicate that, for complex matrices, the sequential application of CZE and MEKC is necessary. It is recommended to use both CE techniques for the analysis of the same sample in order to confirm the results by an orthogonal approach.  相似文献

The acid–base chemistry of some ruthenium ethyne-1,2-diyl complexes, [{Ru(CO)2(η-C5H4R)}22-CC)] (R=H, Me) has been investigated. Initial protonation of [{Ru(CO)2{η-C5H4R}}22-CC)] gave the unexpected complex cation, crystallised as the BF4 salt, [{Ru(CO)2(η-C5H4R}}33-CC)][BF4] (R=Me structurally characterised). This synthesis proved to be unreliable but subsequent, careful protonation experiments gave excellent yields of the protonated ethyne-1,2-diyl complexes, [{Ru(CO)2{η-C5H4R)}2212-CCH)](BF4) (R=Me structurally characterised) which could be deprotonated in high yield to return the starting ethyne-1,2-diyl complexes.  相似文献

[reaction: see text] The enantioselective syntheses of 10-epi-anamarine and 5,10-epi,epi-anamarine have been achieved in 13 to 14 steps. The route relies upon an enantio- and regioselective Sharpless dihydroxylation of either dienoates or trienoates to establish the C-8 to C-11 stereochemistry. A diastereoselective Leighton allylation established the desired C-5 stereochemistry. The route also relies upon a ring-closing metathesis to establish the alpha,beta-unsaturated lactones.  相似文献

1,3-phenyl shifts interconvert imidoylketenes 1 and alpha-oxoketenimines 2 and, likewise, alpha-oxoketenes 3 automerize by this 1,3-shift. These rearrangements usually take place in the gas phase under conditions of flash vacuum thermolysis. Energy profiles calculated at the B3LYP/6-31G(d,p) and B3LYP/6311 + G(3df,2p)//B3LYP/6-31G(d,p) levels demonstrate that electron donating substituents (D) in the migrating phenyl group and electron withdrawing ones (W) in the non-migrating phenyl group, can stabilise the transition states TS1 and TS2 to the extent that activation barriers of ca. 100 kJ mol(-1) or less are obtained; i.e. enough to make these reactions potentially observable in solution at ordinary temperatures. The calculated transition state energies DeltaG(TS1) show an excellent correlation with the Hammett constants sigma(p)(W) and sigma(p+)(D).  相似文献
