Short‐chain carboxylic acids are relevant in pharmaceutical, food quality control, and biomedical analysis. In this study, 11 acids commonly found in drugs and in food products were selected. Wine was chosen as matrix for testing the method. The test compounds were used for comparing the selectivity of four 150 × 2.1 mm zwitterionic hydrophilic interaction LC (HILIC) columns (ZIC‐HILIC 5 μm, 200 Å, and 3.5 μm, 100 Å, ZIC‐pHILIC 5 μm, ZIC‐cHILIC 3 μm, 100 Å) while varying the conditions to optimize for low UV wavelength detection and achieve high sensitivity. Retention using potassium phosphate and ammonium carbonate as mobile‐phase components at pH 6.0, 7.5, and 8.5–8.9 was studied considering recent hypotheses on HILIC mechanism‐related with the Hofmeister series effect and ion hydration. An isocratic method with UV detection at 200 nm and mobile phase consisting of 75% acetonitrile and 10 mM potassium phosphate at pH 6.0 applied to a ZIC‐cHILIC column was found provisionally optimal and partially validated for the 11 analytes. Satisfactory results (R2 from 0.9940 to >0.9999), and recoveries from 93–106% for all analytes evidenced the method as suitable for wine analysis. To the best of our knowledge, no previous study has reported on the direct ZIC‐HILIC separation and UV detection of the acids considered here in wine. 相似文献

The evaporation in vacuo of the matrices used and the particle-induced desorption of matrix molecules in fast-atom bombardment (FAB) contribute to a proposed high pressure region above the FAB matrix known as the selvedge region. If the neutral number density is sufficiently high, ions formed upon bombardment may undergo collisions with molecules, yielding matrix-related cluster ions and, in cases when the analyte is desorbed in neutral form, protonated and deprotonated analyte molecules. Similarities with the chemical ionization (CI experiment have been pointed out previously and are further developed here. If FAB is similar to CI, then the response depends on the structures of the reagent ions — those ions that react with gas phase analyte molecules. We consider here the time dependence of positive and negative ion FAB spectra to attempt to identify the reagent ions of FAB. A model is suggested for the FAB ion source which evaluates similarities to a CI source, as well as spatial aspects that are unique to desorption/ionization techniques. 相似文献

Thymus vulgaris L. is one of the most commonly used medicinal and aromatic plants (MAPs), owing to a range of therapeutic properties of its essential oil. Plant growth, biomass yield, essential oil content and composition are influenced by chemotype, environmental conditions, cultivation techniques and vegetative development. Since in MAPs cultivation special attention is paid on high quality of raw material, the adoption of sustainable agriculture methods is of pivotal importance. Therefore, we evaluated the agronomic and qualitative performances of two Thymus vulgaris L. chemotypes, organically cultivated under the Mediterranean climate of hilly lands of central Italy for three consecutive years (2017–2019). Along the trial, total above-ground dry biomass significantly increased from the 1st to 3rd year after planting and large variations in the main biological, biometric and productive traits were observed between the two chemotypes. The ‘thymol’ chemotype EO obviously showed thymol as the major constituent (51.26–49.87%) followed by γ-terpinene and p-cymene. The ‘linalool’ chemotype EO showed high percentages of oxygenated monoterpenes (about 90%) with linalool (75%), linalyl acetate (8.15%) and b-caryophyllene (3.2%) as main constituents. This study highlighted that T. vulgaris can be successfully organically grown in the hilly lands of Tuscany, with interesting biomass and essential oil yields, even though the plants were in the initial years of crop establishment (start in 2017). The introduction of this species into organic cultivation systems could contribute to obtain high-quality raw material, as well as to enhance crop rotation diversification, which is of pivotal importance in the management of organic farms. 相似文献
