HPLC-based measurement of the chelator 1,2-dimethyl-3-hydroxy-pyrid-4-one (L1) and its iron complex for pharmacokinetic studies in humans

An improved HPLC based method to assay the oral active iron chelator 1,2-dimethyl-3-hydroxypyrid-4-one (L1, CP20) in serum and urine is described. The L1 peak has been well separated from other endogenous compounds, allowing the exact determination of the drug in both biological fluids. Moreover urinary iron excretion due to L1 therapy has been monitored by measuring urine Fe-(L1)₃ complex concentrations using reverse phase HPLC and subsequent detection at 450 nm. In patients and normal volunteers receiving this drug there is a good correlation between urine iron excretion measured by AAS and by the HPLC based method.

An Enantionselective Synthesis of L-Threonine An enantioselective synthesis of L-threonine ( 1 ) is described. Racemic ethyl 2-acetamido-3-oxobutyrate ( 6 ) was synthesized from ethyl acetoacetate ( 2 ) [4][5] and then transformed to the epimeric optically active alcohols 7a and 7b by microbiological reduction with Saccharomyces rouxii. The Mixture 7a/7b could be converted to 1 by slightly modified, known Methods in a yield of ca. 52% with respect to 7a/7b .

Summary DNA sequence analysis of the virus TTV1 identified in archaebacteriumThermoproteus tenax revealed an open reading frame possibly encoding for a protein. To assess the expression of this hypothetical protein F154 via immunochemical methods, related synthetic fragments should allow for production of protein specific antisera. For this purpose the dodecapeptide amide H-Thr-Pro-Thr-Pro-Thr-Pro-Thr-Pro-Ser-Pro-Thr-Pro-NH₂ related to a characteristic repeat in the protein primary structure was prepared by conventional methods of peptide synthesis.Dedicated to Prof. Dr. Karl Schlögl on occasion of his 65th birthday
