The mass spectra of fifteen 1,2-benzisothiazoles are reported; their fragmentation patterns have been investigated by labelling experiments, high resolution mass measurements and defocused metastable ion detection. The parent compound of the series, 1,2-benzisothiazole, eliminates HCN after partial inter-ring hydrogen scrambling. The extent of scrambling has been determined for normal daughter ions produced at different electron beam energies and also for metastable daughter ions, and is compared with the data reported for benzothiazole. Some 3-substituted derivatives show unusual fragmentation patterns. The mechanisms of these processes are under further investigation.  相似文献

Zusammenfassung Es wird eine einfache Methode zur Bestimmung von Molybdän durch indirekte komplexometrische Titration gegeben. Nach der Fällung des Molybdats mit Calciumchlorid bindet man das freie, in Lösung befindliche Calcium mit ÄDTA. Der Niederschlag wird nun ohne zu filtrieren mit einem gemessenen ÄDTA-Überschuß in der Hitze in Lösung gebracht und anschließend wird der ÄDTA-Rest mit Magnesiumlösung rücktitriert. Als Indicator verwendet man Eriochromschwarz T. Die verbrauchte ÄDTA-Menge ist dem Calcium des Calciummolybdats und damit dem Molybdän äquivalent. Der relative Fehler beträgt etwa 1%.  相似文献

Mounting evidence supports the role of amyloidogenesis, oxidative stress, and metal dyshomeostasis in the development of neurodegenerative disorders. Parkinson's Disease is characterized by α-synuclein (αSyn) accumulation and aggregation in brain regions, also promoted by Cu2+. αSyn is modified by reactive carbonyl species, including acrolein (ACR). Notwithstanding these findings, the interplay between ACR, copper, and αSyn has never been investigated. Therefore, we explored more thoroughly the effects of ACR on αSyn using an approach based on LC-MS/MS analysis. We also evaluated the influence of Cu2+ on the protein carbonylation and how the ACR modification impacts the Cu2+ binding and the production of Reactive Oxygen Species (ROS). Finally, we investigated the effects of ACR and Cu2+ ions on the αSyn aggregation by dynamic light scattering and fluorescence assays. Cu2+ regioselectively inhibits the modification of His50 by ACR, the carbonylation lowers the affinity of His50 for Cu2+ and ACR inhibits αSyn aggregation both in the presence and in the absence of Cu2+.  相似文献

Structure‐based drug development is often hampered by the lack of in vivo activity of promising compounds screened in vitro, due to low membrane permeability or poor intracellular binding selectivity. Herein, we show that ligand screening can be performed in living human cells by “intracellular protein‐observed” NMR spectroscopy, without requiring enzymatic activity measurements or other cellular assays. Quantitative binding information is obtained by fast, inexpensive 1H NMR experiments, providing intracellular dose‐ and time‐dependent ligand binding curves, from which kinetic and thermodynamic parameters linked to cell permeability and binding affinity and selectivity are obtained. The approach was applied to carbonic anhydrase and, in principle, can be extended to any NMR‐observable intracellular target. The results obtained are directly related to the potency of candidate drugs, that is, the required dose. The application of this approach at an early stage of the drug design pipeline could greatly increase the low success rate of modern drug development.  相似文献

We developed a new method for real‐time, three‐dimensional tracking of fluorescent particles. The instrument is based on a laser‐scanning confocal microscope where the focus of the laser beam is scanned or orbited around the particle. Two confocal pinholes are used to simultaneously monitor regions immediately above and below the particle and a feedback loop is used to keep the orbit centered on the particle. For moderate count rates, this system can track particles with 15 nm spatial resolution in the lateral dimensions and 50 nm in the axial dimension at a temporal resolution of 32 ms. To investigate the interaction of the tracked particles with cellular components, we have combined our orbital tracking microscope with a dual‐color, wide‐field setup. Dual‐color fluorescence wide‐field images are recorded simultaneously in the same image plane as the particle being tracked. The functionality of the system was demonstrated by tracking fluorescent‐labeled artificial viruses in tubulin‐eGFP expressing HUH7 cells. The resulting trajectories can be used to investigate the microtubule network with super resolution.  相似文献

For chemistry the theoretical representation of the forces connecting atoms in molecules was and is a central problem. The Atomic Orbital and the Molecular Orbital are basic building blocks in the Heitler–London (HL) and in the Linear Combination of Atomic Orbitals–Molecular Orbital (LCAO-MO) methods, which have lead to the construction of modern Valence Bond and Hartree–Fock methods (and related extensions). However, accurate predictions from non semi-empirical methods often require enormous amount of computer power, if applied to molecules of reasonable size and current chemical interest. We have critically re-examined the two basic methods and suggested a few extensions. Merging of the Hartree–Fock with the Heitler–London algorithms, as recently proposed in the Hartree–Fock–Heitler–London (HF–HL) method, reduces the length of the expansions needed in AO or MO ab initio models in the computation of binding energy; this simplification allows easy interpretation of the resulting wave function. The HF–HL method is exemplified with systematic computations on ground and excited state of the hydrides and homonuclear diatomic molecules with atoms of the first and second period of the periodic table. Further, we show that the HF–HL method is derivable from a wave function constructed with a new type of orbital, the Chemical orbital (CO), which embodies the characterization of MO near equilibrium, AO at dissociation and at the united atom. Preliminary computations with CO are included. The new method provides the conceptual origin of both the HF and VB approaches, thus the foundation of an 80 years effort in variational quantum chemistry.  相似文献

The complex Co2(CO)6[μ-η2-(H3CCCCH2CH2OH)] (1) with the ligand 3-pentyn-1-ol (pol) has been synthesized following established procedures. Its structure has been determined by X-ray analysis. The complex Co2(CO)6(mbo) (mbo = 2-methyl-3-butyn-2-ol, HCCC(CH3)2OH), (3), along with the already known Co2(CO)6(bud) (bud = 1,4-butyn-diol, HOCH2CCCH2OH) (2), and Co2(CO)8 were reacted with 2(diphenylphosphino)ethyl-triethoxysilane [Ph2PCH2CH2Si(OCH2CH3)3] (dpts) and tris(hydroxymethyl)phosphine [P(CH2OH)3] (thp). With dpts, mono- and di-substituted complexes were obtained: these were characterized by analytical and spectroscopic techniques. The structures of Co2(CO)6(dpts)2 (5) and of Co2(CO)4(pol)(dpts)2 (8) have been determined by X-ray analysis.Complex (1) was reacted with 3-(triethoxysilyl)propyl isocyanate [(H3CCH2O)3Si(CH2)3NCO] (tsi): the new complex Co2(CO)6[H3CCCCH2CH2OC(O)NH(CH2)3Si(OCH2CH3)3] (9) was obtained and spectroscopically characterized. The complex has also been reacted with tetraethyl orthosilicate (teos); a new inorganic-organometallic material was obtained. Complex (5) has been grafted on the mesoporous material SBA-15. The hybrid inorganic-organometallic materials obtained have been characterized by inductively coupled plasma-mass spectrometry (ICP-MS), infrared spectroscopy (FT-IR) under vacuum conditions, X-ray diffraction (XRD) and scanning electron microscopy coupled to EDS probe (SEM-EDS).  相似文献

Abstract  

The triruthenium complexes containing alkynols ligands have been reacted with 2(diphenylphosphino)ethyl-triethoxysilane (dpts), obtaining novel chemically stable disubstituted complexes. The same disubstituted Ru complexes above described were also synthesized following another approach based on the reaction from the phosphine substituted complex Ru3(CO)10(dpts)2 and the same alkynols ligands used to obtain the above triruthenium complexes. One of these stable complexes, the HRu3(CO)7[C≡C(Me)2(OH)](dpts)2 compound was grafted on the mesoporous silica SBA-15 and on γ-Al2O3 surface in order to obtain novel hybrid materials to employ in heterogeneous catalysis. The resulting materials have been characterized by multidisciplinary approach based on inductively coupled plasma-mass spectrometry (ICP-MS), FT-IR, XRD, physisorption measurements and DR-UV–Visible spectroscopy.  相似文献
