A promising way to interfere with biological processes is through the modulation of protein-protein interactions by means of small molecules acting as peptidomimetics. The 1,4-benzodiazepine scaffold has been widely reported as a peptide-mimicking, pharmacogenic system. While several synthetic pathways to C6-8 substituted benzodiazepines have been disclosed, few 1,4-benzodiazepines substituted at C9 have been reported. Herein, we describe a versatile approach to introduce cyclic, protonatable functionality at C8/C9. Introduction of the piperazine system at C8 and C9 gave access to a unique functionalization of the versatile benzodiazepine skeleton, broadening tailoring options on the benzofused side of the molecule, and the possibility of discovering novel peptidomimetics potentially able to modulate protein-protein interactions. Coupling of activated amino acids with poorly reactive anilines under mild conditions, while avoiding racemization, gave easy access to these compounds. Efficient amino acid activation was obtained by exploiting the rapid formation of acid chlorides under low temperature and acid/base free conditions, using triphenylphosphine and hexachloroacetone. This procedure successfully resulted in high reaction yields, did not produce racemization (ee > 98%, as demonstrated by using chiral solvating agents), and was compatible with the acid sensitive protecting groups present in the substrates. 相似文献
As the COVID-19 outbreak is developing the two most frequently reported statistics seem to be the raw confirmed case and case fatalities counts. Focusing on Italy, one of the hardest hit countries, we look at how these two values could be put in perspective to reflect the dynamics of the virus spread. In particular, we find that merely considering the confirmed case counts would be very misleading. The number of daily tests grows, while the daily fraction of confirmed cases to total tests has a change point. It (depending on region) generally increases with strong fluctuations till (around, depending on region) 15–22 March and then decreases linearly after. Combined with the increasing trend of daily performed tests, the raw confirmed case counts are not representative of the situation and are confounded with the sampling effort. This we observe when regressing on time the logged fraction of positive tests and for comparison the logged raw confirmed count. Hence, calibrating model parameters for this virus’s dynamics should not be done based only on confirmed case counts (without rescaling by the number of tests), but take also fatalities and hospitalization count under consideration as variables not prone to be distorted by testing efforts. Furthermore, reporting statistics on the national level does not say much about the dynamics of the disease, which are taking place at the regional level. These findings are based on the official data of total death counts up to 15 April 2020 released by ISTAT and up to 10 May 2020 for the number of cases. In this work, we do not fit models but we rather investigate whether this task is possible at all. This work also informs about a new tool to collect and harmonize official statistics coming from different sources in the form of a package for the R statistical environment and presents the “COVID-19 Data Hub.”
A bound on the growth of the velocity for the Vlasov–Poisson equation in a torus is given in one and two dimensions. The main tool used in the proof is a partition into fast and slow particles and the ergodic property of the free motion in a torus. 相似文献
Rising global populations and enhanced standards of living in so-called developing countries have led to an increased demand of food, in particular meat, worldwide. While increasing the production of broiler meat could be a potential solution to this problem, broiler meat is plagued by health concerns, such as the development of antimicrobial resistance and lower meat quality. For this reason, the supplementation of poultry feed with vitamins and antioxidant compounds, such as polyphenols, has become an attractive prospect for research in this sector. Such supplements could be obtained by extraction of agricultural byproducts (in particular, grape pomaces and artichoke leaves and bracts), thus contributing to reductions in the total amount of waste biomass produced by the agricultural industry. In this review, the effects of poultry feed supplementation with bioactive extracts from grape pomace (skins and/or seeds), as well as extracts from artichoke leaves and bracts, were explored. Moreover, the various methods that have been employed to obtain extracts from these and other agricultural byproducts were listed and described, with a particular focus on novel, eco-friendly extraction methods (using, for example, innovative and biocompatible solvents like Deep Eutectic Solvents (DESs)) that could reduce the costs and energy consumption of these procedures, with similar or higher yields compared to standard methods. 相似文献
We propose a method for effectively upscaling incompressible viscous flow in large random polydispersed sphere packings: the
emphasis of this method is on the determination of the forces applied on the solid particles by the fluid. Pore bodies and
their connections are defined locally through a regular Delaunay triangulation of the packings. Viscous flow equations are
upscaled at the pore level, and approximated with a finite volume numerical scheme. We compare numerical simulations of the
proposed method to detailed finite element simulations of the Stokes equations for assemblies of 8–200 spheres. A good agreement
is found both in terms of forces exerted on the solid particles and effective permeability coefficients. 相似文献
The Multi-Armed Bandit (MAB) problem has been extensively studied in order to address real-world challenges related to sequential decision making. In this setting, an agent selects the best action to be performed at time-step t, based on the past rewards received by the environment. This formulation implicitly assumes that the expected payoff for each action is kept stationary by the environment through time. Nevertheless, in many real-world applications this assumption does not hold and the agent has to face a non-stationary environment, that is, with a changing reward distribution. Thus, we present a new MAB algorithm, named f-Discounted-Sliding-Window Thompson Sampling (f-dsw TS), for non-stationary environments, that is, when the data streaming is affected by concept drift. The f-dsw TS algorithm is based on Thompson Sampling (TS) and exploits a discount factor on the reward history and an arm-related sliding window to contrast concept drift in non-stationary environments. We investigate how to combine these two sources of information, namely the discount factor and the sliding window, by means of an aggregation function . In particular, we proposed a pessimistic (), an optimistic (), as well as an averaged () version of the f-dsw TS algorithm. A rich set of numerical experiments is performed to evaluate the f-dsw TS algorithm compared to both stationary and non-stationary state-of-the-art TS baselines. We exploited synthetic environments (both randomly-generated and controlled) to test the MAB algorithms under different types of drift, that is, sudden/abrupt, incremental, gradual and increasing/decreasing drift. Furthermore, we adapt four real-world active learning tasks to our framework—a prediction task on crimes in the city of Baltimore, a classification task on insects species, a recommendation task on local web-news, and a time-series analysis on microbial organisms in the tropical air ecosystem. The f-dsw TS approach emerges as the best performing MAB algorithm. At least one of the versions of f-dsw TS performs better than the baselines in synthetic environments, proving the robustness of f-dsw TS under different concept drift types. Moreover, the pessimistic version () results as the most effective in all real-world tasks. 相似文献
In this work, we report that 2-cyano-2-phenylpropanoic acid and its p-Cl, p-CH3 and p-OCH3 derivatives can be used as chemical fuels to control the geometry of the calix[4]arene scaffold in its cone conformation. It is shown that, under the action of the fuel, the cone calix[4]arene platform assumes a “locked” shape with two opposite aromatic rings strongly convergent and the other two strongly divergent (“pinched cone” conformation). Only when the fuel is exhausted, the cone calix[4]arene scaffold returns to its resting, “unlocked” shape. Remarkably, the duration of the “locked” state can be controlled at will by varying the fuel structure or amount. A kinetic study of the process shows that the consume of the fuel is catalyzed by the “unlocked” calixarene that behaves as an autocatalyst for its own production. A mechanism is proposed for the reaction of fuel consumption. 相似文献
Myotonic dystrophy type 1 (DM1) is a multisystemic disease involving multiple organ systems including central nervous system (CNS) and muscles. Few studies have focused on the central motor system in DM1, pointing to a subclinical abnormality in the CNS. The aim of our study was to investigate patterns of cerebral activation in DM1 during a motor task using functional MRI (fMRI). Fifteen DM1 patients, aged 20 to 59 years, and 15 controls of comparable age were scanned during a self-paced sequential finger-to-thumb opposition task of their dominant right hand. Functional MRI images were analyzed using SPM99. Patients underwent clinical and genetic assessment; all subjects underwent a conventional MR study. Myotonic dystrophy type 1 patients showed greater activation than controls in bilateral sensorimotor areas and inferior parietal lobules, basal ganglia and thalami, in the ipsilateral premotor area, insula and supplementary motor area (corrected P<.05). Analysis of the interaction between disease and age showed that correlation with age was significantly greater in patients than in controls in bilateral sensorimotor areas and in contralateral parietal areas. Other clinical and MR characteristics did not correlate with fMRI. Functional changes in DM1 may represent compensatory mechanisms such as reorganization and redistribution of functional networks to compensate for ultrastructural and neurochemical changes occurring as part of the accelerated aging process. 相似文献
