Separate methods for the analyses of soluble carbohydrates in different plants and simmondsins in jojoba seed meal are described. A reliable gas chromatographic procedure for the simultaneous quantification of D-pinitol, myo-inositoL sucrose, 5-alpha-D-galactopyranosyl-D-pinitol. 2-alpha-D-galactopyranosyl-D-pinitol, simmondsin, 4-demethylsimmondsin, 5-demethylsimmondsin and 4,5-didemethylsimmondsin as trimethylsilyl derivatives in jojoba seed meal has been developed. The study of different extraction mixtures allowed for the quantitative recovery of the 9 analytes by a mixture of methanol-water (80:20, v/v) in the concentration range between 0.1 and 4%. Comparison of the separation parameters on three different capillary stationary phases with MS detection allowed for the choice of the optimal gas chromatographic conditions for baseline separation of the analytes.  相似文献

Applications of solid-phase microextraction (SPME) in the measurement of very hydrophobic organic compounds (VHOCs) are limited, partly due to the difficulty of calibrating SPME fibers for VHOCs. This study used a static SPME strategy with a large sample volume (1.6 L) and a five-point calibration procedure to determine the distribution coefficients for a large suite of polychlorinated biphenyls (PCBs) and chlorinated pesticides between a polydimethylsiloxane (PDMS) phase (100 microm thickness) coated on a glass fiber and seawater. An extraction time of 12 days was deemed adequate for equilibrium calibration from kinetic experiments. Two groups of randomly selected fibers divided into three batches (up to nine fibers in each batch) were processed separately with two gas chromatography-mass spectrometry (GC-MS) systems. Matrix effects arising from losses of the analytes to glass container walls and stirring bars were corrected. Relative standard deviations within the same batch were generally smaller than those for the entire group. Furthermore, KfVf (Kf and Vf are the distribution coefficient of an analyte between the polymer-coated fiber and aqueous phase and the fiber volume, respectively) values determined with two GC-MS systems were statistically different. These results indicate the calibrated KfVf values were less affected by the random selection of SPME fibers than by other experimental conditions, and therefore average KfVf values may be used for the same type of commercially available SPME fibers. The relative accuracy of our calibration method was similar to that of a previous study [P. Mayer. W.H.J. Vaes, J.L.M. Hermens, Anal. Chem. 72 (2000) 459] employing different coating thickness and calibration procedure. The present study also obtained a bell-shaped relationship between log Kf and log Kow (octanol-water partition coefficient) for PCB congeners with the maximum log Kf corresponding to log Kow approximately 6.5. This bell-shaped relationship was attributed mainly to steric effects arising from the interplay between the PDMS thickness and molecular sizes of the target analytes.  相似文献

We present a robust protocol based on iterations of free energy perturbation (FEP) calculations, chemical synthesis, biophysical mapping and X‐ray crystallography to reveal the binding mode of an antagonist series to the A2A adenosine receptor (AR). Eight A2AAR binding site mutations from biophysical mapping experiments were initially analyzed with sidechain FEP simulations, performed on alternate binding modes. The results distinctively supported one binding mode, which was subsequently used to design new chromone derivatives. Their affinities for the A2AAR were experimentally determined and investigated through a cycle of ligand‐FEP calculations, validating the binding orientation of the different chemical substituents proposed. Subsequent X‐ray crystallography of the A2AAR with a low and a high affinity chromone derivative confirmed the predicted binding orientation. The new molecules and structures here reported were driven by free energy calculations, and provide new insights on antagonist binding to the A2AAR, an emerging target in immuno‐oncology.  相似文献

The corrosion of metals, i.e., the initiation and acceleration of the surface deterioration of metals through an electrochemical reaction with the surrounding intrusive environment, is a global concern because of the economic and environmental impacts. Corrosion inhibitors are considered the most practical choice among the available corrosion protection techniques due to their effectiveness in terms of functionality and cost. The use of traditional and toxic corrosion inhibitors has led to environmental issues, arousing the need for green counterparts that are environmentally friendly, easily accessible, biodegradable, and cost-effective. In this review, the utilization of green corrosion inhibitors purely acquired from renewable sources is explored, with an in-depth focus on the recent advancements in the use of fruit and vegetable extracts as green corrosion inhibitors. In particular, fruits and vegetables are natural sources of various phytochemicals that exhibit key potential in corrosion inhibition. To shed light on the true potential of such extracts in the protection of steel in acidic environments, the experimental techniques involved in corrosion inhibition and the mechanism of corrosion inhibition are discussed in detail. The study highlights the potential of fruit and vegetable extracts as non-toxic, economical, and effective corrosion inhibitors in the pursuit of green chemistry. In addition to discussing and outlining the current status and opportunities for employing fruit and vegetable extracts as corrosion inhibitors, the current review outlines the challenges involved in the utilization of such extracts in corrosion inhibition.  相似文献

The computation of the reachable set of states of a given dynamic system is an important step to verify its safety during operation. There are different methods of computing reachable sets, namely interval integration, capture basin, methods involving the minimum time to reach function, and level set methods. This work deals with interval integration to compute subpavings to over or under approximate reachable sets of low dimensional systems. The main advantage of this method is that, compared to guaranteed integration, it allows to control the amount of over-estimation at the cost of increased computational effort. An algorithm to over and under estimate sets through subpavings, which potentially reduces the computational load when the test function or the contractor is computationally heavy, is implemented and tested. This algorithm is used to compute inner and outer approximations of reachable sets. The test function and the contractors used in this work to obtain the subpavings involve guaranteed integration, provided either by the Euler method or by another guaranteed integration method. The methods developed were applied to compute inner and outer approximations of reachable sets for the double integrator example. From the results it was observed that using contractors instead of test functions yields much tighter results. It was also confirmed that for a given minimum box size there is an optimum time step such that with a greater or smaller time step worse results are obtained.  相似文献

In closed, cerium-catalyzed glyoxylic acid Belousov-Zhabotinsky systems we found a new dynamic behavior, viz. the observation of two oscillatory regimes separated by a nonoscillatory region. Mechanisms for this intermediate nonoscillatory domain are discussed.

First evidence for the existence of free trifluoromethyl anion CF3 has been obtained. The 3D‐caged potassium cation in [K(crypt‐222)]+ is inaccessible to CF3, thus rendering it uncoordinated (“naked”). Ionic [K(crypt‐222)]+ CF3 has been characterized by single‐crystal X‐ray diffraction, solution NMR spectroscopy, DFT calculations, and reactivity toward electrophiles.  相似文献

The inhibition of the corrosion of mild steel in HCl by some cyclopentadiene‐1,3‐diene derivatives, namely, 1‐(thiophen‐2‐yl)ethanone (2APT), 1‐(1H‐pyrrol‐2‐yl)ethanone (2AP), and (E)‐2‐(1‐hydrazonoethyl)‐1H‐pyrrole (2APH)), was studied experimentally using weight loss and hydrogen gas evolution measurements. The theoretical aspect was studied using the density functional theory and quantitative structure activity relation (QSAR) methods. The results obtained indicated that the studied compounds are good inhibitors for the corrosion of mild steel in HCl. The adsorption of the inhibitor on mild steel surface was found to be spontaneous, exothermic, and obeyed the Langmuir adsorption isotherm model. A good correlation was found between experimental inhibition efficiencies and some calculated quantum chemical parameters and also with the theoretical inhibition efficiencies obtained from QSAR modeling. © 2010 Wiley Periodicals, Inc. ProteinChip surface‐enhanced laser desorption/ionization technology and magnetic beads‐based ClinProt system are commonly used for semi‐quantitative profiling of plasma proteome in biomarker discovery. Unfortunately, the proteins/peptides detected by MS are non‐recoverable. To obtain the protein identity of a MS peak, additional time‐consuming and material‐consuming purification steps have to be done. In this study, we developed a magnetic beads‐based proteomic fingerprinting method that allowed semi‐quantitative proteomic profiling and micropreparative purification of the profiled proteins in parallel. The use of different chromatographic magnetic beads allowed us to obtain different proteomic profiles, which were comparable to those obtained by the ProteinChip surface‐enhanced laser desorption/ionization technology. Our assays were semi‐quantitative. The normalized peak intensity was proportional to concentration measured by immunoassay. Both intra‐assay and inter‐assay coefficients of variation of the normalized peak intensities were in the range of 4–30%. Our method only required 2 μL of serum or plasma for generating enough proteins for semi‐quantitative profiling by MALDI‐TOF‐MS as well as for gel electrophoresis and subsequent protein identification. The protein peaks and corresponding gel spots could be easily matched by comparing their intensities and masses. Because of its high efficiency and reproducibility, our method has great potentials in clinical research, especially in biomarker discovery.  相似文献
