Abstract— Relative potency of 10 fluorescein derivatives as sensitizers of delayed photohemolysis of human erythrocytes has been assessed. Dilute suspensions of washed cells were illuminated in the presence of sensitizer at different concentrations for 1 h and analyzed for percent hemolysis following 23 h of dark incubation. Plots of percent hemolysis versus concentration showed a steep dependence on concentration for all sensitizers. Additional measurements of octanol/water partition coefficients, photon absorption in octanol and in saline and photobleach rates were made. After correction for absorption cross section, the effectiveness values ranged over more than three orders of magnitude with fluorescein being the least potent and rose bengal the most. A reasonable prediction of potency is obtained by taking the product of partition coefficient, relative absorption in octanol versus water and molecular weight of the substituents added to the fluorescein skeleton. The results suggest that the influence of halogen substitution on sensitizing potency is exerted by four factors; (1) distribution of sensitizer into a low polarity region of the cell membrane, (2) absorption efficiency in a low polarity region, (3) triplet quantum yield, and (4) photobleach rate.  相似文献

A supramolecular metal-ligand assembly encapsulates a variety of cationic half-sandwich ruthenium complexes. Due to the chirality of both host and guest, chiral recognition is observed with diastereomeric excesses up to 70%. The lack of effective treatment for neurological disorders has encouraged the search for novel therapeutic strategies. Remarkably, neuroinflammation provoked by the activated microglia is emerging as an important therapeutic target for neurological dysfunction in the central nervous system. In the pathological context, the hyperactivation of microglia leads to neuroinflammation through the release of neurotoxic molecules, such as reactive oxygen species, proteinases, proinflammatory cytokines and chemokines. Cannabidiol (CBD) is a major pharmacologically active phytocannabinoids derived from Cannabis sativa L. CBD has promising therapeutic effects based on mounting clinical and preclinical studies of neurological disorders, such as epilepsy, multiple sclerosis, ischemic brain injuries, neuropathic pain, schizophrenia and Alzheimer’s disease. A number of preclinical studies suggested that CBD exhibited potent inhibitory effects of neurotoxic molecules and inflammatory modulators, highlighting its remarkable therapeutic potential for the treatment of numerous neurological disorders. However, the molecular mechanisms of action underpinning CBD’s effects on neuroinflammation appear to be complex and are poorly understood. This review summarises the anti-neuroinflammatory activities of CBD against various neurological disorders with a particular focus on their main molecular mechanisms of action, which were related to the downregulation of NADPH oxidase-mediated ROS, TLR4-NFκB and IFN-β-JAK-STAT pathways. We also illustrate the pharmacological action of CBD’s derivatives focusing on their anti-neuroinflammatory and neuroprotective effects for neurological disorders. We included the studies that demonstrated synergistic enhanced anti-neuroinflammatory activity using CBD and other biomolecules. The studies that are summarised in the review shed light on the development of CBD, including its derivatives and combination preparations as novel therapeutic options for the prevention and/or treatment of neurological disorders where neuroinflammation plays an important role in the pathological components.  相似文献

A novel class of semifluorinated perfluorocyclohexenyl (PFCH) aryl ether homo/copolymers was successfully synthesized with high yield through the step‐growth polymerization of commercially available bisphenols and decafluorocyclohexene in the presence of a triethylamine base. The synthesized polymers exhibit variable thermal properties depending on the functional spacer group (R). PFCH aryl ether copolymers with random and alternating architectures were also prepared from versatile bis‐perfluorocyclohexenyl aryl ether monomers. The PFCH polymers show high thermal stabilities with a 5% decomposition temperature ranging from 359 to 444 °C in air and nitrogen atmosphere. These semifluorinated PFCH aromatic ether polymers contain intact enchained PFCH olefin moieties, making further reactions such as crosslinking and application specific functionalization possible. © 2013 Wiley Periodicals, Inc.
