Metal-chelating affinity beads have attracted increasing interest in recent years for protein purification. In this study, iminodiacetic acid (IDA) was covalently attached to the poly(glycidyl methacrylate) [PGMA] beads (1.6 μm in diameter). Cu(2+) ions were chelated via IDA groups on PGMA beads for affinity binding of hemoglobin (Hb) from human blood hemolysate. The PGMA beads were characterized by scanning electron microscopy (SEM). The PGMA-Cu(2+) beads (628 μmol/g) were used in the Hb binding-elution studies. The effects of Hb concentration, pH and temperature on the binding efficiency of PGMA-Cu(2+) beads were performed in a batch system. Non-specific binding of Hb to PGMA beads in the absence of Cu(2+) ions was very low (0.39 mg/g). The maximum Hb binding was 130.3 mg/g. The equilibrium Hb binding increased with increasing temperature. The negative change in Gibbs free energy (ΔG°<0) indicated that the binding of Hb on the PGMA-Cu(2+) beads was a thermodynamically favorable process. The ΔS and ΔH values were 102.2 J/mol K and -2.02 kJ/mol, respectively. Significant amount of the bound Hb (up to 95.8%) was eluted in the elution medium containing 1.0 M NaCl in 1 h. The binding followed Langmuir isotherm model with monolayer binding capacity of 80.3-135.7 mg/g. Consecutive binding-elution experiments showed that the PGMA-Cu(2+) beads can be reused almost without any loss in the Hb binding capacity. To test the efficiency of Hb depletion from blood hemolysate, eluted portion was analyzed by fast protein liquid chromatography. The depletion efficiency for Hb was above 97.5%. This study determined that the PGMA-Cu(2+) beads had a superior binding capacity for Hb compared to the other carriers within this study.  相似文献

A simple, rapid, sensitive and environmentally friendly separation and preconcentration procedure, based on the carrier element free coprecipitation (CEFC) of Cu(II) and Cd(II) ions by using an organic coprecipitant, 2-{[4-(4-fluorophenyl)-5-sulphanyl-4H-1,2,4-triazol-3-yl]methyl}-4-{[(4-fluorophenyl) methylene]amino}-5-(4-methylphenyl)-2,4-dihydro-3H-1,2,4-triazol-3-one (MEFMAT) was developed. The analyte ions were determined by flame atomic absorption spectrometric (FAAS) determinations. The optimum conditions for the coprecipitation process were investigated on several commonly tested experimental parameters such as pH of the solution, amount of MEFMAT, sample volume, standing time, centrifugation rate and time. The influences of some anions, cations and transition metals on the recoveries of analyte ions were also investigated, and no considerable interference was observed. The preconcentration factor was found to be 50. The detection limits for Cu(II) and Cd(II) ions based on the three times the standard deviation of the blanks (N:10) were found to be 1.49 and 0.45 μg L− 1, respectively. The relative standard deviations were found to be lower than 3.5% for both analyte ions. The method was validated by analyzing two certified reference materials (CRM-TMDW-500 Drinking Water and CRM-SA-C Sandy Soil C) and spike tests. The procedure was successfully applied to sea water and stream water as liquid samples and tobacco, hazelnut and black tea as solid samples.  相似文献

This paper considers a cyclic-service system with a class of limited service policies that consists of exhaustive limited, gated limited and general decrementing policies. Under these policies, the number of customers served consecutively during a server visit is limited by a vector of integers. The major results in this paper are derivations of expected amount of work left in the queues at the server departures for these three policies. Exact expressions of weighted sum of mean waiting times, known as pseudo-conservation laws, are subsequently obtained. The conservation laws for this class of policies contain unknown boundary probabilities. We estimate these probabilities using corresponding server vacation models. Numerical results presented for the exhaustive limited policy are noted to be very accurate compared with simulation results. Moreover, we have obtained analytical bounds for the weighted sums. Finally, we present a conservation law with mixed service policies, and mean waiting times for symmetric systems.This work was completed while the author was in the Ph.D. program at Rensselaer Polytechnic Institute.This work was partially supported by the Center for Advanced Technologies of the New York State.  相似文献

Linoleic acid attached chitosan beads [poly(LA-Ch)] (1.25 μm in diameter) are obtained by the formation of amide linkages between linoleic acid and chitosan. Poly(LA-Ch) beads are characterized by FTIR, TEM, and swelling studies. Poly(LA-Ch) beads are used for the purification of immunoglobulin-G (IgG) from human plasma in a batch system. The maximum IgG adsorption is observed at pH 7.0 for HEPES buffer. IgG adsorption onto the plain chitosan beads is found to be negligible. Adsorption values up to 136.7 mg/g from aqueous solutions are obtained by poly(LA-Ch) beads. IgG adsorption saw an increase as a result of increasing temperature. Higher amounts of IgG are adsorbed from human plasma (up to 390 mg/g) with a purity of 92%. The adsorption phenomena appeared to follow a typical Langmuir isotherm. It is observed that IgG could be repeatedly adsorbed and desorbed without significant loss when we take into account the adsorption amount. It is concluded that the poly(LA-Ch) beads allowed one-step purification of IgG from human plasma.  相似文献

Alkylphosphate self-assembled monolayers (SAMs) were prepared on Nb-doped SrTiO(3) (Nb-STO) conducting metal oxide substrates. Unlike thiols on gold, the alkylphosphate SAMs on Nb-STO exhibited an electrochemical stability over a wide voltage range from -2 to 2 V. Cyclic voltammetry showed that the SAM modification inhibited the electrochemical activity of the underlying conducting substrate with an efficiency dependent on the chain length. Impedance spectroscopy showed that SAM-modified Nb-STO substrates have a significantly higher resistance than bare substrates.  相似文献

In sensorineural hearing loss, damage to inner hair cells or the auditory nerve may result in dead regions in the cochlea, where the information transmission is disrupted. In cochlear implants, similar dead regions might appear if the spiral ganglia do not function. Shannon et al. [J. Assoc. Res. Otolaryngol. 3, 185-199 (2002)] simulated dead regions of varying size and location using a noiseband vocoder. Phoneme recognition by normal-hearing subjects was measured under two frequency-place mapping conditions: the frequency range corresponding to the dead region was (1) removed or (2) reassigned to bands adjacent to the dead region to simulate the off-frequency stimulation of neurons at the edge of a dead region. The present study extends the results of Shannon et al. by including a frequency transposition mapping condition, where the overall acoustic input frequency range was distributed over the entire remaining nondead region. The frequency transposed map provided more acoustic information when compared to the map with the frequency range corresponding to the dead region removed. However, speech perception did not improve for many simulated dead region conditions, possibly due to the spectral distortions in the frequency-place mapping.  相似文献

Adversarial examples are one of the most intriguing topics in modern deep learning. Imperceptible perturbations to the input can fool robust models. In relation to this problem, attack and defense methods are being developed almost on a daily basis. In parallel, efforts are being made to simply pointing out when an input image is an adversarial example. This can help prevent potential issues, as the failure cases are easily recognizable by humans. The proposal in this work is to study how chaos theory methods can help distinguish adversarial examples from regular images. Our work is based on the assumption that deep networks behave as chaotic systems, and adversarial examples are the main manifestation of it (in the sense that a slight input variation produces a totally different output). In our experiments, we show that the Lyapunov exponents (an established measure of chaoticity), which have been recently proposed for classification of adversarial examples, are not robust to image processing transformations that alter image entropy. Furthermore, we show that entropy can complement Lyapunov exponents in such a way that the discriminating power is significantly enhanced. The proposed method achieves 65% to 100% accuracy detecting adversarials with a wide range of attacks (for example: CW, PGD, Spatial, HopSkip) for the MNIST dataset, with similar results when entropy-changing image processing methods (such as Equalization, Speckle and Gaussian noise) are applied. This is also corroborated with two other datasets, Fashion-MNIST and CIFAR 19. These results indicate that classifiers can enhance their robustness against the adversarial phenomenon, being applied in a wide variety of conditions that potentially matches real world cases and also other threatening scenarios.  相似文献

This study yielded a mathematical expression to calculate the pressure gradient (ΔP/L)m of the flow of a spherical capsule train. An experimental investigation was carried out to determine pressure drops of two-phase mixture flow of spherical ice capsules and water inside the pipelines of cooling systems. Instead of ice capsules, spherical capsules made of polypropylene material whose density (870 kg/m3) is similar to that of ice were used in the experiments. Flow behavior of the spherical capsules, 0.08 m outer diameter, was observed in the measuring section inside plexiglass pipes, 0.1 m inner diameter (ID) and 6 m in length; pressure drops were measured on the 4 m section. The investigation was carried out in the 1.2 × 104 < Re < 1.5 × 105 range and under transport concentration (Ctr) by 5–30%. Dimensionless numbers of the physical event were found out by conducting a dimensional analysis, so that mixture density was expressed in terms of specific gravity and in situ concentration. After arriving at certain conclusions based on the relevant experimental findings and observations, empirical and mathematical models which can be used for calculation of the pressure gradient were developed. Comparison of the mathematical model with the experimental findings revealed that pressure drop values deviated by 2.7% on average for 2.5 × 104 < Re < 1.5 × 105.  相似文献
