Background  

Though Dnmt1 is considered the primary maintenance methyltransferase and Dnmt3a and Dnmt3b are considered de novo methyltransferases in mammals, these three enzymes may work together in maintaining as well as establishing DNA methylation patterns. It has been proposed that Dnmt1 may carry out de novo methylation at sites in the genome with transient single-stranded regions, such as replication origins, and then spread methylation from these nucleation sites in vivo, even though such activity has not been reported.  相似文献

The quenching by molecular oxygen of the fluorescence from a protoporphyrin IX adduct of horseradish peroxidase has been investigated using both intensity and time-resolved techniques. The bimolecular quenching rate constant determined for this process, as evaluated by the conventional Stern-Volmer analysis, was 2 x 10(8) M-1 s-1, among the lowest observed for protein systems. This result suggests that the heme binding site in horseradish peroxidase is relatively inaccessible to oxygen, which may account for the observation of room temperature phosphorescence in aerated solutions from enzymatically created triplet states.  相似文献

Recently, structure 8b has been assigned to the iridoid specionin. In the present letter the total synthesis of 8b is described. Comparison of spectral data reveals that the final product is not identical with natural specionin. Therefore the original structural assignment of specionin must be revised.  相似文献

The Bi2O3---V2O5 system was examined using Raman spectroscopy and solid state 51V wideline, magic-angle spinning (MAS), and nutation NMR spectroscopy. The methods are shown to be complementary in the identification of the various phases and in the characterization of their vanadium site symmetries. Most of the compositions examined (1:1 ≤ Bi:V ≤ 60:1) are multiphasic. Depending on the Bi:V ratio, the following phases have been identified: BiVO4, Bi4V2O11, a triclinic type-II phase, a cubic type-I phase, γ-Bi2O3 doped with V(V) (sillenite), and β-Bi2O3. Detailed spectroscopic characterization reveals that vanadium is tetrahedrally coordinated in all these compounds, and that the degree of symmetry increases with increasing Bi:V ratio. At the highest Bi:V ratios, the combined interpretation of the Raman and NMR data provides strong evidence for the presence of Bi5+O4 tetrahedra.  相似文献

The cloning and expression of autofluorescent proteins in living matter, combined with modern imaging techniques, have thoroughly changed the world of bioscience. In particular, such proteins are widely used as genetically encoded labels to track the movement of proteins as reporters of cellular signals and to study protein-protein interactions by fluorescence resonance energy transfer (FRET). Their optical properties, however, are complex and it is important to understand these for the correct interpretation of imaging data and for the design of new fluorescent mutants. In this Minireview we start with a short survey of the field and then focus on the photo- and thermally induced dynamics of green and red fluorescent proteins. In particular, we show how fluorescence line narrowing and high-resolution spectral hole burning at low temperatures can be used to unravel the photophysics and photochemistry and shed light on the intricate electronic structure of these proteins.  相似文献

Protected alpha-formyl amino acids, themselves available from the corresponding alpha-vinyl amino acids, are stereoselectively transformed into the (Z)-configured alpha-(2'-fluoro)vinyl amino acids via a three-step sequence. The route employs McCarthy's reagent, diethyl alpha-fluoro-alpha-(phenylsulfonyl)methyl phosphonate, and proceeds via the intermediate (E)-alpha-fluorovinyl sulfones and (E)-alpha-fluorovinylstannanes. Several improved approaches for the preparation of tetra-O-benzyl-α-D-glucopyranosyl bromide and tetra-O-pivaloyl-α-D-glucopyranosyl bromide are discussed. The importance of these compounds, which are useful glycosyl donors, was demonstrated by successful preparation of cholesteryl glucopyranosides in an almost neutral medium without the formation of orthoesters. In addition, accurate 1H and 13C NMR resonance assignments of the synthesized cholesteryl glycosides were performed by 2D NMR spectroscopy.  相似文献
