ε-Poly-l-lysine (ε-PL) is a natural preservative for food processing industry. A thermo-responsive polymer, attached with Cu2+ or Ni2+, was prepared for metal-chelate affinity precipitation for purification of ε-PL. The low critical solution temperatures (LCSTs) of these polymers were close to the room temperature (31.0–35.0 °C). The optimal adsorption conditions were as follows: pH 4.0, 0 mol/L NaCl, ligand density 75.00 μmol/g, and 120 min. The ligand Cu2+ showed a stronger affinity interaction with ε-PL and the highest adsorption amount reached 251.93 mg/g polymer. The elution recovery of ε-PL could be 98.42% with 0.50 mol/L imidazole (pH = 8.0) as the eluent. The method could purify ε-PL from fermentation broth and the final product was proved as electrophoretic pure by SDS-PAGE. Moreover, these affinity polymers could be recycled after the purification of ε-PL and the recoveries were above 95.00%.

Tetrahydrobiopterin (BH4) is an essential cofactor for aromatic acid hydroxylases and nitric oxide synthase. Sepiapterin reductase (SPR) catalyzes the final steps of BH4 biosynthesis. Studies on SPR from several insects and other organisms have been reported. However, thus far, enzyme activity of SPR in Musca domestica is kept unknown. In this study, 186 differentially expressed genes including SPR gene from Musca domestica (MDSPR) were screened in subtractive cDNA library. The MDSPR gene was cloned, and the recombinant MDSPI16 protein was expressed as a 51-kDa protein in soluble form. The MDSPR exhibited strong activity to the substrate sepiapterin (SP). The values of Vmax and Km of the MDSPR for SP were 6.83 μM/min and 23.48 μM, and the optimum temperature and pH of MDSPR were 50 °C and 4.0, respectively. This study provides new hypotheses and methods for the production of BH4 using insect-derived SPR.  相似文献

Poly(ethylene glycol) (PEG 4000) and bovine serum albumin (BSA) were investigated with the purpose of evaluating their influence on enzymatic hydrolysis of sugarcane bagasse. Effects of these supplements were assayed for different enzymatic cocktails (Trichoderma harzianum and Penicillium funiculosum) that acted on lignocellulosic material submitted to different pretreatment methods with varying solid (25 and 100 g/L) and protein (7.5 and 20 mg/g cellulose) loadings. The highest levels of glucose release were achieved using partially delignified cellulignin as substrate, along with the T. harzianum cocktail: increases of 14 and 18 % for 25 g/L solid loadings and of 33 and 43 % for 100 g/L solid loadings were reached for BSA and PEG supplementation, respectively. Addition of these supplements could maintain hydrolysis yield even for higher solid loadings, but for higher enzymatic cocktail protein loadings, increases in glucose release were not observed. Results indicate that synergism might occur among these additives and cellulase and xylanases. The use of these supplements, besides depending on factors such as pretreatment method of sugarcane bagasse, enzymatic cocktails composition, and solid and protein loadings, may not always lead to positive effects on the hydrolysis of lignocellulosic material, making it necessary further statistical studies, according to process conditions.  相似文献

NMR and X-ray crystallography are the two most widely used methods for determining protein structures. Our previous study examining NMR versus X-Ray sources of protein conformations showed improved performance with NMR structures when used in our Multiple Protein Structures (MPS) method for receptor-based pharmacophores (Damm, Carlson, J Am Chem Soc 129:8225–8235, 2007). However, that work was based on a single test case, HIV-1 protease, because of the rich data available for that system. New data for more systems are available now, which calls for further examination of the effect of different sources of protein conformations. The MPS technique was applied to Growth factor receptor bound protein 2 (Grb2), Src SH2 homology domain (Src-SH2), FK506-binding protein 1A (FKBP12), and Peroxisome proliferator-activated receptor-γ (PPAR-γ). Pharmacophore models from both crystal and NMR ensembles were able to discriminate between high-affinity, low-affinity, and decoy molecules. As we found in our original study, NMR models showed optimal performance when all elements were used. The crystal models had more pharmacophore elements compared to their NMR counterparts. The crystal-based models exhibited optimum performance only when pharmacophore elements were dropped. This supports our assertion that the higher flexibility in NMR ensembles helps focus the models on the most essential interactions with the protein. Our studies suggest that the “extra” pharmacophore elements seen at the periphery in X-ray models arise as a result of decreased protein flexibility and make very little contribution to model performance.  相似文献

Several novel fused heterocyclic systems which have rings containing N/O or N/S have been synthesized through a facile one-pot method by the treatment of substituted 2-(benzo[d]thiazol-2-yl)phenol and cyanogen bromide in THF and NEt3. This one-pot method contains tough ring-opening of thiazole, special intermolecular rearrangement and ring-closing reactions. The newly synthesized compounds were characterized by HR-MS, 1H-NMR, 13C-NMR spectral data and DFT calculation analysis. 2a was also determined by X-ray crystallographic analysis.  相似文献

The aim of this work was to investigate the efficiency of various MWCNTs as SPE materials for the preconcentration of chlorophenols. The COOH-functionalized MWCNTs and MWCNTs were used as SPE sorbents. To evaluate the capability of MWCNTs for the preconcentration of chlorophenols from water samples, 2,4-chlorophenol, 4-chlorophenol, 2,4,6-chlorophenol, 2,6-chlorophenol, 3,4-chlorophenol, and 2-chlorophenol were used as model compounds. Chlorophenols were extracted with acetone, methanol, ethanol, and dichloromethane, and determined by gas chromatography–mass spectrometry. COOH-functionalized MWCNTs <8 nm were found to be the best sorbent for the tested chlorophenols. For COOH-functionalized MWCNTs <8 nm, the recovery rates for all chlorophenols were higher than 50% when acetone or ethanol was used as eluents. In the case of dichloromethane elution, recovery rates for chlorophenols were from 62.0% for 2,6-DCP to 116.8% for 2,4-DCP; only for 2,4,6-TCP was the recovery rate 30.6%. Similar percentage recoveries were achieved with methanol as the eluent.  相似文献

A practical and step-economic route to Favipiravir, an antiviral drug, was developed. Favipiravir was synthesized in only six steps from 3-aminopyrazine-2-carboxylic acid with an overall yield of about 22.3%. Key intermediates 3 and 6 were obtained in excellent purity via recrystallization from optimized solvents, which was beneficial to large-scale production. In the key synthetic reaction, 3,6-dichloropyrazine-2-carbonitrile (6) was reacted sequentially, in one pot, with KF and 30% H2O2 to give (after crystallization from 95% EtOH) favipiravir as colorless crystals, with a 60% yield for this final step of the synthesis.  相似文献

Based on the Keggin-type polyoxometalate and quaternary phosphonium salt, the (MePh3P) n -POMKeggin compounds were synthesized via one-step mechanochemical process at room temperature. It showed the advantages of convenient operation, lower cost, less pollution, and mass production. Wonderfully, the morphology of compounds presented a strong dependence on the number of crystal water in the source heteropoly acids. A hypothesis of ‘semi-solid nonlocalized waters’ was brought to discuss the formation mechanism of polyhedrons. Antibacterial experiments indicate that the samples have good antibacterial activities, which resulted from the increase of the positive electrical charge of the phosphonium cation caused by the polarization by POM groups. Additionally, a formula of inhibitory zone per unit surface area was designed to more precisely evaluate the antibacterial activity of the materials. The calculation result indicated that the polyhedral particles give a higher surface antibacterial activity than the grain powder. This work developed an alternative synthesis method for composites from a new perspective, and will promote the research of new type of antibacterial agent.  相似文献
