The aim of this study was to investigate functional increments of ion exchange type ligands, which may improve the performance of mixed-modal ligands for antibody capture out of feed solutions with pH above 6.0 and containing sodium chloride concentrations of 150 mM and higher. For this purpose several functional groups such as sulfonyl, sulfanyl, amide, methoxy, short alkyl and aromatic moieties were tested in combination with a strong sulfonic acid and/or a weak carboxylic acid group. Therefore a series of ligands were synthesized and subsequently coupled onto epoxide activated Fractogel® EMD. In the first instance, all materials were tested by static binding capacity measurements (SBC) under test conditions, comprising a wide variety of different sodium chloride concentrations and differing pH values ranging from 4.5 to 7.5. From these preliminary experiment it was found that especially the aromatic groups improved the binding of human immunoglobulin G (h-IgG) under isotonic conditions, while other increments, e.g. thiophilic or amide groups, were not able to increase the capacity significantly. Taking the SBC results into account, the most promising materials were investigated under dynamic binding conditions (DBC) with a reduced selection of test conditions (pH 5.5, 6.5 and 7.4 at 75 and 150 mM NaCl). N-benzoyl-homocysteine (material J) and 3,5-dimethoxybenzoyl-homocysteine (material K) showed 100% DBCs of 37 mg/mL and 32 mg/mL in the presence of 75 mM NaCl and pH 6.5. Material L carrying mercaptobenzoic acid as a ligand and tested with the same solution provided a 100% DBC of 68 mg/mL. The influence of Pluronic F68 in a mock feed solution as well as in cell culture supernatant was investigated with the best performing bio-affinity type adsorbent, material L. Zusammenfassung Die Untersuchungen wurden an diffundierten GaAs-Dioden mit abruptempn-Übergang durchgeführt. Bei tiefer Temperatur zeigte die von den Dioden ausgesandte Strahlung TM-Polarisation. Der PolarisationsgradP nahm indessen oberhalb von etwa 190°K stark ab. Oberhalb dieser Temperatur wurde ausserdem eine stärkere Zunahme des Schwellenstromes festgestellt, ferner trat eine Verzögerung der Laserstrahlung gegenüber dem Stromimpuls auf. Diese Beobachtungen dürften auf eine Änderung der Streumechanismen der Ladungsträger infolge der Erscheinung von Traps sowie auf eine grössere Wahrscheinlichkeit für strahlungslose Übergänge zurückzuführen sein. 相似文献

The correlation functions of the intensity fluctuations are calculated by use of a linearization procedure for the equations of motion which include the coupling of the counter-rotating travelling waves of the ring laser in the cases of Doppler broadening, of homogeneous broadening with a self-induced population-inversion grating, and in intermediate cases. Depending on the strength of the mode competition various stable stationary solutions exist for the amplitudes. The transition between these stable states shows phenomena closely resembling phase transitions such as critical fluctuations, critical slowing down, etc. In particular, when the system passes from the state where both modes are above threshold to the state where one mode is below threshold, the negative cross-correlation of the fluctuations also becomes critical. In the case of the transient behaviour of the ring laser in the unstable region the time development of the amplitudes and the correlation functions are described in a short-time approximation. 相似文献

Poly(ether ether ketone) (PEEK) was irradiated with 4?MeV O+ and 5 and 10?MeV Au+ ions to the fluences from 1012 to 1014?cm?2 and then treated in 5 M/l water solution of LiCl for one month at room temperature. After drying and removal of LiCl surface contamination, the depth distribution of LiCl embeded in PEEK was measured by the neutron depth profilig method (NDP) sensitive to 6Li isotope. Embeded LiCl is believed to map distribution of water diffusing into PEEK interior. The results show that the PEEK irradiated to the fluences above 1.1013cm?2 is prone to water penetration to the depths of few microns. Model studies of prebiotic chemistry have revealed compelling routes for the formation of the building blocks of proteins and RNA, but not DNA. Today, deoxynucleotides required for the construction of DNA are produced by reduction of nucleotides catalysed by ribonucleotide reductases, which are radical enzymes. This study considers potential non‐enzymatic routes via intermediate radicals for the ancient formation of deoxynucleotides. In this context, several mechanisms for ribonucleotide reduction, in a putative H2S/HS. environment, are characterized using computational chemistry. A bio‐inspired mechanistic cycle involving a keto intermediate and HSSH production is found to be potentially viable. An alternative pathway, proceeding through an enol intermediate is found to exhibit similar energetic requirements. Non‐cyclical pathways, in which HSS. is generated in the final step instead of HS., show a markedly increased thermodynamic driving force (ca. 70 kJ mol?1) and thus warrant serious consideration in the context of the prebiotic ribonucleotide reduction. 相似文献

A simple one-pot synthesis of new ferrocene-containing 1,3-thiazinan-2-imines from 3-arylamino-1-ferrocenylpropan-1-ols and phenyl isothiocyanate has been developed. The key intermediate β-hydroxy thioureas were generated in situ using ultrasound irradiation and subsequent cyclization was achieved by the addition of acetic acid. The scope of the reaction towards various 3-arylamino-1-ferrocenylpropan-1-ols has been explored and the corresponding 3-aryl-6-ferrocenyl-N-phenyl-1,3-thiazinan-2-imines were obtained in moderate to high yields (52–90%). 相似文献
