Antibodies are commonly used as recognition elements in immunoassays because of their high specificity and affinity, and have seen extensive use in competitive assays for the detection of small molecules. However, these complex molecules require production either in animals or by mammalian cell cultures, and are not easily tailored through genetic manipulation. Single chain antibodies (scFv), recombinantly expressed molecules consisting of only the antibody's binding region joined via a linking peptide, can provide an alternative to intact antibodies. We describe the characterization of a new monoclonal antibody (mAb), 2G5B5, able to detect the small molecule explosive 2,4,6-trinitrotoluene (TNT) and the scFv derived from its variable regions. The mAb and scFv were tested by surface plasmon resonance to determine their affinity for an immobilized TNT surrogate; dissociation constants were determined to be 1.5 × 10−13 M and 4.8 × 10−10 M respectively. Circular dichroism was used to determine their melting temperatures. The mAb is more stable melting at ∼75 °C while the scFv melts at ∼65 °C. The recognition elements were incorporated into a competitive assay format using a bead-based multiplexing platform to examine their sensitivity and specificity. The scFv was able to detect TNT ∼10-fold more sensitively than the mAb in this assay format, allowing detection of TNT concentrations down to at least 1 μg L−1. The 2G5B gave similar detection limits to a commercial anti-TNT mAb, but was less specific, recognizing 1,3,5-trinitrobenzene (TNB) equally well as TNT. 相似文献

Abstract Task‐specific ionic liquids possessing two Brönsted acid sites with –COOH, HSO? 4, or H2PO? 4 groups have been designed, synthesized, and characterized. Under mild conditions and without any additional organic solvent, the esterification of isopropanol by chloroacetic acid could be carried out in these new task‐specific ionic liquids. They were endowed with unparalleled superiority to natural nucleobase analogues 2–6 to gelate aromatic solvents due to their excellent self-assembly properties. The J-AT nucleoside-based organogelators showed a specific self-complementary base pair recognition characteristic. The three-component reactions of 4-dimethylamino- or 4-methoxypyridine with acetylenedicarboxylates and arylidene cyanoacetates showed very interesting molecular diversity. The cage‐like complex, Ag4L4(NO3)4 ( 1 ) [L = 1, 4‐bis(pyridine‐2‐ylmethoxy)benzene] was synthesized by the reaction of the flexible bidentate ligand and silver nitrate. It was characterized by elemental analysis, IR spectroscopy, TG, and single‐crystal X‐ray analysis. Complex 1 is reported as the first cage‐like cluster constructed by four nitrate anions bridging two [2+2] macrocycles. Phase equilibria of the quaternary NaCl–NaBr–Na2SO4–H2O system at 323 K were studied by the isothermal dissolution equilibrium method. The solubilities of salts and densities of saturated solutions were determined. Solid solutions [Na(Cl, Br)] were found in the experiments. The phase diagram of the quaternary system has no invariant point, but has one univariant curve at the boundary of Na(Cl, Br) and Na2SO4 crystallization fields. The experimental results show that an increase of the NaBr concentration is accompanied by an obvious increase of the solution density and the decrease of the solubilities of NaCl and Na2SO4. 相似文献

An improved Stains‐All (ISA) staining method for phosphoproteins in SDS‐PAGE was described. Down to 0.5–1 ng phosphoproteins (α‐casein, β‐casein, or phosvitin) can be successfully selectively detected by ISA stain, which is approximately 120‐fold higher than that of original Stains‐All stain, but is similar to that of commonly used Pro‐Q Diamond stain. Furthermore, unlike the original Stains‐All protocol that was time consuming and light unstable, ISA stain could be completed within 60 min without resorting to protect the gels from light during the whole staining procedure. According to the results, it is concluded that ISA stain is a rapid, sensitive, specific, and economic staining method for a broad application to the research of phosphoproteins. 相似文献

Adsorption processes on a PMMA chip linked with CZE separations of a group of 13 aliphatic and aromatic mono‐ and di‐amines were studied. Due to the lack of chromophores within aliphatic amines, contact conductivity detection implemented directly onto the chip was used for monitoring of cationic CZE separations. To prevent an adsorption of studied amines to the chip channels, the surface of PMMA chip was modified by dynamic coating. Different surface modifiers, such as aliphatic oligoamines (diethylenetriamine and triethylenetetramine), were added to the BGE solutions filling the chip channels. The effect of various concentrations of surface modifiers on peak profiles and separation parameters of amines was monitored. Of these, mainly, aliphatic di‐amines and aromatic mono‐amines adversely affected the CZE resolution of a whole group of analytes by their strong adsorption to the chip channels. A propionate BGE with pH 3.2 containing 100 μM triethylenetetramine and 25 mM 18‐crown‐6‐ether was found suitable for CZE resolution of 12 from a total of 13 amines studied. Simple dynamic modification of the surface of PMMA chip enabled fast (analysis time lasted 9 min), sensitive (sub‐μM LODs reached) and reproducible (1–3% RSD of the peak areas) CZE analysis of the aliphatic and aromatic amines. 相似文献

The effects of ethanol on the thickness and ionization of adsorbed polymer layer on pigment were investigated. The results showed that the thickness of adsorbed polymer layer decreased with the increase of the ethanol concentration, and then the pigment particles aggregated when the concentration of ethanol is higher than 40%. The zeta potentials became more negative with the increasing of the ethanol concentration, and then changed reversely when the ethanol concentration was higher than 16%. The thickness and structure of adsorbed polymer layer on the particle determined the stability of pigment dispersion. 相似文献

The utilization of solid particles in aqueous foam has a great potential in improving fire fighting efficiency. In this study, aqueous foam supported by micro fly-ash (FA) was prepared and its stability in a specific type of oil was characterized. Firstly, different amount of FA was added to study the influence of FA concentration on foamability. It showed that within a specific extent, foam expansion ratio increased with the increasing of FA concentration. And compared with conventional foams, oil resistance of FA stabilized foams, which was investigated by analyzing drainage rate and evolution process with a self-made apparatus, was remarkably improved when FA concentration exceed 4.8wt.%. Secondly, SiO2 and Al2O3 particles with different median sizes were used to study the effect of particle size on stability. However, the smaller hydrophilic particles didn’t behave better as expected. Moreover, the foam stability in three hydrocarbons was evaluated in the same way. The results indicated that the short chain hydrocarbons had much stronger detrimental effect to both two-phase foam and three-phase foam. But overall, the three-phase foam stabilized by FA exhibited much better oil resistance, so it can be used as a promising material for pool fire extinguishing and prevention.GRAPHICAL ABSTRACT 相似文献
