The Tn antigen (GalNAc-α-1-O-Thr/Ser) is a well-known tumor-associated carbohydrate determinant. The use of glycopeptides that incorporate this structure has become a significant and promising niche of research owing to their potential use as anticancer vaccines. Herein, the conformational preferences of a glycopeptide with an unnatural Tn antigen, characterized by a threonine decorated with an sp2-iminosugar-type α-GalNAc mimic, have been studied both in solution, by combining NMR spectroscopy and molecular dynamics simulations, and in the solid state bound to an anti-mucin-1 (MUC1) antibody, by X-ray crystallography. The Tn surrogate can mimic the main conformer sampled by the natural antigen in solution and exhibits high affinity towards anti-MUC1 antibodies. Encouraged by these data, a cancer vaccine candidate based on this unnatural glycopeptide and conjugated to the carrier protein Keyhole Limpet Hemocyanin (KLH) has been prepared and tested in mice. Significantly, the experiments in vivo have proved that this vaccine elicits higher levels of specific anti-MUC1 IgG antibodies than the analog that bears the natural Tn antigen and that the elicited antibodies recognize human breast cancer cells with high selectivity. Altogether, we compile evidence to confirm that the presentation of the antigen, both in solution and in the bound state, plays a critical role in the efficacy of the designed cancer vaccines. NMR-based binding and functional screening performed with FAXS (fluorine chemical shift anisotropy and exchange for screening) and 3-FABS (three fluorine atoms for biochemical screening) represents a potential alternative approach to high-throughput screening for the identification of novel potential drug candidates. The major limitation of this method in its current status is its intrinsic low sensitivity that limits the number of tested compounds. One approach for overcoming this problem is the use of a cryogenically cooled (19)F probe that reduces the thermal noise in the receiver circuitry. Sensitivity improvement in the two screening techniques achieved with the novel cryogenic (19)F probe technology permits an increased throughput, detection of weaker binders and inhibitors (relevant in a fragment-based lead discovery program), detection of slow binders, and reduction in protein and substrate consumption. These aspects are analyzed with theoretical simulations and experimental quantitative performance evaluation. Application of 3-FABS combined with the cryogenic (19)F probe technology to rapid screening at very low enzyme concentrations and the current detection limits reached with this approach are also presented. 相似文献

High-throughput ligand-based proton NMR screening performed in the presence of a spy molecule and a control molecule is a valuable tool for identifying drug leads. A limitation of the technique is represented by the severe overlap encountered in the screening of large chemical mixtures. An approach for overcoming this overlap problem is the use of multi-selective R(1) filtered and COSY or TOCSY experiments. Application of this methodology to compounds binding to the Sudlow site I of human serum albumin is presented. The screening is performed by simply monitoring the intensity of two signals. The precise measurement of the relative intensity of the two resonances permits determination of the binding constant of the NMR-hit. For a simple competition binding mechanism, the rapidly-derived NMR binding constants are in good agreement with the values derived from full-titration ITC and fluorescence spectroscopy measurements. 相似文献

Two methods of protein extraction for soybean seeds were evaluated in terms of preservation of the metal ions bound to proteins
after the extraction and separation procedures. The proteins were firstly separated according to their molar masses by polyacrylamide
gel electrophoresis. Then, the protein bands were mapped by synchrotron radiation X-ray fluorescence in order to establish
which metal ions were present in each one. Finally, some mapped protein bands were decomposed by microwave-assisted combustion
and Ca, Cu, K, Mg, Mn, and Zn were quantified by inductively coupled plasma mass spectrometry or inductively coupled plasma
optical emission spectrometry. The extraction methods studied were Method A (based on the treatment of ground soybean seeds with hexane and their extraction with Tris–HCl and β-mercaptoethanol) and
Method B (based on the treatment of ground soybean seeds with petroleum ether and their extraction with Tris–HCl, dithiothreitol,
phenylmethanesulfonyl fluoride, sodium dodecyl sulfate and potassium chloride). The best method was Method B, in which a 78% higher extraction efficiency was obtained when compared to Method A. The electrophilic iodocyclization reaction of (Z)- and (E)-5-n-alkylsubstituted 4-alken-1-ols followed by base-induced hydrodeiodination reaction stereoselectively gave, respectively, (Z)- and (E)-alkylidentetrahydrofurans in high yield. Completely different outcomes were observed with (Z)- and (E)-6,6-dimethylhept-4-en-1-ol: their iodocyclization furnished, respectively, threo- and erythro-2-(1-iodo-2,2-dimetylpropyl)tetrahydrofuran with high stereoselectivity. The threo isomer gave clean formation of 6-tert-butyl-3,4-dihydro-2H-pyran by base-induced ring expansion, while erythro isomer underwent a base-induced ring contraction to 1-cyclopropyl-3,3-dimethylbutan-1-one. Moreover, (Z)- and (E)-5-cyclopropylpent-4-en-1-ol underwent a 6-endo-iodocyclization to threo- and erythro-2-cyclopropyl-3-iodotetrahydro-2H-pyran, respectively, that under the same basic treatment, gave two isomeric 6-cyclopropyldihydro-2H-pyrans in a stereoselective fashion. 相似文献

Methyl or silyl dissociation in the CH(2)=CHCH(2)-XH(3) (a-XH(3)(*)(+)) and CH(2)=CHCH=CHCH(2)-XH(3) (p-XH(3)(*) (+)) radical cations (X = C, Si) yields a(+) or p(+) and XH(3)(*). Similarly, the radical anions a-CH(3)(*) (-) and p-CH(3)(*) (-) give the pi-delocalized anion and CH(3)(*) preferentially. In contrast, a-SiH(3)(*) (-) and p-SiH(3)(*-) prefer to dissociate into the pi-delocalized radical and silide. All reactions are endoergic: by 43-50 kcal mol(-)(1) in the radical cations, and easier to some extent in the radical anions, that require 29-33 (X = C) and 13-14 kcal mol(-)(1) (X = Si). The fragmentation energy profiles do not present significant barriers for the backward process in the case of the radical cations. All radical anions exhibit an energy maximum along the dissociation pathway, but the barrier is lower than the dissociation limit. Fragmentation is "activated" more in the anions than in the cations with respect to homolysis in the corresponding neutrals (that requires 72-81 kcal mol(-)(1)). Wave function analysis indicates that the C-X bond cleavage in the hydrocarbon radical ions, although formally comparable to a homolytic process, is at variance with this model, due to the spin recoupling of one of the two C-X bond electrons with the originally unpaired electron. This is basically true also for the silyl-substituted radical anions, in which the initial more delocalized charge distribution might suggest some heterolytic character of the bond cleavage. 相似文献

Synthesized hydrated lamellar acidic crystalline magadiite (H2Si14O29·2H2O) nanocompound was used as host for intercalation of polar n-alkylmonoamine molecules of the general formula H3C(CH2)nNH2 (n = 1–6) in aqueous solution. The original interlayer distance (d) of 1500 pm, determined by X-ray powder diffraction patterns, increases after intercalation. The values correlated with the number of aliphatic amine carbon (nc) atoms: d = [(1312 ± 11) + (21 ± 2)]nc. The amount of intercalated amines (Ns), decreased as nc increased: Ns = [(5.82 ± 0.04) − (0.45 ± 0.01)]nc. The acidic layered nanocompound was calorimetrically titrated with the amines and the thermodynamic data gave exothermic values for all guest molecules, as shown by the correlation: ΔintH = −[(24.45 ± 0.49) − (1.91 ± 0.10)]nc and d = [(1576 ± 16) − (10.8 ± 1.0)]ΔintH. The negative values of the Gibbs energies and the positive entropies also presented the correlations: ΔintG = −[(22.8 ± 0.2) − (0.2 ± 0.1)]nc and ΔintS = [(6 ± 1) + (5 ± 1)]nc, respectively. 相似文献

Molecular distillation was studied for the separation of tocopherols from soya sludge, both experimentally and by simulation,
under different operating conditions, with good agreement. Evaporator temperatures varied from 100°C to 160°C and feed flow
rates ranged from 0.1 to 0.8 kg/h. The process pressure was maintained at 10−6 bar, the feed temperature at 50°C, the condenser temperature at 60°C, and the stirring at 350 rpm. For each process condition,
samples of both streams (distillate and residue) were collected and stored at −18°C before tocopherols analyses. Owing to
the differences between molecular weights and vapor pressures of free fatty acids and tocopherols, tocopherols preferentially
remained in the residue at evaporator temperatures of 100°C and 120°C, whereas for higher temperatures (140°C and 160°C) and
