Merocyanine 540 (MC540) has been used as external probe to determine the interaction of the peptide sequence 125-139 corresponding to the E2 protein of Hepatitis G virus, with lipid bilayers. The probe was incorporated into large unilamellar vesicles (LUVs) or small unilamellar vesicles (SUVs) of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC). When incorporated into bilayers, MC540 shows two absorption maxima corresponding to the monomer (570 nm) and dimer (530 nm) form of the probe. Changes in the probe microenvironment are reflected by a modification in the position and/or intensity of these maxima. Addition of increasing amounts of peptide resulted in a slight decrease of the ratio A570/A530 thus indicating a change in MC540 partition into the membrane, going from a hydrophobic to a more hydrophilic environment. This effect was concomitant with an increase in dimer formation as stated from the values of the apparent dimerization constant (K(app)) obtained. Fluorescence spectra as well as steady state anisotropy measurements were in agreement with the above results indicating that the peptide was able to relocate the probe and displacing MC540 from its initial location into the bilayer. Results with SUVs or LUVs were similar for what curvature does not seem to play any role on peptide activity. These results reflect the ability of peptide to interact with biomimetic membranes in the lipid head group region.  相似文献

Compression isotherms of mixed monolayers of phosphatidylcholine (PC) and sulphatides (S) were carried out, with the lipids being spread on subphases of different pH and different ionic composition. Several common trends can be observed. All monolayers when compressed reach collapse. Collapse pressures are directly related to the molar fractions of the lipids in the monolayer. Miscibility is not ideal, in general, and is independent on the surface pressure of the monolayer, the mixed monolayers being always (with some exceptions) in a more expanded state than those of the pure components. The thermodynamic parameters corresponding to the mixing interactions have been calculated at the molar composition that gives a maximum deviation from ideality.  相似文献

This paper reports on an experimental study of saturated flow boiling of R134a inside a circular vertical quartz tube coated with a transparent heater. The inner diameter of the tube was 1.33 mm and the heated length 235.5 mm. The flow pattern at high vapor qualities and the dryout of the liquid film were studied using a high speed CCD camera at the mass fluxes 47.4 and 124.4 kg/m2 s in up flow at 6.425 bar. The heat fluxes ranged from 5 to 13.6 kW/m2 for the lower mass flux and from 20 to 32.4 kW/m2 for the higher mass flux.

The behavior of the flow close to dryout was found to be different at low and high mass flux. At low mass flux the location of the liquid front fluctuated with waves passing high up in the tube. In between the waves, a thin film was formed, slowly evaporating without breaking up.

At high mass flux the location of the liquid front was more stable. In this case the liquid film was seen to break up into liquid streams and dry zones on the tube wall.  相似文献

To prepare liposomes containing a synthetic hepatitis A virus antigen (HAV) [VP3(110-121)] as a vaccine, the miscibility of this peptide (with negative net charge) with a neutral lipid [dipalmitoylphosphatidylcholine (DPPC)], a negatively charged lipid [dipalmitoylphosphatidylglycerol (DPPG)], and a positively charged lipid [Stearylamine (SA)] was studied through compression isotherms of monolayers. Mixtures with DPPC and SA showed a low degree of interaction with the peptide, the composition of the monolayer being stable through compression. For DPPG-containing monolayers larger positive deviations from ideality were found, and the peptide was squeezed out from the monolayer at a DPPG/VP3(110-121) mole fraction of 0.8/0.2. All this suggests that besides hydrophobic interactions between the peptide and the lipid, electrostatic forces also play a role; thus it seems that neutral and positively charged lipids would be more suitable for preparing stable liposomes with VP3(110-121). A study of enzyme lipolysis by pancreatic phospholipaseA 2 and by vipera berus phospholipaseA 2 on monomolecular mixed films of didecanoyl-lecithin and triolein on an aqueous subphase of pH 8 has been carried out. The influence of the composition of the mixed film, the surface pressure of the film and the amount and type of the injected enzyme on the lipolysis rate were studied.In order to relate the lipolytic activity with the monolayer state, the compression isotherms of the didecanoyl-lecithin/triolein mixed monolayers have also been obtained.The resuls are compared to observations on lipolytic activity of phospholipaseA 2 on the didecanoyl-lecithin/cholesterol mixed monolayers. Triolein improves the kinetic conditions of the lipolysis of lecithin films in a higher degree than cholesterol. Probably it increases the enzyme penetration by the fluidifying effect exerted on the lecithin monolayers.  相似文献
