Compound 1, 2-(4-phenoxyphenylsulfonylmethyl)thiirane, is a potent and selective inhibitor for human gelatinases (J. Am. Chem. Soc. 2000, 122, 6799-6800), enzymes implicated in a number of diseases, including cancer. This compound is showing excellent promise in animal trials in a number of disease models. Large quantities of this compound were necessary for these studies. A convenient four-step synthetic route for compound 1 is described herein. The synthesis is amenable to scale-up to tens of grams and gives an overall yield of 57% for this important compound.  相似文献

Self-assembly of a new precursor [Cu(L)](ClO4)2 (1) (L = macrocyclic ligand) with octacyanometalates [M(CN)8]3- (M = Mo, W) produced two-dimensional cyano-bridged Cu(II)-M(V) bimetallic assemblies [Cu(L)]3n[M(CN)8]2n.6nH2O [M = Mo (2), W (3)] with novel honeycomblike structures, characterized by spectroscopic data, single-crystal X-ray diffraction studies, and magnetic measurements. The crystallographic determination reveals that compounds 2 and 3 are isostructural and crystallize in the triclinic system (P). The Cu atom in a distorted octahedral environment experiences a tetragonal elongation of apical nitrogen atoms exhibiting average Cu-Nax lengths of 2.566 Angstroms for 2 and 2.593 Angstroms for 3, which accounts for the Jahn-Teller effect of a Cu(II) ion. The Cu-NC angles are magnetically important, ranging from 135.7 to 159.2 degrees. Three types of L in the crystal lattice are observed, which are dependent on the relative positions of the pendant hydroxyl groups with respect to the CuN4 basal plane. The positions are correlated with hydrogen bonding of OH groups to neighboring atoms. The magnetic data indicate that ferromagnetic and antiferromagnetic interactions between Cu(II) and M(V) through the CN linkage coexist.  相似文献

Blends of ABS (acrylonitrile–butadiene–styrene) with phenoxy(poly(hydroxyether bisphenol A)) were prepared using a Branender single screw extruder. Scanning and transmission electron micrographs (SEM, TEM) showed a typical two-phase morphology; particle-in-matrix (90/10) (ABS/phenoxy by weight), 70/30, 10/90), island/sea (30/70) and co-continuous (50/50) morphologies. The glass transition temperature (Tg) of SAN was almost unchanged in the blends, while the Tg of phenoxy increased by about 5 °C in the blends. The synergistic effect of tensile modulus and strength was noted in ABS-rich blends, where a drastic drop of ductility was seen, and the results were interpreted in terms of rubber particle migration form SAN to phenoxy phase, which was visualized by TEM. Melt viscosity showed yield in ABS-rich blends, and generally followed the log additivity.  相似文献

Transmission gratings of holographic polymer-dispersed liquid crystals (HPDLC) are prepared under controlled reaction conditions by adding various amounts of chain-transfer agent (CTA). The resulting films have a higher gel content with lower cross-link density, less dark reaction, less grating shrinkage with much smoother LC/polymer interfaces, smaller induction period, and fast saturation of diffraction efficiency, which shows a maximum of 95 % with 1 % CTA. An optimum LC content of 35 % is verified on the basis of morphology and reaction kinetics.  相似文献

The growth kinetics is characterized and the moving species is identified for the formation of Ni2Si by Rapid Thermal Annealing (RTA) of sequentially deposited Si and Ni films on a 100 Si substrate. The interfacial Ni2Si layer grows as the square root of time, indicating that the suicide growth process is diffusion-limited. The activation energy is 1.25±0.2 eV in the RTA temperature range of 350–450° C. The results extend those of conventional steady-state furnace annealing quite fittingly, and a common activation energy of 1.3±0.2 eV is deduced from 225° to 450° C. The marker experiment shows that Ni is the dominant moving species during Ni2Si formation by RTA, as is the case for furnace annealing. It is concluded that the two annealing techniques induce the same growth mechanisms in Ni2Si formation.  相似文献

Rab coupling protein (RCP) is upregulated in head and neck squamous cell carcinoma (HNSCC) and is correlated with the progression and survival of patients. However, the role of RCP in one of the aggressive types of HNSCC, oral squamous cell carcinoma (OSCC), remains elusive. In the present study, we identified the important role of Zeb1 in RCP-induced OSCC epithelial-to-mesenchymal transition (EMT) and invasion. RCP induces Zeb1 expression, and silencing Zeb1 expression significantly inhibits RCP-induced OSCC invasion. In addition, Zeb1 upregulates MT1-MMP expression to promote OSCC EMT and invasion. Furthermore, we observed that the β1 integrin/EGFR/β-catenin signaling cascade mediates RCP-induced Zeb1 expression to promote OSCC invasion. Notably, we provide evidence that resveratrol (REV) strongly inhibits RCP-induced Zeb1 expression through blocking β1 integrin endosome recycling and EGFR activation, leading to suppression of RCP-induced OSCC invasion, demonstrating the important role of RCP in OSCC invasion and its reversion by REV. Collectively, the present study provides evidence for the first time that RCP aggravates OSCC invasion through increasing Zeb1 expression and subsequently upregulating MT1-MMP expression and that this process is reversed by REV, providing novel biomarkers and indicating the therapeutic potential of REV in OSCC.Subject terms: Oral cancer, Cell invasion  相似文献

Two-photon photodynamic therapy (TP-PDT) is a promising approach for the treatment of cancer because of its better penetration depth and superior spatial selectivity. Here, we describe an azo group containing cyclized-cyanine derivatives (ACC1 and ACC2) as a two-photon activated, type I based photosensitizer (PS). These small-molecule and heavy atom-free organic dyes showed marked reactive oxygen species (ROS)-generating ability under physiological conditions, as well as fast loading ability into the cells and negligible dark toxicity. Live cell analyses with one- and two-photon microscopy revealed that these dyes showed higher ROS generation ability upon two-photon excitation than upon one-photon excitation via the type I process. The PSs have superior PDT properties compared to conventional Visudyne and 5-ALA under mild conditions. These characteristics allowed for precise PDT at the target region in mimic tumor spheroids, demonstrating that the developed TP PS could be useful in efficient PDT applications and in designing various PSs.

Azo containing dyes as a two-photon selective and type I based photosensitizers (PSs) were developed that exhibit excellent photodynamic therapy properties under mild condition.  相似文献
