A comparative study of C(18) immobilized on silica, activated carbon and a polyurethane foam, as sorbents for Pb complexed with O,O-diethyl-dithiophosphate in a flow injection preconcentration system is reported. The complex was formed in 1.0 M HCl medium and processed in a simple system using a peristaltic pump, a manual injector-commutator and a mini-column filled with the sorbent. Using ethanol as eluent, the richest 150-mul fraction was collected and measured (after discarding 150, 200 and 0 mul for the activated carbon, foam and C(18), respectively) by flame atomic absorption spectrometry. The optimum concentration of the complexing agent was 0.05% m/v for C(18) and 0.2% m/v for the activated carbon and the polyurethane foam. The best sample loading flow rate was 4.0 ml min(-1) for the activated carbon and 2.0 ml min(-1) for C(18) and the polyurethane foam, while the best elution flow rate was 1.0 ml min(-1) (activated carbon) and 0.6 ml min(-1) (C(18) and foam). It was found that beyond a certain loading sample volume, for a constant analyte mass, the signal decreased. The maximum loading sample volume, for a constant analyte mass, before the signal started to decrease, was 50 ml for the activated carbon and 150 ml for the other materials. By processing 25 ml, the enrichment factors were 23, 55 and 166 for the activated carbon, foam and C(18), respectively. The best limit of detection (3sigma) was 0.3 mug l(-1) for the C(18) (1.2 mug l(-1) for the foam and 3 mug l(-1) for the activated carbon). As shown, the C(18) has a much superior retention performance in comparison to the other two materials.  相似文献

Bacterial nanocellulose (BNC) is a natural biopolymer produced by different strains of acetic acid bacteria. Biocompatibility, lack of immunogenicity, mechanical strength and crystallinity make BNC a highly applicable product for drug delivery and wound dressing. Previously, we demonstrated that diethylditiocarbamate (DETC), a Superoxide Dismutase 1 inhibitor, incorporated into BNC bio-curatives was effective for treating Cutaneous leishmaniasis (CL) lesions, a parasitic disease caused by Leishmania. We herein investigated the interactions between DETC and BNC. For this purpose, DETC was incorporated into BNC and thermal analysis, x-ray diffraction and Scanning electron microscopy were performed. Furthermore, in vitro DETC release and stability tests as well as degradation studies were also performed. Our results show that DETC is well incorporated into BNC, however it is short lived as suggested by degradation experiments. Future use of BNC DETC-based bio-curatives for the treatment of CL shall require further development in order to increase stability of DETC in the bio-curative.

In this work, mathematical modeling was employed to assess the dynamic behavior of the flash fermentation process for the production of butanol. This process consists of three interconnected units as follows: fermentor, cell retention system (tangential microfiltration), and vacuum flash vessel (responsible for the continuous recovery of butanol from the broth). Based on the study of the dynamics of the process, suitable feedback control strategies [single input/single output (SISO) and multiple input/multiple output (MIMO)] were elaborated to deal with disturbances related to the process. The regulatory control consisted of keeping sugar and/or butanol concentrations in the fermentor constant in the face of disturbances in the feed substrate concentration. Another objective was the maintenance of the proper operation of the flash tank (maintenance of the thermodynamic equilibrium of the liquid and vapor phases) considering that oscillations in the temperature in the tank are expected. The servo control consisted of changes in concentration set points. The performance of an advanced controller, the dynamic matrix control, and the classical proportional-integral controller was evaluated. Both controllers were able to regulate the operating conditions in order to accommodate the perturbations with the lowest possible alterations in the process outputs. However, the performance of the PI controller was superior because it showed quicker responses without oscillations.  相似文献

Nanomedicine is considered a promising alternative to improve cancer diagnosis and treatment. Particularly, the use of nanoparticles (NPs) has enabled the encapsulation of highly toxic anticancer drugs, facilitated ultimate targeting, and allowed tailoring of drug delivery. However, when in biological fluids, these NPs are coated by proteins which hide the targeting moieties and suppress the engineered biological outcome. Herein, how the Ki-1 monoclonal antibody (mAb) can preserve its targetability through grafting on the surface of zwitterionic-functionalized nanoparticles, is unveiled. Zwitterions, known for their stealth ability, are used to minimize unspecific NPs protein adsorption and consequently maintain mAb functionality. In this work, Ki-1 mAb is used as it recognizes TNFRSF8 (CD30+) transmembrane protein overexpressed on CD30+ lymphoma cells such as L540 cells. While nonfunctionalized NPs show negligible toxic effects toward L540 cells, the Ki-1-functionalized structure demonstrates cytotoxicity, since they undergo cellular uptake, suggesting a receptor-mediated internalization. This dual-functionalization strategy provides a promising multifunctional nanoplatform toward future personalized medicine applications, minimizing unspecific protein adsorption on NPs and ensuring selective cancer cell targeting.  相似文献

We discuss some results of a study carried out over the past 4 years to investigate the role of Modellus, a software package, in the development of an approach to teaching calculus for Biology majors. The central idea of the teaching approach is to propose the analysis of a mathematical model for a biological phenomenon at the very beginning of the course, in a way that this analysis is interrelated with some of the mathematical concepts listed in the syllabus. In this paper, we focus on the role of the software during the development of one of the activities proposed to the students, the purpose of which was to discuss the relation between secant lines and the instantaneous rate of change. It was found that this software played two roles in the development of this activity: providing information about the phenomenon and the model; and acting as a trigger, making evident to the student an important aspect that contributed to his understanding. Based on our theoretical perspective of digital technology, we believe that students’ interaction with the software played a fundamental role in the thinking collective composed of humans and media involved in mathematical learning.  相似文献

Silver‐based nanocomposites are known to act as biocides against a series of microorganisms and are largely studied as an alternative to substitute conventional antibiotics that show decreasing efficacy. In this work, an eco‐friendly method to synthesize silver nanoparticles assembled on the surface of hexaniobate crystals is reported. By means of ion exchange, K+ ions of layered potassium hexaniobate were partially substituted by Ag+ ions and the resulting material was exposed to UV light. The irradiation allowed the reduction of silver ions with consequent formation of silver nanoparticles located only on the hexaniobate surface, whereas Ag+ ions located in the interlayer space remained in the ionic form. Increasing UV‐light exposure times allowed controlling of the silver nanoparticle size. The antibacterial effects of the pristine potassium hexaniobate and of silver‐containing hexaniobate samples were tested against Escherichia coli (E. coli). The antibacterial efficacy was determined to be related to the presence of silver in hexaniobate. An increasing activity against E. coli was observed with the decrease in silver nanoparticles size, suggesting that silver nanoparticles of distinct sizes interact differently with bacterial cell walls.  相似文献

Point-of-care platforms can provide fast responses, decrease the overall cost of the treatment, allow for in-home determinations with or without a trained specialist, and improve the success of the treatment. This is especially true for microfluidic paper-based analytical devices (μPAD), which can enable the development of highly efficient and versatile analytical tools with applications in a variety of biomedical fields. The objective of this work was the development of μPADs to identify and quantify levels of nitrite in saliva, which has been proposed as a potential marker of periodontitis. The devices were fabricated by wax printing and allowed the detection of nitrite by a colorimetric reaction based on a modified version of the Griess reaction. The presented modifications, along with the implementation of a paper-based platform, address many of the common drawbacks (color development, stability, etc.) associated with the Griess reaction and are supported by results related to the design, characterization, and application of the proposed devices. Under the optimized conditions, the proposed devices enable the determination of nitrite in the 10–1000 μmol L−1 range with a limit of detection of 10 μmol L−1 and a sensitivity of 47.5 AU [log (μmol L−1)]−1. In order to demonstrate the potential impact of this technology in the healthcare industry, the devices were applied to the analysis of a series of real samples, covering the relevant clinical range.  相似文献

In this work, tetramethylammonium hydroxide (TMAH) was used to solubilize the DORM-1 dogfish muscle certified reference material as a model substance for the determination of As, Cd, Pb and Se by electrothermal atomic absorption spectrometry (ET AAS). The sample was mixed with a small amount of TMAH and heated to 60 °C for 10 min in a water bath. After dissolution, As and Se were determined using palladium and magnesium nitrates as a chemical modifier added in solution. For Cd and Pb, best results were obtained with a mixture of 250 μg of each of iridium and rhodium as permanent modifiers. In both cases, the calibration was performed with aqueous solutions in 0.2% v/v HNO3. The temperature program for each analyte was optimized using pyrolysis and atomization curves established with the fish reference material. The detection limits in dry samples and the characteristic mass values were: Cd 0.005 μg g−1 and 0.9 pg; Pb 0.04 μg g−1 and 7.6 pg; As 0.4 μg g−1 and 13 pg and Se 0.6 μg g−1 and 20 pg, respectively. Results from the determination of these elements in the DORM-1 certified fish reference material were within the 95% confidence interval of the certified values.  相似文献
