STRUCTURE EVOLUTION OF POLYMER CHAINS FOR NECKING FORMATION IN HIGH-SPEED FIBER SPINNING PROCESS

Finite element method is used to simulate the high-speed melt spinning process,based on the equation system proposed by Doufas et al.Calculation predicts a neck-like deformation,as well as the related profiles of velocity,diameter, temperature,chain orientation,and crystallinity in the fiber spinning process.Considering combined effects on the process such as flow-induced crystallization,viscoelasticity,filament cooling,air drag,inertia,surface tension and gravity,the simulated material flow behaviors are consistent with those observed for semi-crystalline polymers under various spinning conditions.The structure change of polymer coils in the necking region described by the evolution of conformation tensor is also investigated.Based on the relaxation mechanism of macromolecules in flow field different types of morphology change of polymer chains before and in the neck are proposed,giving a complete prospect of structure evolution and crystallization of semi-crystalline polymer in the high speed fiber spinning process.     

Synthesis of seven-membered ring diazepin-2-ones via palladium-catalyzed highly regioselective cyclization of 2-vinylpyrrolidines with aryl isocyanates

The first palladium-catalyzed ring-expansion reaction of 2-vinylpyrrolidines with aryl isocyanates to form seven-membered ring heterocycles is described. This regioselective reaction requires 5 mol % of Pd(2)(dba)(3).CHCl(3) and 10 mol % of dppp at 40-60 degrees C in THF and results in the formation of 1,3-diazepin-2-ones in good isolated yields. When Pd(OAc)(2) and PPh(3) were utilized in the reaction, an intramolecular hydrogen migration occurs resulting in the formation of conjugated diene derivatives of urea.     

Simultaneous Determination of Enalapril and Enalaprilat in Human Plasma by LC-MS: Application to a Bioequivalence Study

A rapid, simple and specific liquid chromatography-electrospray ionization mass spectrometry (LC-ESI-MS) method has been developed and validated for the simultaneous determination of enalapril and its major active metabolite enalaprilat in human plasma. Benazepril hydrochloride was used as the internal standard. Plasma was deproteinized with acetone and centrifuged. The supernatant was transferred and evaporated to dryness and the residue dissolved in mobile phase. Samples were separated on a C18 column with a mobile phase of methanol–20 mM ammonium acetate (53:47, v/v) containing 0.15% trifluoracetic acid (v/v) with a pH of 3.0. Enalapril, enalaprilat and the internal standard were measured by electrospray positive selective ion monitoring mode. The method was validated over a linear range of 1.56–400 ng mL⁻¹ and the limits of quantification were 1.56 ng mL⁻¹ for both enalapril and enalaprilat using 0.1 mL plasma. Extraction efficiency was more than 84% and recoveries were in range of 93.65–101.17%. The intra-day relative standard deviations (RSD) were less than 8.16 and 7.05% and inter-day RSDs were within 8.42 and 5.72% for enalapril and enalaprilat, respectively. The storage stability of QC samples was investigated under various conditions. The method was successfully applied for the evaluation of the pharmacokinetics and bioequivalence of enalapril and enalaprilat in 20 healthy volunteers after an oral dose of 20 mg enalapril maleate.     

超临界CO2法制备头孢唑啉钠脂质体

采用超临界CO2(scCO2)流体代替有机溶剂一步法制备了头孢唑啉钠药物载体脂质体. 研究了该脂质体的尺寸、稳定性和药物的包封率. 结果表明, 脂质体的尺寸和稳定性依赖于制备压力, 脂质体对头孢唑啉钠的包封率与乙醇和脂浓度有关, 采用超临界CO2法制备脂质体的药物包封率比采用薄膜分散法(Bangham method)制备的包封率高.     

bPAK-interacting exchange factor may regulate actin cytoskeleton through interaction with actin

p21-activated kinase (PAK)-interacting exchange factor (PIX) is known to be involved in regulation of Cdc42/Rac GTPases and PAK activity. PIX binds to the proline-rich region of PAK, and regulates biological events through activation of Cdc42/Rac GTPase. To further investigate the role of PIX we produced monoclonal antibodies (Mab) against bPIX. Three clones; N-C6 against N-terminal half and C-A3 and C-B7 against C- terminal half of bPIX were generated and characterized. N-C6 Mab detected bPIX as a major band in most cell lines. C-A3 Mab recognizes GIT-binding domain (GBD), but it does not interfere with GIT binding to bPIX. Using C-A3 Mab possible bPIX interaction with actin in PC12 cells was examined. bPIX Mab (C-A3) specifically precipitated actin of the PC12 cell lysates whereas actin Mab failed to immunoprecipitate bPIX. Co-sedimentation of PC12 cell lysates with the polymerized F-actin resulted in the recovery of most of bPIX in the cell lysates. These results suggest that bPIX may not interact with soluble actin but with polymerized F-actin and revealed that bPIX constitutes a functional complex with actin. These data indicate real usefulness of the bPIX Mab in the study of bPIX role(s) in regulation of actin cyoskeleton.     

Biomimetic Asymmetric Reduction of Quinazolinones with Chiral and Regenerable NAD(P)H Models

A facile approach to chiral dihydroquinazolinone derivatives has been described via biomimetic asymmetric reduction of quinazolinones with chiral and regenerable NAD(P)H models. The utility of this method was demonstrated by a concise synthesis of the bromodomain protein divalent inhibitor.     

Application of Functionalized Ag Nanoparticles for the Determination of Proteins at Nanogram Levels Using the Resonance Light Scattering Method

A novel method for the determination of proteins in aqueous solutions has been developed based on the enhancement of resonance light scattering (RLS) of Ag nanoparticles in the presence of proteins. Factors including acidity of the media, concentration of Ag hydrosol, reaction time, temperature, and interference of non-protein substances were investigated. Under the optimal conditions, with the enhanced RLS signals at 452nm, the linear ranges of calibration curves were 0–0.8µgmL^–1 for bovine serum albumin (BSA), 0–1.2µgmL^–1 for human serum albumin (HSA), and 0–2.5µgmL^–1 for human -IgG (-IgG), respectively. The detection limits were 1.3ngmL^–1 for BSA, 10ngmL^–1 for HAS, and 5.7ngmL^–1 for -IgG.This method has been applied to the analysis of synthetic samples and real human serum samples, and the results were in good agreement with those reported by the hospital, indicating that the method presented here is not only sensitive and simple, but also reliable and suitable for practical applications.     

Two New Cyclopeptides from Arenaria oreophila (Caryophyllaceae)

Two new cyclopeptides, named arenariphilin A ( 1 ) and arenariphilin B ( 2 ), were isolated from the whole plants of Arenaria oreophila. Their structures were determined as cyclo‐(Thr‐Gly) ( 1 ) and cyclo‐(Ser₁‐Gly ‐Ser₂‐Ile ‐Phe₁‐Phe₂) ( 2 ) on the basis of spectral data, especially by 2D‐NMR.
