Central retinal vein occlusion (CRVO) is a visually disabling condition resulting from a thrombus in the major outflow vessel of the eye. The inflammatory response in CRVO is effectively treated with a dexamethasone (DEX) intravitreal implant. Uncovering the proteome changes following DEX implant intervention in CRVO may identify key proteins that mediate the beneficial effects of DEX. In six Göttingen minipigs, CRVO was induced in both eyes with an argon laser using a well-established experimental model. The right eyes were treated with a DEX intravitreal implant (Ozurdex, Allergan), while the left control eyes received a sham injection. Eight weeks after DEX intervention, retinal samples were collected and analyzed with tandem mass tag-based mass spectrometry. DEX implant intervention resulted in the upregulation of peptidyl-prolyl cis–trans isomerase FKBP5 (FKBP5) and ubiquilin-4. Immunohistochemistry showed expression of FKBP5 in the nuclei in all cellular layers of the retina. Cell adhesion molecule 3, tumor necrosis factor receptor superfamily member 16, and trans-1,2-dihydrobenzene-1,2-diol dehydrogenase were downregulated following DEX intervention. The upregulation of the corticosteroid-sensitive protein FKBP5 suggests that the implant remained active at the molecular level after eight weeks of treatment. Future studies may investigate if FKBP5 regulates the efficacy and duration of the DEX implant.  相似文献

This is the first in a series of papers devoted to an analogue of the metaplectic representation, namely the minimal unitary representation of an indefinite orthogonal group; this representation corresponds to the minimal nilpotent coadjoint orbit in the philosophy of Kirillov–Kostant. We begin by applying methods from conformal geometry of pseudo-Riemannian manifolds to a general construction of an infinite-dimensional representation of the conformal group on the solution space of the Yamabe equation. By functoriality of the constructions, we obtain different models of the unitary representation, as well as giving new proofs of unitarity and irreducibility. The results in this paper play a basic role in the subsequent papers, where we give explicit branching formulae, and prove unitarization in the various models.  相似文献

In this paper we apply a statistical model combining a random coefficient regression model and a latent class regression model. The EM-algorithm is used for maximum likelihood estimation of the unknown parameters in the model and it is pointed out how this leads to a straightforward handling of a number of different variance or covariance restrictions. Finally, the model is used to analyze how consumers' preferences for eight coffee samples relate to sensory characteristics of the coffees. Within this application the analysis corresponds to a model-based version of the so-called external preference mapping.  相似文献

The use of near infrared spectroscopy (NIRS) for rapid determination of the degree of interesterification of blends of palm stearin, coconut oil, and rapeseed oil obtained using an immobilized Thermomyces lanuginosa lipase at 70 °C was investigated. Interesterification was carried out by applying both fixed bed and batch reactors. Calibrations were developed for quantitative determination of solid fat content (SFC) at 10, 20, 30, 35, and 40 °C and free fatty acid (FFA) resulting in root mean square errors of prediction of 1.0, 1.3, 1.4, 1.6, 1.7, and 0.19% (w/w), respectively. The data showed that NIRS could be used to replace the traditional methods for determining FFA and SFC in vegetable oils.It was possible to monitor the activity of the immobilized enzyme for interesterification of margarine oils by predicting the equivalent reaction time in a batch reactor from NIR spectra. Root mean square errors of prediction for two different oil blends interesterified for 300 and 170 min were 21 and 12 min, respectively.  相似文献
