The role of three highly conserved insulin residues Tyr^B26 was studied to better understand the relationship between insulin and receptor from rat adipose tissue plasma membranes, lnsulin analogues with a single amino acid substitution or single N-methylation of the peptide bond in the position B26 were all shortened in the C-terminus of the B-chain by four amino acids. The effect of modifications was followed by the binding to the insulin receptor. From our results, we can deduce several conclusions: (1) the replacement of tyrosine in the position B26 by histidine, [N-MeHis^B26]-des-tetrapeptide-(B27-B30)-insulin-B26-amide and [N-MeGlu^B26]-des-tetrapeptide- (B2-B30)-insulin-B26-amide, have no significant effect on the binding affinity and they show binding affinity 105%, 190% and 208%, respectively, of that of human insulin; (2) [Aad^B26] -des-tetrapeptide-(B27-B30)-insulin-B26-amide and [Phe(4-carboxy^B26)]-des-tetrapeptide- (B27~B30)-insulin-B26-amide affect the potency highly positively in vitro studies; they show binding affinity 529 and 289 %, respectively, of that of human insulin. 相似文献

A facile method for the fabrication of inorganic hollow materials from cuprammonium cellulose composite filaments based on fast pyrolysis has been developed. Unlike Ostwald ripening, approaches based on the Kirkendall effect, and other template methods, this process yielded hollow materials within 100 s. The heterogeneous structure of the cellulose composite fibers and the gradient distribution of the metal oxides are the main reasons for the formation of the hollow structure. The diameter, wall thickness, and length of the hollow microfibers could be conveniently controlled. With their perfect morphology, these hollow structural materials have great potential for use in various fields. 相似文献

A high power cryogenic cooling Tm-doped (2%) GdVO4 laser double-end-pumped by fiber-coupled-diode with the center wavelength of 804.5 nm at 21 ℃ is reported. The highest continuous-wave (CW) power of 2.35 W at 1903 nm is attained at pump power of 24 W. The slope efficiency is 12.5% and the threshold is 3.2 W. It is reported a novel optical parametric oscillator with KTP as medium crystal inside a Nd∶YAG phase conjugate resonator. In the phase conjugate resonator, the laser beam quality is promoted by phase-conjugate mirror, which is realized by stimulated Brillouin scattering effect, and Q-switch is generated simultaneously. A novel wavelength assignment scheme called the wavelength pre-assignment collision schedule (WPCS) is proposed for wavelength-routed networks. The WPCS pre-assigns the wavelength at the forward detection phase, and schedules the potential collision by priority. The potential collision is scheduled at the forward detection phase and the blocking of the wavelength assignment is reduced. Simulation is conducted with several other existing schemes.
