Autophagy is a cellular homeostatic process by which cells degrade and recycle their malfunctioned contents, and impairment in this process could lead to Parkinson’s disease (PD) pathogenesis. Dioscin, a steroidal saponin, has induced autophagy in several cell lines and animal models. The role of dioscin-mediated autophagy in PD remains to be investigated. Therefore, this study aims to investigate the hypothesis that dioscin-regulated autophagy and autophagy-related (ATG) proteins could protect neuronal cells in PD via reducing apoptosis and enhancing neurogenesis. In this study, the 1-methyl-4-phenylpyridinium ion (MPP+) was used to induce neurotoxicity and impair autophagic flux in a human neuroblastoma cell line (SH-SY5Y). The result showed that dioscin pre-treatment counters MPP+-mediated autophagic flux impairment and alleviates MPP+-induced apoptosis by downregulating activated caspase-3 and BCL2 associated X, apoptosis regulator (Bax) expression while increasing B-cell lymphoma 2 (Bcl-2) expression. In addition, dioscin pre-treatment was found to increase neurotrophic factors and tyrosine hydroxylase expression, suggesting that dioscin could ameliorate MPP+-induced degeneration in dopaminergic neurons and benefit the PD model. To conclude, we showed dioscin’s neuroprotective activity in neuronal SH-SY5Y cells might be partly related to its autophagy induction and suppression of the mitochondrial apoptosis pathway. 相似文献

Four linearly independent linear combinations of the ten sextic centrifugal distortion coefficients of triangular triatomic molecules are presented. These combinations are formed in such a way that the resulting expressions depend only on the equilibrium geometry and the harmonic force field parameters of the molecule, quantities which are ordinarily known to considerably better precision than the sextic constants. The relations obtained thus constitute a set of planarity constraints on the ten sextic coefficients which arise in connection with the Nielsen-Amat expansion of the Darling-Dennison Hamiltonian. The robust optimization methodology is known as a popular method dealing with optimization problems with uncertain data and hard constraints. This methodology has been applied so far to various convex conic optimization problems where only their inequality constraints are subject to uncertainty. In this paper, the robust optimization methodology is applied to the general nonlinear programming (NLP) problem involving both uncertain inequality and equality constraints. The uncertainty set is defined by conic representable sets, the proposed uncertainty set is general enough to include many uncertainty sets, which have been used in literature, as special cases. The robust counterpart (RC) of the general NLP problem is approximated under this uncertainty set. It is shown that the resulting approximate RC of the general NLP problem is valid in a small neighborhood of the nominal value. Furthermore a rather general class of programming problems is posed that the robust counterparts of its problems can be derived exactly under the proposed uncertainty set. Our results show the applicability of robust optimization to a wider area of real applications and theoretical problems with more general uncertainty sets than those considered so far. The resulting robust counterparts which are traditional optimization problems make it possible to use existing algorithms of mathematical optimization to solve more complicated and general robust optimization problems. 相似文献

Heavy metals are the harmful elements, regarded as carcinogens. Nevertheless, owing to their physical and chemical properties, they are still used in the production of several commercial products. Utilization of such products increases the chance for the exposure of heavy metals, some of them are categorized as probable human carcinogens (Group 1) by the International Agency for Research on Cancer. Exposure of heavy metals to school children at early age can result severe life time health issues and high chance of emerging cancer. Thus, we have performed study relating to the presence of heavy metals in acrylic color paints commonly used by the school children. Acrylic paints of different colors were assayed for seven potential heavy metals manganese (Mn), cobalt (Co), nickel (Ni), zinc (Zn), arsenic (As), cadmium (Cd) and lead (Pb) using microwave digestion and iCAPQ inductively coupled plasma mass spectrometry (ICP-MS) system. The optimized method including paints digestion reagents nitric acid (HNO3, 65%, 5 mL) and hydrofluoric acid (HF, 40%, 2 mL) have offered excellent method performance with recovery values ranged between 99.33% and 105.67%. The elements were identified in all of the analyzed samples with concentrations ranged from 0.05 to 372.59 µg/g. Cd constitutes the lower percentage (0.05%), whereas Zn constitutes high ratio contribution which was tremendously high (68.33%). Besides, the paints contamination was also color specific, with considerably total heavy metal concentrations found in brunt umber (526.57 µg/g) while scarlet color (12.62 µg/g) contained lower amounts.
