UV resonance Raman spectroscopy has been used to determine the conformational energy landscape of poly-L-lysine (PLL) in the presence of NaClO4 as a function of temperature. At 1 degree C, in the presence of 0.83 M NaClO4, PLL shows an approximately 86% alpha-helix-like content, which contains alpha-helix and pi-bulge/helix conformations. The high alpha-helix-like content of PLL occurs because of charge screening due to strong ion-pair formation between ClO4- and the lysine side chain -NH3+. As the temperature increases from 1 to 60 degrees C, the alpha-helix and pi-bulge/helix conformations melt into extended conformations (PPII and 2.51-helix). We calculate the Psi Ramachandran angle distribution of the PLL peptide bonds from the UV Raman spectra which allows us to calculate the PLL (un)folding energy landscapes along the Psi reaction coordinate. We observe a basin in the Psi angle conformational space associated with alpha-helix and pi-bulge/helix conformations and another basin for the extended PPII and 2.51-helical conformations. 相似文献

A method for flow-through purification of viruses and virus like nano-particles using a combination of binding and size-exclusion chromatography was developed. This technique relies on minimizing the external surface area per unit volume available for virus binding by increasing the mean diameter of the beads used in the column. At the same time the impurity binding capacity of the column is maximized by utilizing beads with multiple functionalities of the optimum size. Purification of different types of viruses and virus-like-particles could be achieved using this technique. Flow-through purification of influenza virus using this technique yielded virus recoveries greater than 70-80% coupled with impurity removal greater than 80%. Finally an approach to optimize and facilitate process development using this technology is presented. Since the impurity binding occurs via a non-specific mechanism and virus recovery is achieved through reduced surface area, the technique is not limited to specific types of viruses and offers the potential as a universal purification tool. 相似文献

We developed a convenient and fast approach to preparing close-packed two-dimensional (2-D) particle arrays on mercury surfaces. Addition of cosolvents, such as alcohols, to aqueous colloidal particle suspensions induces spreading and self-assembly of the particles into 2-D arrays on top of the mercury surface. We can fabricate large-area close-packed 2-D arrays (>70 cm(2)) within 30 s. We attached these 2-D arrays to functional hydrogel films such that the 2-D array spacings were altered by the hydrogel volume response to the environment. We directly observed the hydrogel volume induced 2-D array spacing changes by using confocal laser scanning microscopy to monitor the spacings of fluorescent polystyrene particle 2-D arrays in response to changes in pH, solvent composition, temperature, etc. 相似文献

A method for observing near-surface fluctuations in pH caused by a water–air flux of carbon dioxide under conditions of ambient
atmospheric carbon dioxide levels is developed and tested. Peaks in fluorescence intensity measured as a function of pH and
turbulence are shown to be consistent with predictions from a chemical kinetics model of CO2 exchange. The square root of the frequency of the pH fluctuations scale linearly with independently measured bulk air–water
