We describe the use of iterative in situ click chemistry to design an Akt-specific branched peptide triligand that is a drop-in replacement for monoclonal antibodies in multiple biochemical assays. Each peptide module in the branched structure makes unique contributions to affinity and/or specificity resulting in a 200 nM affinity ligand that efficiently immunoprecipitates Akt from cancer cell lysates and labels Akt in fixed cells. Our use of a small molecule to preinhibit Akt prior to screening resulted in low micromolar inhibitory potency and an allosteric mode of inhibition, which is evidenced through a series of competitive enzyme kinetic assays. To demonstrate the efficiency and selectivity of the protein-templated in situ click reaction, we developed a novel QPCR-based methodology that enabled a quantitative assessment of its yield. These results point to the potential for iterative in situ click chemistry to generate potent, synthetically accessible antibody replacements with novel inhibitory properties.  相似文献

A simple procedure for selective sorption of tungsten is described. The method involves reduction of W(VI) to W(V) with tin(II) chloride (2%, w/v) at 8-9M hydrochloric acid, formation of the W(V)-SCN complex with 0.2M KSCN and its sorption on polyurethane foam within 20 min. The sorbed complex is then eluted with acidified acetone (1 ml of 1M hydrochloric acid and 8 ml of acetone) followed by addition of 1 ml of 0.1M KSCN to the eluent. The method has been applied to the spectrophotometric determination of tungsten in steels and silicates by measuring the absorbance of the eluted solution at 400 nm. Beer's law is obeyed for the range 0.1-12 mug W/ml. Other elements, e.g., Co(III) (50 mug/ml), Cu(II) (10 mug/ml), Ti(IV) (20 mug/ml), V(V) (10 mug/ml) and Mo(VI) (0.5 mug/ml) have no effect on the method. Interference of copper, up to 100 mug/ml has been eliminated by masking with thiourea and that due to molybdenum by prior separation with thioglycollic acid on PUF. Reported herein is a series of pore-containing polymeric nanotubes based on a hydrogen-bonded hydrazide backbone. Nanotubes of suitable lengths, possessing a hollow cavity of about a 6.5 Å diameter, mediate highly efficient transport of diverse types of anions, rather than cations, across lipid membranes. The reported polymer channel, having an average molecular weight of 18.2 kDa and 3.6 nm in helical height, exhibits the highest anion-transport activities for iodide (EC50=0.042 μm or 0.028 mol % relative to lipid), whcih is transported 10 times more efficiently than chlorides (EC50=0.47 μm ). Notably, even in cholesterol-rich environment, iodide transport activity remains high with an EC50 of 0.37 μm . Molecular dynamics simulation studies confirm that the channel is highly selective for anions and that such anion selectivity arises from a positive electrostatic potential of the central lumen rendered by the interior-pointing methyl groups.  相似文献

Cyclo‐oligo‐(1→6)‐β‐D ‐glucosamines functionalized with hydrophobic tails are reported as a new class of transmembrane ion‐transport system. These macrocycles with hydrophilic cavities were introduced as an alternative to cyclodextrins, which are supramolecular systems with hydrophobic cavities. The transport activities of these glycoconjugates were manipulated by altering the oligomericity of the macrocycles, as well as the length and number of attached tails. Hydrophobic tails of 3 different sizes were synthesized and coupled with each glucosamine scaffold through the amide linkage to obtain 18 derivatives. The ion‐transport activity increased from di‐ to tetrameric glucosamine macrocycles, but decreased further when flexible pentameric glucosamine was introduced. The ion‐transport activity also increased with increasing length of attached linkers. For a fixed length of linkers, the transport activity decreased when the number of such tails was reduced. All glycoconjugates displayed a uniform anion‐selectivity sequence: Cl?>Br?>I?. From theoretical studies, hydrogen bonding between the macrocycle backbone and the anion bridged through water molecules was observed.  相似文献

We describe a general synthetic strategy for developing high‐affinity peptide binders against specific epitopes of challenging protein biomarkers. The epitope of interest is synthesized as a polypeptide, with a detection biotin tag and a strategically placed azide (or alkyne) presenting amino acid. This synthetic epitope (SynEp) is incubated with a library of complementary alkyne or azide presenting peptides. Library elements that bind the SynEp in the correct orientation undergo the Huisgen cycloaddition, and are covalently linked to the SynEp. Hit peptides are tested against the full‐length protein to identify the best binder. We describe development of epitope‐targeted linear or macrocycle peptide ligands against 12 different diagnostic or therapeutic analytes. The general epitope targeting capability for these low molecular weight synthetic ligands enables a range of therapeutic and diagnostic applications, similar to those of monoclonal antibodies.  相似文献

We have measured the low frequency elastic properties of dielectric, normal conducting and superconducting metallic glasses at audio-frequencies (f1 kHz) and temperatures down to 10 mK. Our results are discussed in the framework of the tunneling model of glasses. The major assumption of the tunneling model regarding the tunneling states with long relaxation time has been verified, but discrepancies to high frequency measurements have been found. In addition, our experiments on superconducting metallic glasses seem to indicate that the present treatment of the electron-tunneling state interaction is not sufficient.  相似文献

We consider an extension of an almost strictly competitive game, introduced by Aumann (1961), inn-person extensive games by incorporating Selten's subgame perfection. We call it a subgame perfect weakly-almost (SPWA) strictly competitive game, in particular, an SPWA strictly competitive game in strategic form is simply called a WA strictly competitive game. We give some general results on the structure of this class of games. One result gives an easy way to verify almost strict competitiveness of a given extensive game. We show that a two-person weakly unilaterally competitive extensive game, introduced by Kats and Thisse (1992) for normal form games, is SPWA strictly competitive. We remark that some of our main results for SPWA strictly competitive games do not hold for the modification of almost strict competitiveness with trembling-hand perfection.The author is indebted to Mamoru Kaneko for valuable discussions, comments and criticism throughout the paper.
