Detection of peginesatide in equine serum using liquid chromatography-tandem mass spectrometry for doping control purposes

Erythropoietin (EPO) and its recombinant analogues are suspected to be illicitly administered to horses for performance enhancing purposes and, consequently, prohibited in equine sports. Recently, a new erythropoiesis-stimulating agent, peginesatide (Omontys, formerly referred to as Hematide), belonging to the upcoming class of EPO-mimetic peptides, received approval for the treatment of anaemia in humans with chronic kidney disease on dialysis. As the pegylated dimeric peptide of approximately 45 kDa without sequence homology to EPO is not detectable by conventional EPO detection assays, specific methods are bound to be established for horse sports drug testing. Thus, by fortifying equine serum with peginesatide, an approach consisting of a proteolytic digestion with subtilisin after protein precipitation was developed, eventually targeting a proteotypic and xenobiotic pentapeptide which is easily accessible to liquid chromatography- tandem mass spectrometry analysis. The method was validated for qualitative purposes and demonstrated to be specific, precise (relative standard deviations below 14%), sensitive (limit of detection 10 ng mL(-1)) and linear. Being simple, cost-effective and readily transferable to other doping control laboratories, a mass spectrometric assay for the detection of therapeutic concentrations of peginesatide in equine serum is, in terms of preventive doping research, applicable to routine analysis shortly after approval of the drug.     

Synthesis and Characterisation of Bismuth(III) Aminoarenesulfonate Complexes and Their Powerful Bactericidal Activity against Helicobacter pylori

The synthesis and characterisation of nine new tris‐substituted bismuth(III) aminoarenesulfonates of the general formula [Bi(O₃S‐R^N)₃] (R^N=o‐aminophenyl 1 , m‐aminophenyl 2 , 6‐amino‐3‐methoxyphenyl 3 , p‐aminophenyl 4 , 2‐pyridyl 5 , o‐aminonaphthyl 6 , 5‐aminonaphthyl 7 , 4‐amino‐3‐hydroxynaphthyl 8 and 5‐isoquinolinyl 9 ) is described. Two synthetic strategies, using Ag₂O and [Bi(OtBu)₃], were explored and compared. The possibility to access heteroleptic bismuth(III) complexes with the new silver(I) metathesis reaction is demonstrated with the synthesis of the heteroleptic bismuth(III) aminoarenesulfonate complexes [PhBi(O₃S‐P2)₂(dmso)] 10 , [Ph₂Bi(O₃S‐P2)]_∞ 11 and [PhBi(O₃S‐P2)₂]_∞ 12 , of which the solid state structures 10 and 12 are presented (2P‐SO₃^?=2‐pyridinesulfonate). These complexes offer remarkable in‐vitro activity against three standard laboratory strains of Helicobacter pylori (H. pylori) as demonstrated by their exceptionally low minimum inhibitory concentration (MIC) values of 0.049 μg mL^?1 for the strains 251 and B128, which places most MIC values in the nano‐molar region. These results demonstrate the importance of the amino functionality in addition to the sulfonate group on the bactericidal properties against H. pylori.     

Regioselective Synthesis of Trichloromethyl‐Substituted Salicylates and Cyclohexenones by One‐Pot Cyclizations of 1,3‐Bis(trimethylsilyloxy)buta‐1,3‐dienes

A variety of 6‐(trichloromethyl)salicylates (=2‐hydroxy‐6‐(trichloromethyl)benzoates) were prepared by TiCl₄‐mediated cyclization of 1,3‐bis(trimethylsilyloxy)buta‐1,3‐dienes with 1,1,1‐trichloro‐4,4‐dimethoxybut‐3‐en‐2‐one. The employment of trimethylsilyl trifluoromethanesulfonate (Me₃SiOTf) as Lewis acid resulted in the formation of trichloromethyl‐substituted cyclohexenones. The cyclizations proceeded with good‐to‐very‐good regioselectivities.     

The iodine supply of humans depending on site, food offer and water supply

In Central Europe, the iodine content of weathered soils of new red sandstone, Muschelkalk, keuper and lower strata of new red sandstone, which developed from marine sediment is low. Boulder clays and diluvial sands also exhibit low iodine concentrations. In the same areas, the iodine concentration of drinking water decreases from 9 to 1 g/l with increasing distance from the oceans. Cereals and foodstuffs produced from them are extremely iodine-poor. The iodine content of plants increases with the proportion of leaves. Soil contamination raises the iodine content of foodstuffs and improves the iodine supply of animals. The iodination of mineral mixtures increased the iodine content of cow's milk from 17 to 81 g/l. The iodination of packed cooking salt and of the mineral mixtures for cattle and pigs resulted in a mean increase of the iodine intake of adults from about 30 g/day to 48 and 59, respectively. Although iodine intake remained considerably below the calculated requirement for adults (100 to 150 g/day), it led to the disappearance of Struma konnata and reduced the frequency of struma in children.     

Two-dimensional double-quantum 2H NMR spectroscopy in the solid state under OMAS conditions: correlating 2H chemical shifts with quasistatic line shapes.

Solid-state 2H NMR spectroscopy is a well-established and versatile method to study molecular orientation and dynamics in selectively deuterated samples. Herein, we introduce a 2D 2H double-quantum (DQ) NMR experiment performed under fast magic-angle spinning with a slight offset of the magic angle (OMAS). The experiment combines 2H chemical-shift resolution with DQ-filtered quasistatic 2H line shapes. In this way, it is possible to separate 2H resonances and to independently determine 2H quadrupole couplings at multiple sites. While 2H chemical shifts are resolved in the 2H DQ dimension, the quadrupole parameters can be obtained from characteristic line shapes which are reintroduced in the second dimension by the magic-angle offset. The 2D 2H DQ OMAS experiment is demonstrated on L-histidine which was deuterated at multiple sites by recrystallisation from D2O.     

Fungal upgrading of wheat straw for straw-thermoplastics production

Combining biologic pretreatment with storage is an innovative approach for improving feedstock characteristics and cost, but the magnitude of responses of such systems to upsets is unknown. Unsterile wheat straw stems were upgraded for 12 wk with Pleurotus ostreatus at constant temperature to estimate the variation in final compositions with variations in initial moisture and inoculum. Degradation rates and conversions increased with both moisture and inoculum. A regression analysis indicated that system performance was quite stable with respect to inoculum and moisture content after 6 wk of treatment. Scale-up by 150× indicated that system stability and final straw composition are sensitive to inoculum source, history, and inoculation method. Comparative testing of straw-thermoplastic composites produced from upgraded stems is under way.

The 2,5-dititanabicyclo[2.2.0]hex-1(4)-ene (bis-titanocene-mu-(Z)-1,2,3-butatriene complex)3 is formed starting from [Cp2Ti(eta2-Me3SiC2SiMe3)] by in situ generated titanocene and 1,4-dichlorobut-2-yne via the 1-titanacyclobut-3-yne (2).
