This study highlights the straight-forward synthesis of substituted 1,2-amino alcohols from simple and readily available aromatic methyl ketones. Starting from acetophenone derivatives, the straight-forward synthesis strategy involved an initial bromination of the alpha-positioned methyl group in the first step, followed by a simple hydrolysis to the hydroxyketone (2-hydroxyacetophenone). The hydroxylated intermediate was subsequently converted from Silicibacter pomeroyi to the final 1,2-amino alcohol by using the transaminase. The transaminase-catalyzed reaction proceeded with yields up to 62 % and always excellent enantiomeric excess of >99 %. Interestingly, the keto-enol-tautomerism of the hydroxyl ketone yields an unexpected amino alcohol isomer.  相似文献

Soluble polyphosphide anions were successfully generated in a number of organic solvents by the reaction between shelf‐stable red phosphorus and potassium ethoxide. The species were identified by 31P NMR spectroscopy in solution and by X‐ray crystal‐structure determination of (Bu4N)2P16 in the solid state. The monitoring of phenolic compounds in wastewaters in a simple manner is of great importance for environmental control. Here, a novel screen printed laccase-based microband array for in situ, total phenol estimation in wastewaters and for water quality monitoring without additional sample pre-treatment is presented. Numerical simulations using the finite element method were utilized for the characterization of micro-scale graphite electrodes. Anodization followed by covalent modification was used for the electrode functionalization with laccase. The functionalization efficiency and the electrochemical performance in direct and catechol-mediated oxygen reduction were studied at the microband laccase electrodes and compared with macro-scale electrode structures. The reduction of the dimensions of the enzyme biosensor, when used under optimized conditions, led to a significant improvement in its analytical characteristics. The elaborated microsensor showed fast responses towards catechol additions to tap water – a weakly supported medium – characterized by a linear range from 0.2 to 10 μM, a sensitivity of 1.35 ± 0.4 A M−1 cm−2 and a dynamic range up to 43 μM. This enhanced laccase-based microsensor was used for water quality monitoring and its performance for total phenol analysis of wastewater samples from different stages of the cleaning process was compared to a standard method.  相似文献

Four-arm star-shaped poly(2-isopropyl-2-oxazolines) (PiPrOx4) are synthesized by cationic polymerization on t-butylcalix[4]arene macroinitiator. The obtained samples differ by polymerization degree of arms NPiPrOx = 9 and 25 and are characterized in chloroform. The behavior in aqueous solutions is studied by light scattering methods and compared with the results of investigation of eight-arm star with similar structure. Three types of particles are observed in solution of short-arm PiPrOx4 at room temperature, whereas only two particle types are present in long-arm star solution. Arm shortening leads to widening of the phase transition interval. The arm number decreasing reduces the phase transition temperature by 1°C.  相似文献

Mass spectrometric approaches have recently gained increasing access to molecular immunology and several methods have been developed that enable detailed chemical structure identification of antigen-antibody interactions. Selective proteolytic digestion and MS-peptide mapping (epitope excision) has been successfully employed for epitope identification of protein antigens. In addition, "affinity proteomics" using partial epitope excision has been developed as an approach with unprecedented selectivity for direct protein identification from biological material. The potential of these methods is illustrated by the elucidation of a beta-amyloid plaque-specific epitope recognized by therapeutic antibodies from transgenic mouse models of Alzheimer's disease. Using an immobilized antigen and antibody-proteolytic digestion and analysis by high resolution Fourier transform ion cyclotron resonance mass spectrometry has lead to a new approach for the identification of antibody paratope structures (paratope-excision; "parex-prot"). In this method, high resolution MS-peptide data at the low ppm level are required for direct identification of paratopes using protein databases. Mass spectrometric epitope mapping and determination of "molecular antibody-recognition signatures" offer high potential, especially for the development of new molecular diagnostics and the evaluation of new vaccine lead structures.  相似文献

The complexes of general formula [ML]2[Mn(NCS)4](ClO4)2 (where M = Cu(II), Ni(II); L = N-dl-5,7,7,12,14,14-hexamethyl-1,4,8,11-tetraazacyclotetradeca-4,11-diene) were obtained and the crystal structures of both heteronuclear compounds were determined at 173 K. Complex [CuL]2[Mn(NCS)4](ClO4)2 (1) crystallizes in a monoclinic space group, C2/c, with a = 41.297(9) A, b = 7.571(2) A, c = 16.417(4) A, beta = 96.97(15) degrees, Z = 8, whereas complex [NiL]2[Mn(NCS)4](ClO4)2.H2O (2) crystallizes in a monoclinic space group, P2/c, with a = 21.018(5) A, b = 7.627(2) A, c = 16.295(4) A, beta = 104.47(1) degrees, Z = 4. The magnetic behaviour of (1) and (2) has been investigated over the temperature range 1.8-300 K. Complex (1) displays ferromagnetic coupling inside the trinuclear core of CuMnCu and compound (2) behaves like a mononuclear Mn(II) system. The magnetic properties of the second compound (2) with a similar trinuclear structure shows that Ni(II) ions have a diamagnetic character and a rather weak zero-field splitting at the central Mn(II) ion occurs. Finally, the magnitudes of the Cu(II)-M(II) interactions with M = Ni and Mn have been compared and qualitatively justified.  相似文献
