The scaling of the polyelectrolyte scattering peak in chitosan solutions, as deduced from the relation qmax∼cpα was studied by synchrotron SAXS as a function of the charge density of the polymer. We observe a variation in the α exponent corresponding to the limit of the ionic condensation, by varying the degree of acetylation of the polymer. The nature of the solution medium also affects the polyelectrolyte peak, and it is shown that in alcoholic/water mixtures, the lower dissociation of the acid induces a lower charge density, thus influencing the polyelectrolyte ordering. 相似文献

The mass spectral behavior of a number of organometallic complexes containing the Group 10 metals Ni, Pd, and Pt, together with various thiolate ligands were studied. For Pd, two main types of complexes, differing by the substituents on the phosphorus atom were studied. Types I and II were substituted with bis(diphenylphosphino)ferrocene and bis(diisopropylphosphino)ferrocene ligands, respectively. The Ni complexes, except for one, and the Pd Type I complexes had no molecular radical cations (M(+.)) in their EI spectra. On the other hand, all the Pt complexes showed intense M(+.) ions in their EI spectra indicating that these complexes were more stable as radical cations than those of Ni and Pd. The FAB and MALDI spectra of all the complexes displayed intense quasi-molecular ions (MH(+)) and the fragmentations in both modes were similar. The MALDI spectra of several complexes displayed only M(+.) ions while one gave evidence of both MH(+) and M(+.) ions. Several Pd Type II complexes yielded intense M(+.) in their EI spectra. 相似文献

Abstract The chemical dehydrochlorination of polyvinyl chloride was studied in solution in tetrahydrofuran. The dehydrochlorinated products were characterized by their chlorine contents and by UV/visible spectroscopy. The absorption spectra of dehydrochlorinated PVC consist of a series of absorption maxima between 200 and 600 nm due to the formation of conjugated polyenic sequences. In this article we describe a new method for polyenic sequences evaluation based on deconvolution of the experimental spectrum. Cytochrome P450 enzymes (CYPs or P450s) are the most important enzymes involved in the phase I metabolism of drugs (and other xenobiotics) in humans, and the corresponding drug metabolites are needed as reference substances for their structural confirmation and for pharmacological or toxicological characterization. We have previously shown that biotechnological synthesis of such metabolites is feasible by whole-cell biotransformation with human CYPs recombinantly expressed in the fission yeast Schizosaccharomyces pombe. It was the aim of this study to compare the activity of seven human microsomal CYPs (CYP2C9, CYP2D6, CYP3A4, CYP3A5, CYP3A7, CYP17, and CYP21) upon coexpression with NADPH-cytochrome P450 oxidoreductases (CPRs) from various origins, namely, human CPR (hCPR) and its homologues from fission yeast (ccr1) and the bishop’s weed Ammi majus (AmCPR), respectively. For this purpose, 28 recombinant strains were needed, with five of them having been constructed previously and 23 strains being newly constructed. Bioconversion experiments showed that coexpression of a CPR does not only influence the reaction rate but, in some cases, also exerts an influence on the metabolite pattern. For CYP3A enzymes, coexpression of hCPR yielded the best results, while for another two, hCPR was equally helpful as ccr1 (both CYP17 and CYP21) or AmCPR (CYP17 only), respectively. Interestingly, CYP2D6 displayed its highest activity when coexpressed with ccr1 and CYP2C9 with AmCPR. These results corroborate the view of CPR as a well-suited bio-brick in synthetic biology for the construction of artificial enzyme complexes. 相似文献

In the present study, three types of silica‐based monoliths, i.e. the first and second generations of commercial silica monolithic columns and a wide‐pore prototype monolith were compared for the analysis of large biomolecules. These molecules possess molecular weights between 1 and 66 kDa. The gradient kinetic performance of the first‐generation monolith was lower than that of the second generation, for large biomolecules (>14 kDa) but very close with smaller ones (1.3–5.8 kDa). In contrast, the wide‐pore prototype column was particularly attractive with proteins larger than 19 kDa (higher peak capacity). Among these three columns, the selectivity and retention remained quite similar but a possible larger number of accessible and charged residual silanols was noticed on the wide‐pore prototype material, which led to unpredicted small changes in selectivity and slightly broader peaks than expected. The peak shapes attained with the addition of 0.1% formic acid in the mobile phase remained acceptable for MS coupling, particularly for biomolecules of less than 6 kDa. It was found that one of the major issues with all of these silica‐based monoliths is the possible poor recovery of large biomolecules (principally with monoclonal antibody fragments of more than 25 kDa). 相似文献

Thiamine (vitamin B1) is an essential nutritional factor metabolized inside the body in its mono-, di-, and triphosphate forms. Although the action of thiamine and thiamine diphosphate have been intensely investigated, many questions remain unanswered and the role of thiamine triphosphate is still especially unknown. To probe recent hypotheses on the implication of thiamine triphosphate in a new phosphorylation pathway involving synaptic proteins, we synthesized a series of thiamine di- and triphosphate analogues that are resistant to both enzymatic and chemical hydrolyses. The key step in the preparation of the title compounds is the coupling of thiamine propyl disulfide with adequately protected methylenebis-phosphonic acid, the corresponding triphosphate analogue, and difluoromethylenebisphosphonic acid. 相似文献

Adsorption of cations (Na(+), Ca(2+), Ba(2+)) onto negatively charged (pH 10.4) hematite (alpha-Fe(2)O(3)) particles has been studied. The oxide material was carefully prepared in order to obtain monodisperse suspensions of well-crystallized, quasi-spherical particles (50 nm in diameter). The isoelectric point (IEP) is located at pH 8.5. Adsorption of barium ions onto oxide particles was carried out and the electrophoretic mobility was measured throughout the adsorption experiment. Comparison with calcium adsorption at full coverage reveals a higher uptake of Ba(2+). In both cases it shows also that chloride ions coadsorb with M(2) ions. Simultaneous uptake of the positive and negative ions explains why the electrophoretic mobility does not reverse to cationic migration. A theoretical study of the surface speciation has been carried out, using the MuSiC model. It reveals the presence of negative as well as positive sites on both sides of the point of zero charge (PZC) of the hematite particles, which may explain the coadsorption of Ba(2+) and Cl(-) at pH 10.4. The effective charge of the oxide particles, calculated from the electrophoretic mobility, is in very good agreement with the results found with the MuSiC modelization and the chloride/barium adsorption ratio. It also verifies the theory of ionic condensation. Calorimetric measurements gave a negative heat for the overall reaction occurring when Ba(2+)/Cl(-) ions adsorb onto hematite. Despite the fact that anions (Cl(-) and OH(-)) adsorption onto mineral oxides is an exothermic phenomenon, it is likely that barium and calcium adsorption is endothermic, denoting the formation of an inner-sphere complex as reported in the literature. 相似文献
