Nanostructured core-shell particles with tailor-made affinity surfaces were used to generate microstructured affinity surfaces by microspotting the particles to form densely packed amorphous nanoparticle layers. These layers provided a large reactive surface for the specific binding of protein ligands from aqueous solution. Biofunctional core-shell particles were synthesized for this purpose that consisted of a silica core with a diameter of 100 nm and an organic shell a few nm thick. The nanoparticle core was prepared by sol-gel chemistry and the shell formed in suspension by organosilane chemistry. The shell provided amino groups or carbonyl groups at its outer surface for subsequent covalent immobilization of streptavidin, rabbit IgG antibodies or goat IgG antibodies. AlexaFluor 647-conjugated and biotinylated cytochrome C and CyDye-labeled anti-rabbit IgG and anti-goat IgG were probed as model analytes. The core-shell nanoparticles were spotted using a pin-ring micro-arrayer onto microscope glass slides that were coated with a polycation monolayer by dip-coating prior to nanoparticle deposition. Amorphous particle layers of well-defined thicknesses in the range of 100 nm to 2 microm were obtained by printing aqueous particle suspensions containing 5-500 mg/mL (0.5-50 wt%) of silica particles. The specific affinity of the plotted nanoparticulate capture surface was demonstrated by binding Cy3-labeled donkey anti-rabbit IgG and Cy5-labeled mouse anti-goat IgG to immobilized rabbit IgG and goat IgG particles. The signal intensity per spot increased for any given analyte concentration when the amount of particles per spot was augmented. This was attributed to the increasing integration of receptor molecules per surface footprint, which shifted the binding equilibrium towards the formation of the receptor-ligand complex. Additionally, the locally-increased supply of receptor molecules at the nanoparticulate microchip surface resulted in a wide dynamic range of 4 fM-20 nM (covering six orders of magnitude). 相似文献

In information theory, the link between continuous information and discrete information is established through well-known sampling theorems. Sampling theory explains, for example, how frequency-filtered music signals are reconstructible perfectly from discrete samples. In this Letter, sampling theory is generalized to pseudo-Riemannian manifolds. This provides a new set of mathematical tools for the study of space-time at the Planck scale: theories formulated on a differentiable space-time manifold can be equivalent to lattice theories. Cloud point separation of selected dyes was studied. The use of dyes made possible observation of the dynamics of surfactant-rich phase separation by color video. The pictures were interpreted by means of ImageC software and degrees of whiteness were calculated. It was found that separation was slow and equilibrium was not achieved even after a period of more than 10 h. The separated surfactant-rich phase had a heterogeneous structure. The globules of the surfactant-rich phase were also observed in the micellar aqueous phase. The surfactant concentration could be as high as one or two orders of magnitude above the expected cmc values and was not decreased to critical micelle concentration by centrifugation. The presence of sodium chloride was important and improved separation. Separation of dyes was in the range 73-98% and depended upon the surfactant, the temperature, the electrolyte content, and the dye. In each system considered, appropriate conditions had to be selected to obtain high recovery of the dye. The presence of the electrolyte was the most important parameter and it improved the separation of dyes. However, addition of the electrolyte could also account for precipitation of the dyes, as observed for the systems containing Direct Pink. The best recoveries were observed for Direct Yellow and oxyethylated nonylphenol (98% at 55 degrees C in the presence of NaCl). Centrifugation gave recoveries similar to those for prolonged heating but it shortened the time of phase separation. 相似文献

Self-assembled columnar AlGaN/GaN nanocavities, with an active region of GaN quantum disks embedded in an AlGaN nanocolumn and cladded by top and bottom AlN/GaN Bragg mirrors, were grown. The nanocavity has no cracks or extended defects, due to the relaxation at the Si interface and to the nanocolumn free-surface to volume ratio. The emission from the active region matched the peak reflectivity by tuning the Al content and the GaN disks thickness. Quantum confinement effects that depend on both the disk thickness and the inhomogeneous strain distribution within the disks are clearly observed. 相似文献
