Studies on the refolding of the reduced-denatured insulin with size exclusion chromatography

The refolding of the reduced-denatured insulin from bovine pancreas was investigated with the size exclusion chromatography (SEC). It was shown that the reduced-denatured insulin originally denatured with 7.0 mol·L-1 guanidine hydrochloride (GuHCI) or 8.0 mol·L-1 urea could not be refolded with a non-oxidized mobile phase. Although the oxidized and reduced glutathione (GSSG and GSH) were employed in the oxidized mobile phase, the reduced-denatured insulin still could not be renatured. However, in the presence of 2.0 mol·L-1 urea in the oxidized mobile phase employed, the reduced-denatured insulin can be refolded with SEC, and the aggregation of denatured insulin can be diminished by urea. In addition, the disul-fide exchange of reduced-denatured insulin also can be accelerated with GSSG/GSH in the oxidized mobile phase. The three disulfide bridges of insulin were formed correctly and the reduced-unfolded insulin can be renatured completely. The results were further tested with re-versed-phase liquid chromatography (RPLC) and hydrophobic interaction chromatography (HIC).

采用均相反应的方法合成了6个含羟基芳基或氨基芳基的磷酰胺酯衍生物:[(ClCH_2CH_2)_2NP(O)(OPh)(XArY)](X:O,NH;Y:m或p-OH或NH_2).用元素分析、红外光谱、氢核磁共振谱及质谱证实了它们的组成及结构.     

复合催化剂上CO2加氢合成C2烃类

介绍了由CO₂+H₂合成C2⁺烃的几种复合催化剂体系的研究进展，比较和评价了复合催化剂体系的活性和选择性及对C2⁺烃类生成的影响。着重于复合催化剂体系对C4⁺烃的生成及产物分布的影响并简述反应机理。     

New unsaturated polyesters as injectable drug carriers

New unsaturated polyesters of poly(fumaric acid-glycol-sebacic acid) copolymers and poly(maleic anhydride-glycol-sebacic acid) copolymers were prepared by melt polycondensation of the corresponding mixed monomers: sebacic anhydride, fumaric acid or maleic anhydride and glycol. Methyl-methacrylate (MMA) was used as crosslinker and dimer acid was used as thinner.In vitro studies showed that those copolymers are degradable in phosphate buffer at 37 °C and poly(fumaric acid-glycol-sebacic acid) has proper drug release rate as drug carriers. The biocompatibility of poly(fumaric acid-glycol-sebacic acid) copolymers under mice skin was also evaluated; macroscopic observation and microscopic analysis demonstrated that the copolymer is biocompatible and well tolerated in vivo. The injected poly(fumaric acid-glycol-sebacic acid) [molar ratio M_{fumaric acid}:M_glycol:M_{sebacic acid} = 1.75:2.20:0.25] containing 5% adriamycin hydrochloride (ADM) in the mice bearing Sarcoma-180 tumor exhibited a good antitumor efficacy. The volume doubling time (VDT) (18 ± 2.5 days) of the tumor growth by this treatment was longer than that (7 ± 0.9 days) by the subcutaneous injection of ADM.     

掺硼多晶金刚石膜的电化学性能研究

采用EACVD(Electron Assisted Chemical Vapor Deposition)方法制备了掺硼金刚石膜, 并用扫描电镜、拉曼光谱及霍尔效应等测试方法对其表面形貌、生长特性、载流子浓度以及导电性能进行了分析. 测试结果表明, 掺硼金刚石膜是由微米级晶粒组成的多晶膜, 其载流子浓度为4.88×1020 cm-3, 电阻率为0.03 Ω·cm, 是高品质金刚石膜. 用该金刚石膜制作电化学电极, 利用循环伏安法分别测量了金刚石膜电极在氯化钾空白底液、亚铁氰化钾溶液和左旋半胱氨酸溶液中的循环伏安曲线, 发现该金刚石膜电极在水溶液中具有宽的电化学窗口(约为3.7 V)和接近零的背景电流, 在生物制剂的检测中具有很高的灵敏度和良好的稳定性, 是一种理想的电化学电极材料.     

超临界CO2法制备头孢唑啉钠脂质体

采用超临界CO2(scCO2)流体代替有机溶剂一步法制备了头孢唑啉钠药物载体脂质体. 研究了该脂质体的尺寸、稳定性和药物的包封率. 结果表明, 脂质体的尺寸和稳定性依赖于制备压力, 脂质体对头孢唑啉钠的包封率与乙醇和脂浓度有关, 采用超临界CO2法制备脂质体的药物包封率比采用薄膜分散法(Bangham method)制备的包封率高.     

Synthesis and magnetic properties of μ-phenolato copper (Ⅱ) binuclear complexes with different exogenous bridges

The syntheses and magnetic properties are reported for a series of copper(Ⅱ) complexes prepared from a pentadentate binucleating ligand 2,6-diformyl-4-methylphenol di(benzoyl-hydrazone) (H3L). These complexes incorporate different exogenous ions (X-) into a bridging position to form copper(Ⅱ) binuclear complexes of the formula [Cu2(H2L)X]2+, where X-= Br-(1), Cl-(2), HO-(3), C2H5O-(4) and C3H3N2- (5). The complexes have been characterized with variable temperature magnetic susceptibility (4.2-300 K) and the observed data were fit to those from a modified Bleaney-Bowers equation by least-squares method, giving the exchange integral 2J = -6.2 cm-1 for 1, -76.4 cm-1 for 2, -241.9 cm-1 for 3, -231.1 cm-1 for 4 and -343.8 cm-1 for 5. This suggested that there is an antiferromagnetic interaction between the Cu(Ⅱ) ions and the sequence of the effect of some exogenous bridging ligands on magnetic coupling is corresponding to that in spectrochemical series.     

1,3-二(2,6-二甲基苯基)-2-四氢咪唑基-苯亚甲基-二吡啶基-二氯合钌 化合物的合成

设计了由1,3-二(2,6-二甲基苯基)-2-四氢咪唑基-苯亚甲基-三苯基膦-二氯合钌(7)和吡啶反应生成无膦型金属钌卡宾化合物1,3-二(2,6-二甲苯基)-2-四氢咪唑基-苯亚甲基-2-吡啶基-二氯合钌(8), 8作为高效催化剂用于丙烯腈和烯丙基苯的交叉交互置换反应. 新化合物7, 8经核磁共振氢谱、碳谱和高分辨率质谱予以证实.     

Sharpless四唑合成中捕获双四唑锌配合物

The reaction of malononitrile with ZnCl₂ and NaN₃ in the presence of 2,2′-bipyridine and water affords the first bis(tetrazole) Zn-complex intermediate, mono(2,2′-bipyridine)bis(tetrazolyl)methane aqua zinc(Ⅱ), [Zn(2,2′-bpy)(BTZ)(H₂O)]·(H₂O)₂ (1) which gives a clue for Demko-Sharpless′ tetrazole synthesis.
