A simple and highly sensitive electrochemical assay for ligation-free and polymerase chain reaction (PCR)-free microRNA (miRNA) expression profiling is described in this work. The electrode used in the assay was made of a monolayer of stem-looped capture probes (CPs) comprising of a miRNA complementing region at one end and detection probes (DPs) receiving region at the other. It engaged an electrocatalytic reaction between electrochemically activated glucose oxidase (GOx) and glucose to enhance its sensitivity. Briefly, upon hybridizing to its target miRNA, the stem loop is unlocked exposing the DP receiving region. A subsequent hybridization with the DPs brought them together with an amplifier, the activated GOx, onto the electrode. The activated GOx exhibited excellent catalytic activity towards electrooxidation of glucose. MicroRNA detection could therefore be conducted in 60 mM glucose in phosphate-buffered saline. A detection limit of 4.0 fM and a linear calibration curve up to 10 pM were obtained under optimal conditions. The assay was applied to profile human let-7 miRNA expressions in cultured cancer cells. 相似文献

A novel fingerprinting method, bioactivity fingerprint analysis, based on an ultrafiltration–ultraperformance liquid chromatography–multistage tandem mass spectrometry (UPLC–MS n ) method is proposed for the quality control of herbal medicines from the bioactivity viewpoint concerning the efficacy of herbal medicines. The bioactivity fingerprints reflecting the anti-inflammatory activities of radix Aconiti and radix Aconiti preparata were established. With use of ultrafiltration UPLC–MS n , 11 cyclooxygenase-2 ligands from radix Aconiti preparata and 14 cyclooxygenase-2 ligands from radix Aconiti were found after incubation with cyclooxygenase-2. Twelve of the cyclooxygenase-2 ligands were identified by the ultraperformance UPLC–MS n method. The enrichment factor of each peak in the bioactivity fingerprint was calculated and was demonstrated to be characteristic, which makes bioactivity fingerprint analysis for the quality control of herbal medicines possible from the viewpoint of their bioactivities. Two catechin epimers and their non‐covalent complexes with γ‐cyclodextrin were studied by using ion mobility coupled with mass spectrometry (IM‐MS). Rapid separation of complexes was achieved with the peak‐to‐peak resolution reaching 0.86 after optimization of IM condition. Collision cross section (CCS) was measured to explore the structural difference of complexes. A gap of 11.75 Å2 between two complexes was found. Molecular modeling and theoretical CCS calculation were adopted to explain the measurement results. Two binding ways of both complexes were found and the calculated CCS corresponds accurately to the measured CCS. Quantification of catechins in mixtures was performed and the relative error was less than 15%, indicating the effectiveness of quantification by IM‐MS. 相似文献

An orthogonal test was used to design different mixture ratios of molybdenum disulfide(MoS2), graphite, and SiO2 particles, which were filled with polytetrafluoroethylene (PTFE) composite. MoS2-, graphite-, and SiO2-modified PTFE was obtained by pressing and sintering, and the processing parameters were determined using progressive studies and experiments. The friction and wear properties of different PTFE composites lubricated with natural seawater were analyzed using an MMU-5G wear tester. A laser scanning confocal microscope was employed to examine the morphological characteristics of the worn surface. Moreover, the influence of particle proportions on the tribological property of composites was analyzed. Results show that the addition of SiO2, MoS2, and graphite can increase the bearing capacity, improve the wear resistance, reduce the friction coefficient, and increase the self-lubricating ability of the PTFE matrix. 相似文献

Single crystal surfaces with highly coordinated sites very often hold high specific activities toward oxygen reduction reaction (ORR) and others. Transposing their high specific activity to practical high-surface-area electrocatalysts remains challenging. Here, ultrathin Pt(100) alloy surface is constructed via epitaxial growth. The surface shows 3.1–6.9 % compressive strain and bulk-like characteristics as demonstrated by site-probe reactions and different spectroscopies. Its ORR activity exceeds that of bulk Pt3Ni(100) and Pt(111) and presents a 19-fold increase in specific activity and a 13-fold increase in mass activity relative to commercial Pt/C. Moreover, the electrochemically active surface area (ECSA) is increased by 4-fold compared to traditional thin films (e.g. NSTF), which makes the catalyst more tolerant to voltage loss at high current densities under fuel cell operation. This work broadens the family of extended surface catalysts and highlights the knowledge-driven approach in the development of advanced electrocatalysts. 相似文献
