A micro fluorescent analysis system is proposed using silicon micromachining. GaN blue light-emitting diode (LED) monolithically integrated on a silicon substrate is used as a light source for the fluorescent analysis system. The blue light suits the excitation of several dyes used commonly in fluorescent analysis. Silicon photodiode (Si-PD) that matches the visible and near infrared fluorescent wavelengths of dyes is integrated on a silicon substrate. Polydimethylsiloxane (PDMS) micro-channels are also stacked for flowing dye-sensitized liquid. Therefore, the proposed system is an integrated system that can be composed on a silicon platform, i.e. a bottom layer of Si-PD, a middle layer of GaN-LED on silicon substrate and a top layer of micro PDMS channel. An aperture is opened into the GaN-LED layer by deep reactive ion etching to create a ring-shaped GaN-LED and a through-hole for detection. The light from the ring-shaped GaN-LED in the middle layer excites the dye-sensitized liquid in the top micro-channel layer. The fluorescence emitted from dye is detected by the Si-PD on the bottom layer at an angle larger than 90 degrees from the direction of excitation. Therefore, the detection optics consist basically of a dark-field illumination optical system. In order to evaluate the performance of the integrated system, fluorescence of fluorescein isothiocyanate (FITC) solution flowing in the micro channel is measured. From the measurement, the noise, sensitivity and limit of detection in the fabricated system are evaluated for FITC dye to be 0.57 pA, 1.21 pA μM(-1) and 469 nM, respectively. From these results, a compact fluorescence analysis system is demonstrated.  相似文献

Green tea is manufactured from the leaves of the plant Camellia sinensis Theaceae and has been regarded to possess anti-cancer, anti-obesity, anti-atherosclerotic, anti-diabetic, anti-bacterial, and anti-viral effects. Many of the beneficial effects of green tea are related to the activities of (-)-epigallocatechin gallate (EGCG), a major component of green tea catechins. For about 20 years, we have engaged in studies to reveal the biological activities and action mechanisms of green tea and EGCG. This review summarizes several lines of evidence to indicate the health-promoting properties of green tea mainly based on our own experimental findings.  相似文献

We report on the development of a microfluidic system for the electrical detection of single pollen allergen particles. Our device consists of 500 nm electrode gaps fabricated in an 800 nm wide fluidic channel. We flowed pollen allergen particles of average size 330 nm along the channel via fluid pumping and simultaneously monitored temporal change in dc current flowing through the sensing electrodes. Current spikes were detected, which can be attributed to a capacitance discharging upon trapping/detrapping of single allergens in the electrode gap. This sensing mechanism may open new avenues for a highly sensitive pollen allergen sensor.  相似文献

The obtained material was a second-stage compound with a d-spacing of 0.367 nm for the intercalated layers. We have directly probed the Ba 5d states in the ferroelectric barium titanate BaTiO3 using two bulk-sensitive spectroscopic probes, resonant x-ray emission spectroscopy (RXES) and x-ray absorption spectroscopy in the partial fluorescence mode (PFY-XAS) at the Ba-L3 edge. The results are compared with those of the non-ferroelectric barium sulfate BaSO4. While the RXES spectra point to a localized character for the Ba 5d states in both compounds, the main peak of the PFY-XAS spectrum, corresponding to the dipolar transitions from 2p to 5d, is found to be significantly broader for BaTiO3 than for BaSO4. On the basis of band structure calculations, this broadening is ascribed to strong hybridization between the unoccupied Ba 5d and O 2p states in the ferroelectric. This suggests that the hybridization between the conduction states of the Ba2+ and O2? ions, and not only Ti4+ and O2?, plays a central role in determining the electronic structure of BaTiO3, and is therefore likely to be indirectly correlated with the occurrence of ferroelectricity in this material.  相似文献

We immobilized human cytochrome P450 (CYP), a membrane-bound enzyme, onto both smooth and nanostructured surfaces of gold electrodes via a naphthalene thiolate monolayer film. Rapid electron transfer of CYP with an electrode as a redox partner took place when the enzyme was immobilized onto an electrode surface with nanostructures. This structure was easily prepared by conventional sputtering techniques. A well-defined pair of peaks was observed at ? 0.175 V (vs. SHE) with the largest heterogeneous electron transfer rate constant of 340 s? 1 for human CYP. The positive redox potential shift of 45 mV upon drug (testosterone) binding was clearly detected, which corresponded to a change in the spin states of heme iron in CYP. The present study showed that gold sputtered surfaces are very useful for direct electron transfer reactions of human CYP isoforms.  相似文献
