The shortest known type 1 copper binding loop (that of amicyanin, Ami) has been introduced into three different cupredoxin beta-barrel scaffolds. All of the loop-contraction variants possess copper centers with authentic type 1 properties and are redox active. The Cu(II) and Co(II) sites experience only small structural alterations upon loop contraction with the largest changes in the azurin variant (AzAmi), which can be ascribed to the removal of a hydrogen bond to the coordinating thiolate sulfur of the Cys ligand. In all cases, loop contraction leads to an increase in the pK(a) of the His ligand found on the loop in the reduced proteins, and in the pseudoazurin (Paz) and plastocyanin (Pc) variants the values are almost identical to that of Ami ( approximately 6.7). Thus, in Paz, Pc, and Ami, the length of this loop tunes the pK(a) of the His ligand. In the AzAmi variant, the pK(a) is 5.5, which is considerably higher than the estimated value for Az (<2), and other controlling factors, along with loop length, are involved. The reduction potentials of the loop-contraction variants are all lower than those of the wild-type proteins by approximately 30-60 mV, and thus this property of a type 1 copper site is fine-tuned by the C-terminal loop. The electron self-exchange rate constant of Paz is significantly diminished by the introduction of a shorter loop. However, in PcAmi only a 2-fold decrease is observed and in AzAmi there is no effect, and thus in these two cupredoxins loop contraction does not significantly influence electron-transfer reactivity. Loop contraction provides an active site environment in all of the cupredoxins which is preferable for Cu(II), whereas previous loop elongation experiments always favored the cuprous site. Thus, the ligand-containing loop plays an important role in tuning the entatic nature of a type 1 copper center.  相似文献

Methods of utilizing the Youden plot obtained from interlaboratory comparison tests are proposed. The Youden plot is a powerful tool for visually evaluating between- or within-laboratory errors. However, there are no generally applicable methods to indicate the anomalies in the between- and the within-laboratory errors in qualitative terms. An evaluation frame was therefore developed in this study as an indication method. Moreover, the Youden plot is considered to be useful for visually assessing the comprehensive performance of laboratories using two types of results. Although the confidence ellipse has been proposed in ISO 13528:2005 for this purpose, robust determination of the parameters has not yet been implemented. In the present study, robust determination of one of the parameters, the correlation coefficient, was developed. Our proposals were analytically validated using two types of statistical models. In addition, the properties were evaluated through computational simulations. From the results obtained, it can be concluded that our proposals are practically applicable to actual interlaboratory comparison tests.  相似文献

Bio‐inspired functional microcapsules have attracted increasing attention in many fields from physical/chemical science to artificial‐cell engineering. Although particle‐stabilised microcapsules are advantageous for their stability and functionalisation potential, versatile methods for their functionalisation are desired to expand their possibilities. This study reports a water‐in‐oil microdroplet stabilised with amphiphilic DNA origami nanoplates. By utilising DNA nanotechnology, DNA nanoplates were designed as a nanopore device for ion transportation and to stabilise the oil–water interface. Microscopic examination revealed the microcapsule formed by the accumulation of amphiphilic DNA nanoplates at the oil–water interface. Ion current measurements revealed the nanoplate pores functioned as channel to transport ions. These findings provide a general strategy for the programmable design of microcapsules to engineer artificial cells and molecular robots.  相似文献

ABSTRACT

In this study, ultrasonic measurements were performed on a single crystal of cubic PrNi2Cd20, down to a temperature of 0.02?K, to investigate the crystalline electric field ground state and search for possible phase transitions at low temperatures. The elastic constant (C11?C12)/2, which is related to the Γ3-symmetry quadrupolar response, exhibits the Curie-type softening at temperatures below ~30?K, which indicates that the present system has a Γ3 non-Kramers doublet ground state. A leveling-off of the elastic response appears below ~0.1?K toward the lowest temperatures, which implies the presence of level splitting owing to a long-range order in a finite-volume fraction associated with Γ3-symmetry multipoles. A magnetic field–temperature phase diagram of the present compound is constructed up to 28?T for H || [110]. A clear acoustic de Haas–van Alphen signal and a possible magnetic-field-induced phase transition at H ~26?T are also detected by high-magnetic-field measurements.  相似文献

An exclusive measurement has been made of the Coulomb dissociation of the two-neutron halo nucleus 11Li at 70 MeV/nucleon at RIKEN. Strong low-energy (soft) E1 excitation is observed, peaked at about Ex = 0.6 MeV with B(E1) = 1.42(18) e2fm2 for Erel < or = 3 MeV, which was largely missed in previous measurements. This excitation represents the strongest E1 transition ever observed at such low excitation energies. The spectrum is reproduced well by a three-body model with a strong two-neutron correlation, which is further supported by the E1 non-energy-weighted cluster sum rule.  相似文献
