In the process of drug discovery, drug-induced liver injury (DILI) is still an active research field and is one of the most common and important issues in toxicity evaluation research. It directly leads to the high wear attrition of the drug. At present, there are a variety of computer algorithms based on molecular representations to predict DILI. It is found that a single molecular representation method is insufficient to complete the task of toxicity prediction, and multiple molecular fingerprint fusion methods have been used as model input. In order to solve the problem of high dimensional and unbalanced DILI prediction data, this paper integrates existing datasets and designs a new algorithm framework, Rotation-Ensemble-GA (R-E-GA). The main idea is to find a feature subset with better predictive performance after rotating the fusion vector of high-dimensional molecular representation in the feature space. Then, an Adaboost-type ensemble learning method is integrated into R-E-GA to improve the prediction accuracy. The experimental results show that the performance of R-E-GA is better than other state-of-art algorithms including ensemble learning-based and graph neural network-based methods. Through five-fold cross-validation, the R-E-GA obtains an ACC of 0.77, an F1 score of 0.769, and an AUC of 0.842.  相似文献

Organic charge-transfer complexes (CTCs) can function as versatile second near-infrared (NIR-II) theranostic platforms to tackle complicated solid tumors, while the structure–property relationship is still an unanswered problem. To uncover the effect of molecular stacking modes on photophysical and biochemical properties, herein, five ferrocene derivatives were synthesized as electron donors and co-assembled with electron-deficient F4TCNQ to form the corresponding CTCs. The crystalline and photophysical results showed that only herringbone-aligned CTCs (named anion-radical salts, ARS NPs) possess good NIR-II absorption ability and a photothermal effect for short π–π distances (<3.24 Å) and strong π-electron delocalization in the 1D F4TCNQ anion chain. More importantly, the ARS NPs simultaneously possess ·OH generation and thiol (Cys, GSH) depletion abilities to perturb cellular redox homeostasis for ROS/LPO accumulation and enhanced ferroptosis. In vitro experiments, FcNEt-F4 NPs, and typical ARS NPs, show outstanding antitumor efficiency for the synergistic effect of NIR-II photothermal therapy and ferroptosis, which provides a new paradigm to develop versatile CTCs for anti-tumor application.

Optical imaging and phototherapy in the second near-infrared window(NIR-Ⅱ, 900–1700 nm) can reduce tissue auto-fluorescence and photon scattering, which facilitates higher spatial resolution and deeper tissue penetration depth for solid tumor theranostics. Herein, a polycyclic naphthalenediimide(NDI) based chromophore 13-amino-4,5-dibromo-2,7-di(dodecan-6-yl)-1 H-isoquinolino[4,5,6-fgh]naphtho[1,8-bc][1,9]phenanthroline-1,3,6,8(2H,7H,9H)-tetraone(NDI-NA) was designed and synthesized. With large polycyclic π-systems, NDI-NA molecule possesses broad near-infrared(NIR) absorption(maximum at777 nm) and emission(maximum at 921 nm). By nanoprecipitation, NDI-NA nanoparticles(NPs) were formed in aqueous solution with J-aggregative state, which showed huge red-shift in both absorption spectrum(maximum at 904 nm) and emission spectrum(maximum at 1,020 nm), endowing NDI-NA NPs efficient NIR-Ⅱ fluorescence imaging capability. Besides, the NPs present effective tumor-targeting capability in vivo based on the enhanced permeation and retention(EPR) effect. More importantly, NDI-NA NPs simultaneously have high photothermal conversion efficiency(30.8%) and efficient reactive oxygen species generation ability, making them remarkably phototoxic to cancer cells. The polycyclic chromophore based multifunctional NDI-NA NPs as NIR-Ⅱ phototheranostic agents possess bright future for clinical NIR-Ⅱ imaging-guided cancer phototherapy.  相似文献

The interfacial tension, wettability, foam and emulsification properties of di-tetrapropylene diphenyl ether disulfonates (C12-DADS) gemini-type surfactant have been studied and compared to mono-tetrapropylene diphenyl ether disulfonates (C12-MADS) and tetrapropylene benzene sulfonates (C12-BAS). The results show that the C12-DADS have a lower interfacial activity compared to that of C12-MADS and C12-BAS. The wettability of the three surfactants decreases in the order C12-BAS?>?C12-DADS?>?C12-MADS. For foam properties, the foamability and foam stability of C12-MADS and C12-DADS is lower than that of C12-BAS. In addition, the emulsification ability increase in the order C12-DADS?12-MADS?12-BAS.  相似文献

Under atmospheric pressure, nano-polyethylene fibres were prepared via in situ ethylene extrusion polymerization, with MCM-41 and SBA-15 supported zirconocene dichloride (Cp2ZrCl2) catalytic systems, respectively. The effects of the geometrical structures and surface properties of MCM-41 and SBA-15 on the morphology of the resultant polyethylene, catalytic activity and polymerization rate were investigated and compared in various polymerization conditions. The possible formation mechanism of nano-polyethylene fibres with MCM-41 and SBA-15 supported Cp2ZrCl2 as catalyst was discussed.  相似文献

A novel brush-type amphiphilic copolymer of PSMA-b-PTMSPMA was synthesized via ATRP technique. As-synthesized polymer was characterized by GPC and 1H NMR. It was of interest that the resultant polymer could self-assemble into micelles with different morphologies and sizes in selective solvents by adjusting the copolymer concentrations. These aggregates could be prepared into novel stable organic/inorganic hybrid nanomaterials by the gelation process. The size and structure of these aggregates and the corresponding hybrid nanomaterials were observed by TEM.  相似文献
