Modeling of biological reactions

Cell membranes consist of lipid bilayers in which proteins are embedded. Many cell functions are carried out at the cell boundary which interface with water. Here we describe the response to an anesthetic of a lipid bilayer and of an enzyme separately. While both systems are markedly affected by anesthetics at appropriately high concentrations, the result at the clinical concentrations seems best explained as principally an effect on the protein somewhat accentuated by its presence in the lipid bilayer. Thus the lipoprotein complex seems to have the properties of the protein alone, except with somewhat greater induced sensitivity due to the lipid matrix in which it is immersed.     

Ultratrace determination of phosphorus in ultrapurified water by a slope comparison method

The analytical method for the determination of phosphorus in ultrapurified water was developed. Ultrapurified water was evaporated to concentrate phosphorus and the final sample volume for analysis was 10 ml. In 0.55 mol l⁻¹ HCl, orthophosphate forms molybdophosphate, and then the molybdophosphate forms ion associate with Malachite Green (MG), which can be collected on a tiny membrane filter (diameter: 5 mm, and effective filtering diameter: 2 mm). After the ion associate on the membrane filter is dissolved together with the membrane filter in 1 ml of methyl cellosolve (MC), the absorbance of MC solution is measured at 627 nm by a flow injection-spectrophotometric detection technique. When 10 ml of the sample solution was used for the procedures and absorbance measurement, the calibration graph is linear up to about 500 ng l⁻¹ of phosphorus and the detection limit was 8 ng l⁻¹ (S/N=3). For the determination of phosphorus in an ultrapurified water, 10-40 ml of sample solutions were transferred into poly(tetrafluoroethylene) (PTFE) beaker and evaporated to 5 ml or to dryness. To them, 0.003 mol l⁻¹ HCl was added to get 10 ml of final solution, which was used as sample. Phosphate is determined by comparing the slope of the varied sample volume after evaporation/concentration with a slope of the standard calibration graph (a slope comparison method: SCM). The SCM enables to evaluate the concentration of phosphate in ultrapurified waters more sensitively and accurately.     

Carbon-13 NMR chemical shift and electronic structure of polypeptide as studied by tight-binding MO theory: poly (β-benzyl L-aspartate) with the right-handle α-helix and left-handed α-helix forms

An attempt was made to calculate ¹³C NMR chemical shifts of poly(β-benzyl L-aspartate) having the right-handed α-helix (α_R-helix) and left-handed α-helix (α_L-helix) forms by a tight-binding MO sum-over-states theory within the extended Hückel framework, in order to examine whether or not the conformation-dependent ¹³C chemical shifts previously determined by the cross polarization-magic angle spinning technique are reproduced by a change of electronic structure of the polymer. It is found that the relative displacements of the observed C_α, C_β and carbonyl ¹³C chemical shifts between the α_R- and α_L-helices are reproduced qualitatively by the calculation.     

Trace and ultratrace analysis methods for the determination of phosphorus by flow-injection techniques

Trace (≤1 mg/l or 30 μM) and ultratrace (≤1 μg/l or 30 nM) analysis methods for phosphorus determination by flow-injection analysis are reviewed. Most of the methods cited in this review are fundamentally based on the reaction of orthophosphate with molybdate to form heteropoly acids, such as molybdenum yellow and molybdenum blue, and some of the methods are based on the formation of such secondary reactions as ion associates and their aggregates with bulky cations, such as cationic dyes and quaternary ammonium ions. The heteropoly acids themselves can be measured by spectrophotometry, and the ion associate formed with a cationic dye, Malachite Green (MG), can be measured based on the coloration of MG. Light scattering detection methods can be used for measuring the aggregates of ion associates formed with bulky cations. Highly sensitive detection of phosphorus can be accomplished by fluorophotometry; Rhodamine B (RB) and its analogues react with molybdophosphate to form ion associates, which shows fluorescence quenching of RB: LOD is about 5 nM. The detection method based on the chemiluminescence of luminal oxidized with molybdophosphoric acids is probably the most sensitive of all the detection methods reported so far: LOD of the method is as low as 1 nM. The LOD of the molybdenum blue method can be improved by using a liquid core waveguide: LOD is 0.5 nM.     

A new tomato pregnane glycoside from the overripe fruits

A new pregnane glycoside has been isolated from the overripe fruits of Cherry tomato (Mini tomato), Lycopersicon esculentum var. cerasiforme (DUNAL) ALEF. The structure was determined to be 3-O-beta-lycotetraosyl 3beta-hydroxy-5alpha-pregn-16-en-20-one on the basis of spectroscopic analysis. The seasonal variation of the tomato saponin is discussed.     

Capillary zone electrophoretic studies of ion association between inorganic anions and tetraalkylammonium ions in aqueous-dioxane media

Ion association between inorganic anions and symmetrical tetraalkylammonium ions, R4N+ (R = Me, Et, Pr, n-Bu, n-Am, and 2-methyl butyl {isoamyl = iAm}) was investigated using ordinary silica capillary by capillary zone electrophoresis. An improved version of the Williams-Vigh method was used for the first time to measure the mobilities of the inorganic anions. Plots of log Kass against log dielectric constant in various media, revealed a smaller change in Kass compared to dielectric constant. These plots suggest that the Bjerrum's equation is inadequate in accounting for the associations of ions in a CZE setup.
