The present work describes the systematic development of a robust, precise, and rapid reversed‐phase liquid chromatography method for the simultaneous determination of eprosartan mesylate and its six impurities using quality‐by‐design principles. The method was developed in two phases, screening and optimization. During the screening phase, the most suitable stationary phase, organic modifier, and pH were identified. The optimization was performed for secondary influential parameters—column temperature, gradient time, and flow rate using eight experiments—to examine multifactorial effects of parameters on the critical resolution and generated design space representing the robust region. A verification experiment was performed within the working design space and the model was found to be accurate. This study also describes other operating features of the column packed with superficially porous particles that allow very fast separations at pressures available in most liquid chromatography instruments. Successful chromatographic separation was achieved in less than 7 min using a fused‐core C18 (100 mm × 2.1 mm, 2.6 μm) column with linear gradient elution of 10 mM ammonium formate (pH 3.0) and acetonitrile as the mobile phase. The method was validated for specificity, linearity, accuracy, precision, and robustness in compliance with the International Conference on Harmonization Q2 (R1) guidelines. The impurities were identified by liquid chromatography with mass spectrometry.  相似文献

Enantioselective analysis or separation is very essential for improved therapeutic effects of drugs as the pure enantiomeric drug formulations display potential benefits over racemates. In this work, we carried out (i) the synthesis of a nanocomposite of β-cyclodextrin and 3D graphene (G/β-CD NC), and (ii) its application for the detection of fluoxetine enantiomers [(RS)-FLX)] using a thin-layer chromatographic method. The synthesized nanocomposite was introduced into silica gel slurry while preparation of thin-layer plates. The separation conditions were optimized by altering pH, temperature, and mobile phase composition. The method is simple and easy to be optimized, and it can therefore be exploited to assess and monitor routine work of enantiomeric purity of drug enantiomers. The average precision (as measured by RSD) was in the region of 1.35‒1.65% for the enantiomers of (RS)-FLX. The measured limit of detection and limit of quantification for (RS)-FLX enantiomers were 1.8 and 5.4 mg mL‒1, respectively.

We analyze the screening of an external Coulomb charge in gapless graphene cone, which is taken as a prototype of a topological defect. In the subcritical regime, the induced charge is calculated using both the Green’s function and the Friedel sum rule. The dependence of the polarization charge on the Coulomb strength obtained from the Green’s function clearly shows the effect of the conical defect and indicates that the critical charge itself depends on the sample topology. Similar analysis using the Friedel sum rule indicates that the two results agree for low values of the Coulomb charge but differ for the higher strengths, especially in the presence of the conical defect. For a given subcritical charge, the transport cross-section has a higher value in the presence of the conical defect. In the supercritical regime we show that the coefficient of the power law tail of polarization charge density can be expressed as a summation of functions which vary log periodically with the distance from the Coulomb impurity. The period of variation depends on the conical defect. In the presence of the conical defect, the Fano resonances begin to appear in the transport cross-section for a lower value of the Coulomb charge. For both sub and supercritical regime we derive the dependence of LDOS on the conical defect. The effects of generalized boundary condition on the physical observables are also discussed.  相似文献

β-phase polyvinylidene fluoride (PVDF)–BaTiO3 nanocomposite samples have been prepared by solution mixing method. XRD data represent that the crystallinity of PVDF decreases with increase in loading level of BaTiO3 nanoparticles. DSC curve represents that the melting point of PVDF is lightly affected by loading concentration of BaTiO3. The morphology and microstructure of PVDF and PVDF embedded by BaTiO3 nanofillers were investigated by using inverted contrast microscopy (ICM) and scanning electron microscopy (SEM). FTIR interferrometry is proven that PVDF and BaTiO3 are not chemically interacting; therefore, interaction of BaTiO3 is van der Waals type of interaction. The thermally stimulated discharge current (TSDC) of PVDF and PVDF–BaTiO3 nanocomposites sample was characterized by single peak. The observed TSDC peak is discussed on the basis of dipolar and interfacial polarization.  相似文献

Herein, we demonstrate that a bioinspired assembly of silica nanoparticles with polyamines as structure‐directing agents similar to that known for the biosilicification of diatoms can pave the way for the efficient encapsulation of sulfonated copper–phthalocyanine in a hybrid microcapsule structure, in which the organic component provides a capable environment for its catalytic activity in epoxidation reactions and the nanoassembled structure imparts stability.  相似文献
